This 60 day study investigated the effects of organically certified nucleotide yeast-derived prot... more This 60 day study investigated the effects of organically certified nucleotide yeast-derived protein source (Nu-Pro®) on growth, feed efficiency, fillet proximate composition and digestive enzymes in rainbow trout (Onchorhyncus mykiss). Diets were isonitrogenous and isocaloric. Three experimental diets wherein 20%, 40% and 60% of the fish meal content was replaced by Nu-Pro®, were compared to the control diet in which the crude protein content was anchovy fish meal and corn gluten meal. The rainbow trout (initial weight 27 g/fish) were fed twice daily in 200-l Aqaria. Live weight gain increase ranged from 125-195% in fish fed the experimental diets. Results indicated that up to 40% fish meal protein can be replaced by Nu-Pro® without compromising growth rates, feed efficiency or the fillet biochemical composition in the rainbow trout. Glutathione S-transferases (GSTs) are enzymes involved in the metabolism of many disease-causing ... more Glutathione S-transferases (GSTs) are enzymes involved in the metabolism of many disease-causing electrophilic substrates and protect the cells against oxidative stress. In the present study, we investigated the GSTM1, GSTT1 and GSTP1 gene polymorphisms in diabetic patients and healthy individuals and searched whether polymorphisms in GST genes are associated with diabetes mellitus (DM) in the Turkish population. The study population consisted of 98 unrelated healthy individuals and 98 patients with DM. Genotyping of GSTM1, GSTT1 and GSTP1 genes was performed using real time polymerase chain reaction with a Light Cycler instrument. Patients had a higher frequency of the GSTM1 null genotype than the control group (Odds ratios, OR = 3.7; 95% confidence intervals, CI = 2.05-6.70). However, there was no significant difference in the frequencies of the GSTT1 and GSTP1 gene polymorphisms between the patients and control group. The combined analysis of these three GST genotypes showed a further DM risk increase (OR = 5.7, 95% CI = 1.51-31.07). This is the first study to determine the association of diabetes with GST gene polymorphism in the Turkish population. These results show that GSTM1 null genotype may play a significant role in the aetiopathogeneses of DM and the GSTM1 gene may be a useful marker in the prediction of DM susceptibility of the Turkish population.

Pure and extract forms of E.E are applied to the nasal cavity of rats for rhinosinusitis. Nasal mucosa, soft palate and trachea of rats were harvested in the first hour, 3rd day and 3rd week for histopathological evaluation. This experimental study revealed antiinflammatory effects of E.E. It has also been shown that E.E reduced fibrosis. The antiinflammatory effect of all forms of E.E has been found to be at their maximum level in the 3rd day with a decreasing manner to 3rd week. Additionally, pure form of E.E caused severe epithelial loss in hyperacute stage. E.E also reduced fibrosis besides its antiinflammatory effect. The action mechanism of antiinflammatory effect of different levels of extract forms need to be further analysed through related enzymatic pathways by different dose and time studies.

In this study, it was aimed to investigate Zinc Phthalocyanine-1 (ZnPc-1) against SARS-CoV-2 main protease (MPro) by in silico method. In our research, Zn-Pc-1 was enabled to target the active site of the SARS-CoV-2 MPro receptor with PDB ID:6LU7. Molecular docking study was done with Autodock program. The result of molecular docking was found to be -10.6 kcal/mol. Absorption, distribution, metabolism, excretion, and toxicity (ADMET) screening of ligands assists US to state their absorption properties, toxicity, and drug-likeness. Ligand molecules obtained from PubChem in smiles format were loaded on SWISSADME and PROTOX-II webservers for ADMET screening. The three compounds in propolis were obtained from the PubChem database. Compounds were located at the active site of the SARS-CoV-2 MPro receptor with PDB ID:6LU7. Molecular docking work was done with Autodock program. Molecular docking results were found as -8.7 kcal/mol in limonin, -7.5 kcal/mol in quercetin and -7.7 kcal/mol in kaempferol. The effects of punicalagin and punicalin on adipogenesis were investigated in this study. To examine the effect of punicalagin and punicalin on adipocyte differentiation, various concentrations of punicalagin and punicalin (2-10 µM) were applied to differentiated 3T3-L1 cells. Glyceraldehyde-3-phosphate dehydrogenase (GPDH) activity, Oil red O staining, intracellular triglyceride levels, and gene expressions of transcription factors (Peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT-enhancer-binding proteins-α (C/EBPα), Sterol regulatory element-binding protein 1c (SREBP-1c)) and lipolysis-associated genes (hormone-sensitive lipase (HSL), Perilipin A, tumor necrosis factor-α (TNF-α)) were examined in order to investigate the effects of punicalagin and punicalin on adipocyte differentiation. Punicalagin and punicalin applications caused a continuous decrease in cell size and intracellular triglyceride accumulation. GPDH activity and transcription gene expressions decreased significantly in groups that were applicated punicalagin and punicalin at high concentrations. Punicalagin, but not punicalin, down-regulated the expression of HSL and perilipin A and up-regulated the expression of TNF-α in a dose-dependent manner. In conclusion, both punicalagin and punicalin were able to inhibit the adipocyte differentiation.
Abstract Aim: Major side effects of cyclophosphamide administration are immunosuppression and mye... more Abstract Aim: Major side effects of cyclophosphamide administration are immunosuppression and myelosuppression. The immunomodulatory effects of plant bioactive compounds on chemotherapy drug-induced immunosuppression may have significant effects in cancer treatment. For this reason, we investigated the immunomodulatory effect of myricetin, apigenin, and hesperidin in cyclophosphamide-induced immunosuppression in rats. Methods: In our study, a total of 64 rats were used, and divided into eight equal groups. These groups were: control, cyclophosphamide, cyclophosphamide + myricetin (100 mg/kg), cyclophosphamide + myricetin (200 mg/kg), cyclophosphamide + apigenin (100 mg/kg), cyclophosphamide + apigenin (200 mg/kg), cyclophosphamide + hesperidin (100 mg/kg), and cyclophosphamide + hesperidin (200 mg/kg). Myricetin, apigenin, and hesperidin pretreatments were performed for 14 d, while cyclophosphamide application (200 mg/kg) was performed only on the 4th day of the study. Levels of humoral antibody production, quantitative hemolysis, macrophage phagocytosis, splenic lymphocyte proliferation, and natural killer cell cytotoxicity were determined. In addition, we measured pro-inflammatory cytokines, and followed lipid peroxidation and antioxidant markers and examined the histology of bone marrow, liver and spleen in all groups. Results: During cyclophosphamide treatment, all three phytochemicals increased the levels of humoral antibody production, quantitative hemolysis, macrophage phagocytosis, splenic lymphocyte proliferation, antioxidant markers, and natural killer cell cytotoxicity. Moreover, the agents decreased the levels of pro-inflammatory cytokines and mediators, reduced lipid peroxidation markers, and reduced tissue damage in liver, spleen, and bone marrow. Conclusion: Our study demonstrated that myricetin, apigenin, and hesperidin can reduce the immunosuppressive effect of cyclophosphamide by enhancing both innate and adaptive immune responses, and these compounds may be useful immunomodulatory agents during cancer chemotherapy.
