To address the increasing concerns surrounding possible impacts of graphene-related materials on ... more To address the increasing concerns surrounding possible impacts of graphene-related materials on the aquatic environment, this study focused on computational predictions of binding between models of graphenes in the nm size range (nanographenes, nGs) and the aryl-hydrocarbon receptor (tAHR) and P450 cytochromes (tCYPs) of rainbow trout (Oncorhynchus mykiss). The tAHR plays a key role in the induction of detoxifying and early immune responses and tCYPs are essential for detoxifying planar hydrophobic chemicals such as nGs. After 3D modelling of those trout proteins, docking algorithms predicted the size-dependance profiles of nGs binding-scores to tAHR and tCYPs in the low nM range (high binding-affinities). Virtual oxidations of nGs to nGOs (carboxy-, epoxy-and/or hydroxy-oxidations) further lowered the corresponding binding-scores in level/type-oxidation manners. This work discovered computational star-shaped-similar molecules exhibiting lower binding-scores (higher-affinities) to S-SLSF with probable cross-binding properties of their targeted α-helices but with lower hydrophobicities and smaller molecular sizes. No... more Viral haemorrhagic septicaemia virus (VHSV) is one of the worst viral threats to fish farming. Non-virion (NV) gene-deleted VHSV (dNV-VHSV) has been postulated as an attenuated virus, because the absence of the NV gene leads to lower induced pathogenicity. However, little is known about the immune responses driven by dNV-VHSV and the wild-type (wt)-VHSV in the context of infection. Here, we obtained the immune transcriptome profiling in trout infected with dNV-VHSV and wt-VHSV and the pathways involved in immune responses. As general results, dNV-VHSV upregulated more trout immune genes than wt-VHSV (65.6% vs 45.7%, respectively), whereas wt-VHSV maintained more non-regulated genes than dNV-VHSV (45.7% vs 14.6%, respectively). Rainbow trout Ficoll-purified red blood cells (RBCs) cultured in vitro undergo morphological changes, especially when exposed to stress, and enter a new cell stage that we have coined shape-shifted RBCs (shRBCs). We have characterized these shRBCs using transmission electron microscopy (TEM) micrographs, Wright⁻Giemsa staining, cell marker immunostaining, and transcriptomic and proteomic evaluation. shRBCs showed reduced density of the cytoplasm, hemoglobin loss, decondensed chromatin in the nucleus, and striking expression of the B lymphocyte molecular marker IgM. In addition, shRBCs shared some features of mammalian primitive pyrenocytes (extruded nucleus surrounded by a thin rim of cytoplasm and phosphatidylserine (PS) exposure on cell surface).
