The water rat, Nectomys squamipes, closely involved in schistosomiasis transmission in Brazil, ha... more The water rat, Nectomys squamipes, closely involved in schistosomiasis transmission in Brazil, has been found naturally infected simultaneously by Schistosoma mansoni and Echinostoma paraensei. Laboratory experiments were conducted to verify parasitic interaction in concurrent infection. It was replicated four times with a total of 42 water rats and essayed two times with 90 mice pre-infected with E. paraensei. Rodents were divided into three groups in each replication. A wild strain recently isolated from Sumidouro, RJ, and a laboratory strain of S. mansoni from Belo Horizonte (BH) was used. Rats infected with E. paraensei were challenged 4 weeks later with S. mansoni and mice 2 or 6 weeks after the infection with S. mansoni. Necropsy took place 8 weeks following S. mansoni infection. The N. squamipes treatment groups challenged with S. mansoni RJ strain showed a significant decrease (80 and 65%) in the S. mansoni parasite load when compared with their respective control groups. There was a significant change or no change in the hosts challenged with the BH strain. The persistence time of E. paraensei within host was extended in relation to control groups, with a consequent enhancement of the number of recovered worm. An E. paraensei strain-specific influence on S. mansoni parasitism is reported. This paper presents some experimental data about this interaction in N. squamipes and Mus musculus.
Linkage studies have identified the human leukocyte antigen (HLA)-DRB1 as a putative rheumatoid a... more Linkage studies have identified the human leukocyte antigen (HLA)-DRB1 as a putative rheumatoid arthritis (RA) susceptibility locus (SL). Nevertheless, it was estimated that its contribution was partial, suggesting that other non-HLA genes may play a role in RA susceptibility. To test this hypothesis, we conducted microarray transcription profiling of peripheral blood mononuclear cells in 15 RA patients and analyzed the data, using bioinformatics programs (significance analysis of microarrays method and GeneNetwork), which allowed us to determine the differentially expressed genes and to reconstruct transcriptional networks. The patients were grouped according to disease features or treatment with tumor necrosis factor blocker. Transcriptional networks that were reconstructed allowed us to identify the interactions occurring between RA SL and other genes, for example, HLA-DRB1 interacting with FNDC3A (fibronectin type III domain containing 3A). Given that fibronectin fragments can stimulate mediators of matrix and cartilage destruction in RA, this interaction is of special interest and may contribute to a clearer understanding of the functional role of HLA-DRB1 in RA pathogenesis.

The identification of rhG-CSF was carried out in pharmaceutical preparations by non-reducing poly... more The identification of rhG-CSF was carried out in pharmaceutical preparations by non-reducing polyacrylamide gel electrophoresis and western blotting with specific antibodies, showing a single band in the 19 kDa region. The potency was assessed by the neutropenia mouse bioassay giving values between 88.4 and 122.4% of the stated potency. The precision index expressed by weight was between 141 and 432 for the independent assays. Batch-to-batch, the samples met the requirements for the safety test and bacterial endotoxins test. The biological and immunological results showed the quality of the products in clinical use and the specifications established contribute to assuring the safety and efficacy of biological medicines. The physiological performance of acariquara (Minquartia guianensis) seedlings submitted to water ... more The physiological performance of acariquara (Minquartia guianensis) seedlings submitted to water deficit and the recovery of physiological parameters during rehydration were investigated in a greenhouse experiment. The analyzed parameters were: leaf water potential, gas exchange and chlorophyll a fluorescence. After thirty-five days, non-irrigated plants exhibited a leaf water potential 70 % lower compared to control plants (irrigated daily) and the stomatal conductance reached values close to zero, inducing a severe decrease in gas exchange (photosynthesis and transpiration). Six days after the beginning of the rehydration of drought-stressed plants, the results demonstrated that water stress did not irreversibly affect gas exchange and quantum efficiency of photosystem II (PSII) in M. guianensis seedlings, since four to six days after rehydration the plants exhibited total recovery of the photosynthetic apparatus. We conclude that M. guianensis presented good tolerance to water stress and good capacity to recover the physiological performance related to leaf water status, photosynthesis and photochemical efficiency of PS II under hydric stress, suggesting substantial physiological plasticity during the juvenile phase for this tree species.

A fast, sensitive, and specific liquid chromatography-tandem mass spectrometry (LC-MS/MS) method ... more A fast, sensitive, and specific liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for the simultaneous determination of quinine and doxycycline in pharmaceutical formulations. The LC-MS/MS method was carried out on a Sun Fire Waters C18 column (50 mm × 3.0 mm I.D.) and the mobile phase consisted of acetonitrile:0.1% formic acid (75:25, v/v), run at a flow rate of 0.45 mL/min (split 1:3). The injection volume was 10 µL for both standard and samples. The triple quadrupole mass spectrometer equipped with an electrospray source in positive mode (ES+) was set up in multiple reaction monitoring mode (MRM), monitoring the transitions of 325.0 > 307.0 and 445.0 > 428.1, for quinine and doxycycline, respectively. The total analysis time was 2 min and the method was linear in the concentration range of 10–1500 ng/mL for both compounds. Method validation investigated parameters such as the specificity, linearity, precision, accuracy, and robustness, giving results within the acceptable range. Moreover, the proposed method was successfully applied for determination of quinine and doxycycline in nanocapsule formulations to support the quality control.
