This study was being carried since November 2011 and so on Sam- ples that used for water quality parameters analysis were collected once in a month. Water quality parameters, dissolved oxygen (DO), salinity, pH, temperature and electrical conduc- tivity (E.C) was measured directly in the field with the help of devices. Other water param- eters as suspended solids (SS), chemical oxygen demand (COD), total alkalinity, total hardness, total ammonia nitrogen, nitrite, nitrate, ammonia, phosphate, sulfate, sulfite, free chlorine, sodium, magnesium, calcium, lead, copper and cadmium are analyzed. In this study conducted for Horohon (Hafik-Sivas) water quality of the stream is followed by wa- ter quality methods during a year. Furthermore, the present state of water quality was found to be suitable for trout farming.
Carbonic anhydrase (CA) from erythrocytes of rainbow trout Oncorhynchus mykiss was purified by af... more Carbonic anhydrase (CA) from erythrocytes of rainbow trout Oncorhynchus mykiss was purified by affinity chromatography through a process comprising hemolysate preparation, sepharose- 4B-l-tyrosine-sulfanylamide affinity gel chromatography, and dialysis. Then the effects of three antibiotics—sodium ampicillin, streptomycin, and trimethoprim plus sulfamethoxazole—on this enzyme were investigated in vitro and in vivo. Inhibition effects were determined in vitro by means of the Lineweaver–Burk and activity–antibiotic regression graphs. The dissociation constant of the enzyme inhibitor complex (Ki) and 50% inhibitory values were 2.16 ± 0.40 and 1.87 mM for sodium ampicillin, 7.40 ± 0.310 and 5.21 mM for streptomycin, and 18.50 ± 0.35 and 9.74 mM for trimethoprim–sulfamethoxazole. The in vivo experiment demonstrated that CA was significantly inhibited by trimethoprim–sulfamethoxazole but not by sodium ampicillin and streptomycin.

Toplam aerobik mezofilik bakteri sayilari su, deri ve yem numunelerinde sirasiyla 101-104 kob/ml, 102-105 kob/g, 103-108 kob/g olarak tespit edilmistir. Oxygenating and degassing columns, simple effective devices for economically adding oxygen to wat... more Oxygenating and degassing columns, simple effective devices for economically adding oxygen to water, have been used successfully in aquaculture. Increasing the oxygen content of water enables fish culturists to increase fish loads and improve the rearing environment. Tests conducted in the present study were to evaluate the effectiveness of the column, injected pure oxygen or air into, at a rainbow trout hatchery. For this purpose, concentration of dissolved oxygen and carbon dioxide in the column outflow were monitored. Although injecting pure oxygen increased the oxygen concentration and reduced the carbon dioxide, injecting air had a little effect on the carbon dioxide level. Notwithstanding, the oxygen and carbon dioxide concentrations were not high enough to affect fish production, we recommended that users of the air injected column closely monitor the concentration of the gases to avoid from negative effects.
Abstract Rainbow trout broodstocks were fed diets with varying levels of diludine [0 (D0; control... more Abstract Rainbow trout broodstocks were fed diets with varying levels of diludine [0 (D0; control), 0.25 (D1), 0.5 (D2) and 1 g kg−1 (D3)] during 2 consecutive reproduction seasons. Fish were started to be fed 3 months before spawning for the first reproduction season and they were kept on the same diets as well as an additional dietary group with 3 g kg−1 dietary diludine (D4) until the following reproduction season, providing short and long-term dietary treatments. Gametes storage at 6 °C for four and fifteen days prior to the fertilization in the first and second reproduction season, respectively, was also tested. Short-term feeding trial did not influence reproductive performance when the eggs were fertilized immediately after stripping, whereas survival at eyed-stage embryos significantly decreased in eggs, from fish fed diludine free diet, fertilized after 4-day storage. Dietary diludine level corresponded with increasing level of vitamin E in the eggs, which positively correlated with survival at eyed-stage embryos from the eggs with or without 15-day storage after long-term feeding. Fish fed the diet with highest level of diludine (D4) produced eggs higher in number but smaller in size in comparison to those from the other four experimental groups. In eggs fertilized after 15-day storage, survival rate was 20-fold higher in D4 diet fed fish in comparison to the other experimental groups. Our overall results indicate that dietary diludine has vitamin E sparing effect in the eggs and protects gamete viability during the storage when it is provided at 3 mg kg−1 dietary level, which result in lower lipid peroxidation and higher reproductive performance.
