We propose a novel multi-scale modeling framework for infectious disease spreading, borrowing ideas and modeling tools from the so-called Refractory Density (RD) approach. We introduce a microscopic model that describes the probability of infection for a single individual and the evolution of the disease within their body. From the individual-level description, we then present the corresponding population-level model of epidemic spreading on the mesoscopic and macroscopic scale. We conclude with numerical illustrations taking into account either a white Gaussian noise or an escape noise to showcase the potential of our approach in producing both transient and asymptotic complex dynamics as well as finite-size fluctuations consistently across multiple scales. A comparison with the epidemiology of coronaviruses is also given to corroborate the qualitative relevance of our new approach.

A slowly ramped voltage-clamp electrophysiology protocol serves as closed-loop feedback controlled experiment for the subsequent current-clamp open-loop protocol on the same cell. In this way, the voltage-clamped experiment determines dynamically stable and unstable (hidden) steady states of the current-clamp experiment. The transitions between observable steady states and observable spiking states in the current-clamp experiment reveal stability and bifurcations of the steady states, completing the steady-state bifurcation diagram.

The dominant modeling framework for understanding cortical computations are heuristic firing rate models. Despite their success, these models fall short to capture spike synchronization effects, to link to biophysical parameters and to describe finite-size fluctuations. In this opinion article, we propose that the refractory density method (RDM), also known as age-structured population dynamics or quasi-renewal theory, yields a powerful theoretical framework to build rate-based models for mesoscopic neural populations from realistic neuron dynamics at the microscopic level. We review recent advances achieved by the RDM to obtain efficient population density equations for networks of generalized integrate-and-fire (GIF) neurons -- a class of neuron models that has been successfully fitted to various cell types. The theory not only predicts the nonstationary dynamics of large populations of neurons but also permits an extension to finite-size populations and a systematic reduction to low-dimensional rate dynamics. The new types of rate models will allow a re-examination of models of cortical computations under biological constraints.
