Although treatment for EC has improved with the introduction of Paclitaxel (Tax) chemotherapy, the majority of patients will develop resistance to the treatment, leading to poor prognosis. One of the causes of chemoresistance is the increased ability to undergo autophagy. In this study, we identified that miR-218 was significantly down-regulated in Tax-resistant EC cells compared to the non-drug resistant cell lines, and overexpression of miR-218 sensitized paclitaxel resistant EC cells to paclitaxel. Moreover, we demonstrated that miR-218 directly binds to the 3'-UTR of HMGB1 gene. In this study, the influence of NaOH and thermal pretreatment of dewatered activated sludge (DAS)... more In this study, the influence of NaOH and thermal pretreatment of dewatered activated sludge (DAS) on the high-solid solubilisation and anaerobic digestion was separately investigated by monitoring common parameters. The results indicated that COD, proteins and carbohydrates were efficiently solubilised in both NaOH and thermal pretreated DAS samples. For NaOH pretreatment, the concentrations of volatile fatty acids (VFAs) and total ammonium nitrogen (TAN) firstly increased followed by decreasing with NaOH dose increasing. However, they decreased with the severity of thermal pretreatment. During the batch digestion experiments (at 37°C), for 80mg NaOHg(-1) total solid (TS) DAS pretreatment it resulted in a 6.99% decrease in cumulative methane yield (CMY) compared to untreated DAS. While for 80, 100, 120°C and 20mg NaOH pretreatment, CMY increased by 15%, 42%, 71% and 35%, respectively, in comparison to untreated DAS.

Based on the results, the autotrophic denitrification supported by PIP could completely remove about 50 mg·L(-1) of nitrate within 4 days, and almost 80% of nitrate was changed into N2O (under acetylene blocking) without residual nitrite or ammonium. While only 53% of the nitrate was removed using acid-washed iron (AWI) instead of PIP, about 70% was converted into ammonium. Furthermore, a layer of FeOOH converted from hematite (α-Fe2O3) and magnetite (Fe3O4), which may block direct chemical nitrate reduction, was observed on the iron surface when PIP was used to support hydrogenotrophic denitrification. To explore the role of miRNAs in colorectal cancers (CRC), we have deep sequenced 48 pairs of fro... more To explore the role of miRNAs in colorectal cancers (CRC), we have deep sequenced 48 pairs of frozen CRC samples, of which 44 pairs produced high quality sequencing data. By using a combined approach of our bias reduction small RNA (smRNA) deep sequencing protocol and Illumina small RNA TruSeq method for sample bar coding, we have obtained data from samples of relatively large size with bias reduced digital profile results. This novel approach allowed us to validate many previously published results using various techniques to profile miRNAs in CRC tissues or cell lines and to characterize 'true' miRNA signatures highly expressed in colon/rectum (CR) or CRC tissues. The original biobed system comprises a clay layer at the bottom, a biomixture layer and a grass layer on top. The composition of the biomixture, which in the original Swedish model consisted of soil: peat: wheat straw (1:1:2 by volume), determines the efficiency of the system. Since the use of peat is not environmentally or economically feasible in China, this study tested the potential of replacing it with a locally available material, spent mushroom substrate (SMS). Three biomixtures containing different SMS (Pleurotus eryngii, Flammulina velutipes and Lentinus edodes) were compared with a biomixture containing peat, as in the original Swedish design, and a control containing soil alone. The fungicide chlorothalonil and the insecticide imidacloprid were used as model pesticides in the tests. Microbial activity (measured as respiration and phenoloxidase and hydrolytic activity) and pesticide dissipation were studied. Microbial activity was higher in the three biomixtures containing SMS than in the original-type biomixture. Among the SMS biomixtures, that containing SMS from L. edodes was the most biologically active. However, pesticide dissipation was comparable in all four biomixtures and significant differences were only found between biomixtures and the soil-alone control. Based on the physicochemical characteristics, biological activity and preliminary results on pesticide degradation, SMS are suitable and can therefore be used as a substitutes for peat.

Lovastatin is a proteasome inhibitor that enhances gefitinib-induced antiproliferation in non-small cell lung cancer. The objective of this study is to investigate the mechanism of lovastatin-induced antiproliferation in gefitinib-resistant human cholangiocarcinoma.Two gefitinib-resistant cholangiocarcinoma cell lines, SSP-25 and HuH-28, were used in this study to determine how to compensate gefitinib resistance. The combined effect of these two drugs was examined using the MTT assay, qPCR, immunoblotting, flow cytometry, and in vivo xenograft. Results indicated that lovastatin enhanced TNF-α-induced cell death in vitro. In addition, the combination of lovastatin with gefitinib enhanced accumulation of TNF-α. Furthermore, the treatment induced a synergistic cytotoxic effect and antiproliferation through apoptosis in SSP-25 cells and cell cycle arrest in HuH-28 cells. TiO2@(AC/Fe3O4) (AC is activated carbon) was prepared by using AC and Fe3O4 as joint support. The... more TiO2@(AC/Fe3O4) (AC is activated carbon) was prepared by using AC and Fe3O4 as joint support. The morphological features, crystal structure, and magnetism of the final product were characterized. The results indicate that TiO2 particles formed on the surface of AC and Fe3O4; the sizes of TiO2 and Fe3O4 were 0.5 and 0.7 μm respectively, and that of AC fell within a wide range. The highly crystalline cubic structures of the TiO2 particles was in accord with the standard X-ray diffractometry spectrum of magnetite and anatase. The maximum saturation magnetization of TiO2@(AC/Fe3O4) was 75 emu g(-1), which was enough to support magnetic recovery. The rate of methylene blue (MB) removal photocatalyzed by TiO2@(AC/Fe3O4) was higher by 50% than that achieved with AC/Fe3O4 photocatalysis, and similar to that achieved with TiO2@AC. The removal rate (kobs) decreased drastically from 1.77 × 10(-2) to 9.36 × 10(-3)min(-1) when the initial concentration of MB solution increased from 2.0 to 5.0 mg L(-1). The kobs value increased from 9.41 × 10(-3) to 1.34 × 10(-2)min(-1) with increasing photocatalyst dosage from 0.2 to 1.0 g, then slightly decreased to 1.33 × 10(-2)min(-1) at 2.0 g dosage.

