Summary Factors affecting the successful rapid proliferation and rooting of male pistachio (Pista... more Summary Factors affecting the successful rapid proliferation and rooting of male pistachio (Pistacia vera L.) cv. Atlı, were studied. The most suitable type of cytokinin [6 benzyladenine (BA), kinetin (Kin), or thidiazuron (TDZ)], and the effect of eight different concentrations of BA (0.0675, 0.125, 0.25, 0.5, 1.0, 2.0, 4.0, or 8.0 mg l–1) were evaluated to optimise shoot proliferation. The auxins -naphthaleneacetic acid (NAA), indole-3-acetic acid (IAA), and indole-3-butyric acid (IBA), each at 2.0 mg l–1, or different concentrations (0.5, 1.0, 2.0, 4.0, or 8.0 mg l–1) of IBA, and the effect of explant size (1.0, 2.0, 3.0, or 4.0 cm) were assessed for root induction. The highest number of new microshoots per explant (5.64 ± 0.07) was obtained 4 weeks after culturing on Murashige and Skoog (MS) medium supplemented with 1.0 mg l–1 BA. Again, shoot length was highest in 1.0 mg l–1 BA, and decreased as the BA concentration increased. IBA was most effective in promoting root formation. The highest rooting frequency (73%) of microshoots was recorded for explants 4.0 cm in length, 4 weeks after culturing. In vitro-rooted plantlets were transferred to polyethylene pots filled with a 1:1:1 (v/v/v) mixture of soil, sand and peat.This treatment resulted in 90% survival of plantlets, which were acclimatised in a greenhouse.

Siirt. The in vitro procedure involved four steps that included (1) induction of shoot initials from the regenerated mature leaf tissue, (2) regeneration and elongation of shoots from the shoot initials, (3) rooting of the shoots, and (4) acclimatization of the plantlets. The induction of shoot initials was achieved on an agarified Murashige and Skoog (MS) medium with Gamborg vitamins supplemented in different concentrations of benzylaminopurine (BA) and indole-3-acetic acid (IAA). This study presents an efficient improvement in the in vitro propagation protocol for one cloned ... more This study presents an efficient improvement in the in vitro propagation protocol for one cloned genotype of lentisk (Pistacia lentiscus L.) by assessing the effects of gibberellic acid (GA3) concentrations, different cytokinins and amino acids and their concentrations on shoot proliferation, the effects of shoot length on rooting and the effects of compost type (sterile and non-sterile) on acclimatization. The best growth medium for multiple shoot induction was the MS medium supplemented with a combination of 1 mg l−1 BA, 100 mg l−1 tryptophan and 0.5 mg l-1 GA3, which gave a mean shoot length of 1.64 ± 0.07 cm and a mean bud number of 5.46 ± 0.16. The best results in terms of root length, rooting rate and the number of roots per shoot were obtained with 2 cm long shoots. The rooted plantlets were readily acclimatized in the sterile compost. Ancak, anac cogaltiminin halen tohumlar ile yapilma nedeni, istenilen ozeliklere sahip olgun bir anactan in vitro kultur baslatma guclugudur. Olgun anaclardan alinan eksplantlarda gorulen kontaminasyon, fenolik salinimi ve kahverengilesme problemleri kultur baslatmanin onundeki en buyuk engellerdendir. Bu arastirmada disi buttum (Pistacia khinjuk Stocks) agaclarinin surgunlerinden alinan 1-2 cm boyundaki apikal ya da lateral uclardan in vitro kultur baslatilmasi amaclanmistir. Eksplantlarin saldigi fenolik bilesikler her hangi bir adsorbant bilesik kullanilmadan % 92 oraninda uzaklastirilmistir. Calismamizin sonucunda, apikal surgun uclarinin % 10’luk NaOCI’te 30 dakika boyunca bekletilmesiyle, %32.5 oraninda yasayan ve suren eksplant elde edilmistir.

