In present study used twenty-eight male Sprague Dawley rats were randomly divided into four equal groups containing of seven rats per group. Group 1; peros physiological saline to the rat, Group 2; STZ 60 mg/kg/IP single dose, Group 3; PGE 10 mg/kg/20days/peros, Group 4; STZ as 60 mg/kg/IP + PGE 10 mg/kg/20days/peros. After the end of the experimental procedure, the rats were sacrificed, blood and testicular tissues were taken, and biochemical and histopathological examinations were performed. The administration of PGE was shown that the activities of CAT, SOD, GPx and GSH increased and the levels of MDA decreased in diabetic rats. Oral administration of PGE reduced the levels of LPO and improved the antioxidant activity in plasma and testis tissues while compared with the Groups 4. ...
The potential health-promoting effects of probiotics against intoxication by pesticides is a topi... more The potential health-promoting effects of probiotics against intoxication by pesticides is a topic of increasing commercial interest with limited scientific evidence. In this study, we aimed to investigate the positive effects of probiotic Saccharomyces boulardii on the male reproductive system under low dose neonicotinoid pesticide exposure conditions. We observed that acetamiprid and imidacloprid caused a degeneration and necrosis of the spermatocytes in the tubular wall, a severe edema of the intertubular region and a hyperemia. This was concomittant to increased levels of 8-hydroxy-2′-deoxyguanosine reflecting oxidative stress, and an increase in caspase 3 expression, reflecting apoptosis. According to our results, Saccharomyces boulardii supplementation mitigates these toxic effects. Further in vivo and clinical studies are needed to clarify the molecular mechanisms of protection. Quercetin, a potent antioxidant, has been shown to have a positive effect on semen in different species. This study aims to improve the efficiency of the Merino ram sperm using quercetin. The ejaculates collected from Merino rams were used for the dilution of the sperm (sperm density ≥ 3×109 spermatozoa/ml, motility ≥ 75%). Tris diluent was used to dilute the semen. The ejaculates were divided into 4 equal volumes at 32 °C. Three groups were formed by adding quercetin (15, 30, 45 μg/mL) at different doses. Antioxidant (quercetin) did not participate in the 4th group (the control group). Spermatozoa were diluted and frozen to approximately 4x108 spermatozoa per ml with Tris diluent. For this purpose, 6 Merino rams were used during the breeding season. The ejaculates were pooled after being collected from the rams. Pooled ejaculates were divided into 6 groups as control (C; no additive) Naringin (1, 2, and 4 mM) and Diosmin (2 and 4 mM) groups, and then reconstituted with TRIS-based diluent. Pooled semen was equilibrated, placed in 0,25 mL straws with 10 × 107 sperm cells in each straw and frozen in liquid nitrogen vapor. After waiting for 24 hours, the straws were thawed at 37 oC for 25 s and analyzes were made. There was no statistical difference in total motility between the groups. D2 and D4 preserved the integrity of the plasma membrane better than the other groups. Aim: The aim of this study was to investigate the effects of different doses of lipoic acid adde... more Aim: The aim of this study was to investigate the effects of different doses of lipoic acid added into dilution extender, following equilibration and freeze-thawing of ram semen on spermatologic and biochemical parameters. Materials and Methods: The ejaculates collected from 6 mature Konya Merino rams were used in the research. Ejaculates containing approximately 400x106 spermatozoa were diluted at 37 oC with the Tris-based extender containing lipoic acid at the doses of 1, 2, 4 mM and no additive (control), and then, they were frozen. Results: After equilibration, all doses of lipoic acid did not affect the sperm parameters whereas lipoic acid 1 mM (74.3%) ameliorated motility compared to both control (62.5%) and lipoic acid 4 mM (65.0%) (p 0.05). Conclusion: Addition of 1 mM lipoic acid to ram semen increases the motility after freeze-thawing process. However, dose treatments of lipoic acid in different extenders and animal species-breeds are required to be performed.

Ejaculates were collected via artificial vagina from four Merino rams and microscopically evaluated and pooled at 37 ◦C. The pooled semen samples were diluted in a Tris-based extender and separated to groups including curcumin (1, 2, 4 mM), ellagic acid (1, 2, 4 mM), methionine (1, 2, 4 mM) and no antioxidant (control). The diluted semen was slowly cooled to 5 ◦C and then the samples were frozen in liquid nitrogen vapour, plunged into liquid nitrogen (-196oC) for storage. Ejaculates were collected using artificial vaginas from five Merino rams, microscopically evaluated and pooled at 37 °C. The pooled semen samples were diluted in a Tris-based extender, including cysteine (2 mM and 4 mM), trehalose (10 mM and 25 mM) and no antioxidant (control). Diluted semen samples were kept in tubes and cooled from 37 to 5 °C in a cold cabinet, and maintained at 5 °C.