In this study, we identified the presence of gene deletion and missense mutation leading to inactivation or underexpression of liver kinase B1 (LKB1) tumor suppressor and excluded the involvement of LKB1 gene hypermethylation in ICC tissues. Immunohistochemical analysis showed that LKB1 was underexpressed in a portion of 326 ICC tissues compared to their adjacent normal tissues. By statistical analysis underexpression of LKB1 in ICC tissues significantly correlated with poor survival and malignant disease characteristics in ICC patients. A combined system using nano MgO and nanofiltration (NF) membrane was established to purify pollu... more A combined system using nano MgO and nanofiltration (NF) membrane was established to purify polluted water in this experiment. The turbidity, permanganate index, UVA254, colony counts and the concentrations of NO3-, NO2-, NH4+, Fe, Mn and Mg of the effluents from each unit of this combined system were measured to investigate the pollutant removal of this system. Based on the results obtained, the combined nano MgO-NF system could efficiently remove many kinds of pollutants in this experiment, including organic matter, nitrogen species, heavy metals, suspended solids and bacteria. And the effluents could meet the standard of drinking water. Furthermore, increasing the nano MgO dosage could not elevate the removal ratio of the pollutants, but only increase the Mg content of the effluent. Thus, 0.05 g L(-1) of nano MgO may be a suitable dosage for 2,000 L of polluted water treatment. Also, the operating pressure of NF membrane had no significant effect on pollutant removal when the operating pressure of NF membrane was increased from 0.3 to 0.9 MPa.

This key enzyme in deoxyribonucleotide biosynthesis is the target of established clinical agents such as hydroxyurea (HU) and gemcitabine because of its critical role in DNA replication and repair. Herein we report that BRCA-1-defective human breast cancer cells are more sensitive than wild-type BRCA-1 counterparts to COH29 in vitro and in vivo. Microarray gene expression profiling showed that COH29 reduces expression of DNA repair pathway genes, suggesting that COH29 interferes with these pathways. The turbidity, permanganate index, UVA254, colony counts and the concentrations of NO3-, NO2-, NH4+, Fe, Mn and Mg of the effluents from each unit of this combined system were measured to investigate the pollutant removal of this system. Based on the results obtained, the combined nano MgO-NF system could efficiently remove many kinds of pollutants in this experiment, including organic matter, nitrogen species, heavy metals, suspended solids and bacteria. And the effluents could meet the standard of drinking water. Furthermore, increasing the nano MgO dosage could not elevate the removal ratio of the pollutants, but only increase the Mg content of the effluent. Thus, 0.05 g L(-1) of nano MgO may be a suitable dosage for 2,000 L of polluted water treatment. Also, the operating pressure of NF membrane had no significant effect on pollutant removal when the operating pressure of NF membrane was increased from 0.3 to 0.9 MPa.
The aim of this study was to determine the presence of high-risk HPV-16 in patients with HNSCC, a... more The aim of this study was to determine the presence of high-risk HPV-16 in patients with HNSCC, assess the impact of HPV status on treatment response and survival in this select cohort treated with combined modality therapy and to identify the differences in HIF-1alpha and VEGF expression in HPV-positive and -negative tumors. Patients had resectable, untreated stage III, IV HNSCC of the oral cavity, oropharynx, hyopharynx or larynx, and stage II cancer of the base of tongue, hypopharynx and larynx. HPV status was determined by conventional PCR in fresh frozen biopsy samples and by Taqman PCR assay on formalin-fixed, paraffin-embedded specimens. HIF-1alpha and VEGF expression were assessed by quantitative real-time PCR (RT-PCR). Multivariate Cox proportional hazards regression analysis was used to calculate hazard ratios (HRs) and 95% confidence intervals (CIs) based on HPV status. COH29, a novel antimetabolite drug developed at City of Hope Cancer Center, has anticancer activi... more COH29, a novel antimetabolite drug developed at City of Hope Cancer Center, has anticancer activity that stems primarily from the inhibition of human ribonucleotide reductase (RNR). This key enzyme in deoxyribonucleotide biosynthesis is the target of established clinical agents such as hydroxyurea (HU) and gemcitabine because of its critical role in DNA replication and repair. Herein we report that BRCA-1-defective human breast cancer cells are more sensitive than wild-type BRCA-1 counterparts to COH29 in vitro and in vivo. Microarray gene expression profiling showed that COH29 reduces expression of DNA repair pathway genes, suggesting that COH29 interferes with these pathways. It is well-established that BRCA1 plays a role in DNA damage repair, especially homologous recombination (HR) repair, to maintain genome integrity.
