Several reproducible animal experimental models, in which human physiologic conditions can be mimicked, are available for studying glioblastoma multiforme. In an ideal rat model, the tumor is of glial origin, grows in predictable and reproducible patterns, closely resembles human gliomas histopathologically, and is weakly or nonimmunogenic. In the current study, we used MRI and histopathologic evaluation to compare the most widely used allogeneic rat glioma model, C6-Wistar, with the F98-Fischer syngeneic rat glioma model in terms of percentage tumor growth or regression and growth rate. Methotrexate-loaded biocompatible nanoparticles were tested for preliminary efficacy in glioma tr... more Methotrexate-loaded biocompatible nanoparticles were tested for preliminary efficacy in glioma treatment. Behenic acid nanoparticles, prepared by the coacervation method, were loaded with the ester prodrug didodecylmethotrexate, which was previously tested in vitro against glioblastoma human primary cultures. Nanoparticle conjugation with an ApoE mimicking chimera peptide was performed to obtain active targeting to the brain. Biodistribution studies in healthy rats assessed the superiority of ApoE-conjugated formulation, which was tested on an F98/Fischer glioma model. Differences were observed in tumor growth rate (measured by MRI) between control and treated rats. In vitro tests on F98 cultured cells assessed their susceptibility to treatment, with consequent apoptosis, and allowed us to explain the apoptosis observed in glioma models.
Aberrant activation of the canonical Wingless-type MMTV integration site family (WNT)/β-catenin s... more Aberrant activation of the canonical Wingless-type MMTV integration site family (WNT)/β-catenin signaling pathway is critical for gliomas. In 74 gliomas of different histological grade and in 24 glioblastoma cell lines, protein expression of WNT member 3a (WNT3a), β-catenin and transcription factor 4 (TCF4) was investigated by immunohistochemistry, western blotting, immunofluorescence and immunocytochemistry. In tumors and cell lines, WNT3A expression was assessed at the mRNA level by quantitative real-time polymerase chain reaction. WNT3a was overexpressed at the protein and mRNA levels in malignant astrocytic tumors and cell lines. Cytoplasmic expression of β-catenin was detected in high-grade gliomas and cell lines, with evidence of nuclear translocation on fractionated protein extracts. GAGs accumulate in the oligodendrogliomas and in the oligodendroglial component of mixed gliomas ... more GAGs accumulate in the oligodendrogliomas and in the oligodendroglial component of mixed gliomas induced in rats by transplacental ENU. We studied the period between birth and the appearance of the earliest tumoral lesions and did not find any accumulation preceding the early neoplastic proliferations. These findings, together with the observation that GAGs are present only in the oligodendroglial component of tumors, are in agreement with our previous interpretation that the tumoral transformation of oligodendroglia takes place after it has acquired the ability to interfere with GAG metabolism during myelinogenesis.
Recent surveys indicate a decreasing north-to-south gradient in the mortality and incidence rates... more Recent surveys indicate a decreasing north-to-south gradient in the mortality and incidence rates of amyotrophic lateral sclerosis (ALS) in Italy, possibly indicating a different susceptibility to ALS in these populations. Piemonte, a region of northwestern Italy, experienced a considerable migration from other regions in Italy between 1940 and 1975; we therefore analyzed the effects of place of birth and migration upon the risk of developing ALS. Data on all ALS cases occurring in Piemonte during the period 1971-1990 were collected. Standardized incidence ratios (SIRs) for patients born outside Piemonte were calculated, using the Piemonte-born population as reference. A total of 962 ALS cases were identified during the study period, corresponding to a mean annual crude incidence rate of 1.37/100,000 population (95% confidence interval, 1.29-1.46). The SIRs of patients born in three southern Italian regions and of foreign-born persons were significantly higher than those of persons born in Piemonte and or other regions in Italy, and increased with age. This observation may be explained by an interaction between environmental and genetic factors or by selective migration.
We report the case of a girl who developed leukoencephalopathy and adrenal atrophy and died at 3 ... more We report the case of a girl who developed leukoencephalopathy and adrenal atrophy and died at 3 years of age. Histologically, demyelination, gliosis, perivascular lymphocytic cuffing and sudanophilia were present in the brain. The adrenal cortex was atrophic. Ultrastructurally, there were numerous cytoplasmic inclusions in brain macrophages, consisting of two leaflets separated by an intervening space of variable low electron density. Brain tissue cholesterol esters contained a high proportion of long chain fatty acids. The findings are discussed and compared with those in the literature. Emphasis is placed on the fact that the disease occurred in a girl in apparent contradiction to the commonly accepted X-linked hereditary transmission of adrenoleukodystrophy. Some possible genetic explanations for our case are put forward.
