Due to unique optical properties of high birefringent (Hi-Bi) fibers for sensing and coherent opt... more Due to unique optical properties of high birefringent (Hi-Bi) fibers for sensing and coherent optical communications, there is a strong interest in process optimization at preform fabrication and fiber drawing stages. Boron-doped silica cladding acts as stress-applying part resulting in polarization properties of Hi-Bi fibers that are strongly dependent on chemical composition. Using modified chemical vapor deposition (MCVD) technique, B2O3-doped silica preform rods were synthesized under different precursor gas flow conditions. Qualitative information about B2O3-SiO2 system composition was derived from etching behavior in nonbuffered HF solution and subsequent microstructural examination using scanning electron microscope. Significant degree of B2O3 incorporation was seen in case of high BCl3:SiCl4 ratio and mild oxygen-deficient processing conditions. Increasing the B2O3 content to ~26 mol% led to a corresponding increase in coefficient of thermal expansion (CTE) to a maximum value of 2.35 ppm/K. The value of refractive index (RI), on the other hand, was found to decrease with increased B2O3 incorporation. A qualitative correlation between B2O3 and SiO2 system composition and physical properties such as CTE and RI was established.
We have carried out a systematic investigation of salts- and alcohols-induced conformational alte... more We have carried out a systematic investigation of salts- and alcohols-induced conformational alterations on the trifluoroacetic acid (TFA)-treated ferricytochrome c by soret absorption spectroscopy, far UV circular dichroism (CD), tryptophan fluorescence, and 1-anilino-8-naphthalene sulfonate (ANS) binding. TFA induces the unfolding of native cytochrome c obtained from horse heart leading to loss of secondary structure. The addition of increasing concentration of salts and alcohols leads to increase in MRE value at 222 and 208 nm indicating an increase in the α-helical content leading to formation of compact dimensional structure. Cytochrome c is a heme protein in which the resonance energy of tryptophan is transferred to heme resulting in quenched tryptophan fluorescence. Addition of alcohols leads to increase in tryptophan and ANS fluorescence. The tryptophan and ANS fluorescence in case of salts shows decreased fluorescence intensity. TFA-induced unfolded cytochrome c showed the soret absorption maximum at 394 nm. However, an intermediate state in presence of alcohols and salts showed the absorption maxima at 398 nm and 402 nm, respectively. Among all the salts and alcohols studied, K3Fe(CN)6 and butanol were found to be most effective as examined by the above-mentioned spectroscopic techniques. The order of effectiveness of alcohols was found to be butanol > propanol > ethanol > methanol. The following effective trend in the case of salts was obtained: K3Fe(CN)6 > K2SO4>KClO4 > KCl. These results suggest that alcohols induce an intermediate with molten globule–like conformation on the TFA unfolded state, whereas salts induce a refolded intermediate approaching native-like conformation.

Oral administration of vanadate to diabetic animals have been shown to stabilize the glucose home... more Oral administration of vanadate to diabetic animals have been shown to stabilize the glucose homeostasis and restore altered metabolic pathways. However, vanadate exerts these effects at relatively high doses with several toxic effects. Low doses of vanadate are relatively safe but unable to elicit any antidiabetic effects. The present study explored the prospect of using low doses of vanadate with Trigonella foenum graecum, seed powder (TSP), another antidiabetic agent, and to evaluate their antidiabetic effect in diabetic rats. Alloxan diabetic rats were treated with insulin, vanadate, TSP and low doses of vanadate with TSP for three weeks. The effect of these antidiabetic compounds was examined on general physiological parameters, Na+/K+ ATPase activity, membrane lipid peroxidation and membrane fluidity in liver, kidney and heart tissues. Expression of glucose transporter (GLUT4) protein was also examined by immunoblotting method in experimental rat heart after three weeks of diabetes induction. Diabetic rats showed high blood glucose levels. Activity of Na+/K+ ATPase decreased in diabetic liver and heart. However, kidney showed a significant increase in Na+/K+ ATPase activity. Diabetic rats exhibited an increased level of lipid peroxidation and decreased membrane fluidity. GLUT4 distribution was also significantly lowered in heart of alloxan diabetic rats. Treatment of diabetic rats with insulin, TSP, vanadate and a combined therapy of lower dose of vanadate with TSP revived normoglycemia and restored the altered level of Na+/K+ ATPase, lipid peroxidation and membrane fluidity and also induced the redistribution of GLUT4 transporter. TSP treatment alone is partially effective in restoring the above diabetes-induced alterations. Combined therapy of vanadate and TSP was the most effective in normalization of altered membrane linked functions and GLUT4 distribution without any harmful side effect.

Trigonella foenum graecum seed powder (TSP) and sodium orthovanadate (SOV) have been reported to ... more Trigonella foenum graecum seed powder (TSP) and sodium orthovanadate (SOV) have been reported to have antidiabetic effects. However, SOV exerts hypoglycemic effects at relatively high doses with several toxic effects. We used low doses of vanadate in combination with TSP and evaluated their antidiabetic effects on antioxidant enzymes and membrane-linked functions in diabetic rat brains. In rats, diabetes was induced by alloxan monohydrate (15 mg/100 g body wt.) and they were treated with 2 IU insulin, 0.6 mg/ml SOV, 5% TSP and a combination of 0.2 mg/ml SOV with 5% TSP for 21 days. Blood glucose levels, activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), Na+/K+ ATPase, membrane lipid peroxidation and fluidity were determined in different fractions of whole brain after 21 days of treatment. Diabetic rats showed high blood glucose (P < 0.001), decreased activities of SOD, catalase and Na+/K+ ATPase (P < 0.01,P < 0.001 andP < 0.01), increased levels of GPx and MDA (P < 0.01 andP < 0.001) and decreased membrane fluidity (P < 0.01). Treatment with different antidiabetic compounds restored the above-altered parameters. Combined dose ofTrigonella and vanadate was found to be the most effective treatment in normalizing these alterations. Lower doses of vanadate could be used in combination with TSP to effectively counter diabetic alterations without any toxic effects.

ABSTRACT Development of the methods for generating distinct mechanisms derived from a given famil... more ABSTRACT Development of the methods for generating distinct mechanisms derived from a given family of kinematic chains has been persued by a number of researchers in the past, as the distinct kinematic structures provide distinct performance characteristics. A new method is proposed to identify the distinct mechanisms derived from a given kinematic chain in this paper. Kinematic chains and their derived mechanisms are represented in the form of an extended adjacency matrix [EA] using the graph theoretic approach. Two structural invariants derived from the eigen spectrum of the [EA] matrix are the sum of absolute eigen values EA∑ and maximum absolute eigen value EAmax. These invariants are used as the composite identification number of a kinematic chain and mechanism and are tested to identify the all-distinct mechanisms derived from the family of 1-F kinematic chains up to 10 links. The identification of distinct kinematic chains and their mechanisms is necessary to select the best possible mechanism for the specified task at the conceptual stage of design.
