En diversas degeneraciones retinianas, independientemente de su mecanismo primario, se ha observa... more En diversas degeneraciones retinianas, independientemente de su mecanismo primario, se ha observado un aumento del estres oxidativo que sufren los fotorreceptores y que produce la muerte progresiva de las distintas celulas de la retina. Como consecuencia de esta perdida celular la degeneracion retiniana conlleva la alteracion visual y la perdida gradual de otras funciones no visuales de la retina. Actualmente no hay tratamiento curativo frente a estas degeneraciones retinianas y es necesario profundizar en los mecanismos fisiopatologicos implicados para poder desarrollar herramientas de prevencion y diagnostico precoz y conseguir terapias curativas. Se estan estudiando diversas terapias para ralentizar la evolucion de la degeneracion, tales como: la terapia genica, el trasplante celular, los factores neurotroficos, los inhibidores de la mTOR y agentes antiapoptoticos y antioxidantes. En esta linea de antioxidantes se encuentran los flavonoides, que son un grupo ubicuo de sustancias polifenolicas que estan presentes en una gran variedad de plantas. El te verde contiene grandes cantidades de una catequina basada en flavonoides, el galato de epigalocatequina (EGCG). Varios estudios de laboratorio han demostrado que flavonoides como el EGCG son poderosos antioxidantes con multiples propiedades adicionales. El modelo animal P23H reproduce una forma de degeneracion retiniana autosomica dominante en el humano. Nuestro estudio ha tenido como objetivo caracterizar los distintos parametros de dano oxidativo de la degeneracion retiniana en el modelo animal P23H, con vistas a su posible correlacion con las pruebas funcionales durante la enfermedad y su aplicacion como marcadores para poder evaluar con precision la eficacia de las terapias antioxidantes, como el EGCG, en la degeneracion retiniana. En este trabajo se han analizado, en ratas normales de referencia y ratas P23H linea 1, la evolucion en edades de hasta 120 dias de los parametros de dano oxidativo mediante la cuantificacion de malonildialdehido y 4-hidroxialquenales, como un indicador del grado de peroxidacion lipidica; de dano nitrosativo, mediante la determinacion de nitritos; de defensas antioxidantes mediante la determinacion tanto de la actividad total antioxidante como de la actividad de la catalasa. Tambien se ha seguido la evolucion de la enfermedad valorando la funcion visual mediante optomotor y electrorretinografia. Asi mismo se ha estudiado el efecto protector del EGCG administrado intraperitonealmente a los animales P23H sobre los mismos parametros de dano oxidativo, defensas antioxidantes y funcionalidad visual de la retina. Nuestros resultados muestran una correlacion entre los parametros bioquimicos con las pruebas funcionales de evolucion de la enfermedad de modo que se pueden utilizar estas determinaciones en la evaluacion de la eficacia de los tratamientos antioxidantes frente a degeneraciones retinianas. Por otro lado, podemos concluir que el tratamiento con EGCG desciende los niveles tanto de dano oxidativo como nitrosativo, aumenta los niveles de defensas antioxidantes y mejora la funcionalidad visual en este modelo animal. Por lo tanto, se abre un campo de investigacion para determinar si la degeneracion retiniana puede ser mitigada por el EGCG con el fin de proporcionar un beneficio clinico en estos pacientes.

La expresión del ataque por radicales libres a los fosfolípidos de la membrana celular es el fenómeno de la peroxidación lipídica. Este proceso se inicia con la pérdida de átomos de hidrógeno de los restos acilo de los fosfolípidos, y con la formación de radicales peroxilo y alcoxilo, y lipoperóxidos, endoperóxidos e hidroperóxidos. A su vez, éstos inician una reacción en los fosfolípidos vecinos, propagando por toda la membrana la peroxidación, que sólo puede detenerse por el agotamiento del sustrato lipídico o la presencia de moléculas antioxidantes.