Non è semplice indagare l’influenza dello stato emotivo dell’esaminatore sull’analisi che sta com... more Non è semplice indagare l’influenza dello stato emotivo dell’esaminatore sull’analisi che sta compiendo al microscopio e, più ancora, l’influenza del suo vissuto generale, quello che insieme alla memoria e alla coscienza corrisponde alla sua identità. Forse però è ancora più interessante spiegare l’inverso e cioè le evocazioni che gli oggetti nel campo microscopico operano sul vissuto individuale e in particolare sulla memoria “implicita”,meglio conosciuta come inconscio. È un argomento di difficile discussione perché non può essere fondato su basi anatomiche o organiche reali, ma poggia su concetti il più delle volte opinabili o comunque su substrati non dimostrabili. In primo luogo mi riferisco al primo stadio della percezione, quando questa subisce le “inferenze inconsce” secondo Von Helmholtz1, rapide e inconsapevoli, che rappresentano correzioni della percezione in base all’esperienza passata mediante atti di giudizio. Ne ho già parlato a proposito del riconoscimento e delle interpretazioni,e riprendo qui il discorso perché si tratta molto verosimilmente di fenomeni iponoici, incontrollabili e automatici, perché poggianti sull’esperienza implicita e l’abitudine, ma anche sulla memoria esplicita oppure persino sulle proprietà dell’insieme dell’apparato recettore.

L’invenzione del microscopio, avvenuta un secolo prima, ha consentito l’esplorazione della realtà biologica ben al di là delle possibilità dell’occhio umano consentendo l’esplorazione a livello cellulare,che ha cominciato a dare frutti proprio in quell’epoca. Come ho già detto, per una sempre migliore osservazione il materiale da esaminare è stato progressivamente elaborato in modo sempre più complesso per poter essere studiato al microscopio e giungere a noi sotto forma di percezione visiva.Questa ovviamente è rimasta la stessa, ma la qualità e quantità delle immagini sono fortemente migliorate nel tempo. Alla classica istologia a un certo punto si sono aggiunte la chimica, poi l’istochimica, l’istoenzimologia, l’immunoistochimica, senza contare la microscopia elettronica, l’osservazione in fluorescenza e molte altre tecnicalità che includono per esempio il cell sorting, il miglioramento delle colture in vitro fino all’analisi molecolare della cellula singola con tecniche laser. Si è scesi a livelli di ingrandimento impensabili fino a qualche decennio fa con la microscopia elettronica e fino all’uso del microscopio a scansione. Anche il microscopio ottico ha subito una profonda evoluzione fino agli attuali sofisticati microscopi polifunzionali.Rimane però incontrovertibile che la via finale dell’introito dell’informazione è rimasta la percezione visiva. I progressi tecnici verificatisi nei vari campi l’hanno arricchita e affinata, ma contemporaneamente hanno anche profondamente modificato il vissuto specifico che gioca un ruolo fondamentale nell’interpretazione finale.

Le tecniche chirurgiche sono cambiate nel tempo e così quelle di neuroradiologia (Risonanza Magnetica funzionale, a diffusione, a più di 1, 5 Tesla, spettroscopia, angiografia, etc.) che hanno consentito diagnosi più precoci. Giungono al tavolo operatorio tumori più piccoli e si cerca di salvaguardare di più il tessuto sano. Anche per le modalità tecniche di asportazione del tumore, la quota di tessuto che giunge al patologo è nettamente diminuita rispetto a un tempo. Come ho già detto nel capitolo “L’attenzione”, non è raro il caso in cui questa quota non sia rappresentativa del tumore e il patologo debba utilizzare per la diagnosi elementi estranei alla patologia, come per esempio l’aspetto macroscopico fornito dal neurochirurgo e soprattutto quello neuroradiologico, e fornire al neurochirurgo formule diagnostiche interlocutorie. L’etichetta diagnostica del patologo diventa non più trasferibile direttamente alla terapia, ma dev’essere mediata. Spesso il patologo si sente in obbligo di informare il neurochirurgo su quale sia l’interpretazione da dare alla sua risposta. Molto meglio sarebbe se il neurochirurgo conoscesse a fondo la dinamica biologica dei tumori per interpretare la diagnosi fornitagli dal patologo. Questo vale anche per il neuroradiologo e per il neuro-oncologo. Ma, è sempre così? Capita che la preparazione biologica sui tumori lasci a desiderare. Nella mia esperienza ne ho conosciuti più d’uno che hanno regolarmente studiato al microscopio i tumori da loro operati.