A hundred percent of germination was obtained when isolated embryos were cultured in vitro, a result alike those reported in the literature with traditional dormancy breaking treatments. In vitro seedling development was optimal in a modified full-strength Murashige and Skoog (1962) [1] medium with Gamborg vitamins supplemented with 1 mgl-1 BA, 30 gl-1 sucrose, 6.3 gl-1 agar. An average of 2-4 shoots was produced from the initial seedlings after 14 days of culture. In vitro developed plantlets were successfully acclimatized and transferred to soil. In vitro embryo culture thus allows for the production of apricot seedlings that can be transferred to natural orchards.

In this study, the effect of different sodium chloride (NaCl) concentrations (0, 50, 100, 150, 200, 250 mM) on growth and physiological parameters of Pistacia lentiscus L. seedlings raised in in vitro condition for 4 weeks was investigated. For this purpose the seeds of Lentisk were germinated in Murashige and Skoog, (1962) basal mediums containing different NaCl concentrations. The morphological, physiological and biochemical changes that occurred in the seedlings were measured and recorded after exposure to salt stress. These results show that visible leaf damage of Lentisk seedlings are affected by high salt concentrations. High salinity concentrations significantly reduce root and stem lengths, relative water content (RWC), total chlorophyll, Cl-a, Cl-b and carotenoid values after the culture periods. Bu çalışmada, kirazın lateral tomurcukları kullanılarak kültür başlatılmasına, sitokininlerden be... more Bu çalışmada, kirazın lateral tomurcukları kullanılarak kültür başlatılmasına, sitokininlerden benzilaminopurin (BAP), thidiazuron (TDZ) ve kinetinin bazı konsantrasyonlarının etkisi üzerinde çalışılmıştır. Tomurcuklar, 0900-Ziraat çeşidine ait 5 yaşındaki ağaçların bir yaşlı dallarından dormant dönemde alınmıştır. Araştırmada BAP ve kinetinin 0.1, 0.5, 1.0, 2.0 ve 4.0 mgl-1 konsantrasyonları incelenmiş, TDZ’de bunlarla beraber 0.05 mgl-1 konsantrasyonuna yer verilmiştir. Kiraz tomurcuklarından kültür başlatmak için kinetinin 4 mgl-1 konsantrasyona kadar besi ortamına ilavesinin olumlu bir etkisinin olmadığı anlaşılmıştır. Kullanılan sitokinin tip ve konsantrasyonları içinde en başarılı sonuçlar 2 mgl-1 BAP ve 2 mgl-1 TDZ ilaveli besi ortamlarından alınmıştır. Bu çalışmada, kirazın lateral tomurcukları kullanılarak kültür başlatılmasına, sitokininlerden be... more Bu çalışmada, kirazın lateral tomurcukları kullanılarak kültür başlatılmasına, sitokininlerden benzilaminopurin (BAP), thidiazuron (TDZ) ve kinetinin bazı konsantrasyonlarının etkisi üzerinde çalışılmıştır. Tomurcuklar, 0900-Ziraat çeşidine ait 5 yaşındaki ağaçların bir yaşlı dallarından dormant dönemde alınmıştır. Araştırmada BAP ve kinetinin 0.1, 0.5, 1.0, 2.0 ve 4.0 mgl-1 konsantrasyonları incelenmiş, TDZ’de bunlarla beraber 0.05 mgl-1 konsantrasyonuna yer verilmiştir. Kiraz tomurcuklarından kültür başlatmak için kinetinin 4 mgl-1 konsantrasyona kadar besi ortamına ilavesinin olumlu bir etkisinin olmadığı anlaşılmıştır. Kullanılan sitokinin tip ve konsantrasyonları içinde en başarılı sonuçlar 2 mgl-1 BAP ve 2 mgl-1 TDZ ilaveli besi ortamlarından alınmıştır. Ancak, TDZ bulunan besi ortamlarında gelişen rozet bitkilerin canlılığı, kültür süresinin sonuna doğru azalmıştır.
