Recently it has been demonstrated that MTs participate in several cellular functions such as regulation of growth, and anti-oxidative defenses. Moreover, pesticides can induce their synthesis. The aim of the current work was to determine the effects of isoproturon either pure or formulated as Matin (suspension containing 500 g x l(-1) isoproturon) on the metallothionein and total protein content of the aquatic worm Tubifex tubifex. MT levels in exposed worms increased significantly after 7 and 15 days of exposure to 50 mg x l(-1) of herbicide. Isoproturon reduced metal (Cu, Zn, and Cd) content of metallothioneins, and it also increased total protein content of the worms. The results suggest that MT induction may not be considered as a specific biomarker of metal exposure but it can be used as a non specific biomarker of isoproturon effect in the worm.
Abstract Polycyclic aromatic hydrocarbons (PAHs) and heavy metals can influence smoked fish quali... more Abstract Polycyclic aromatic hydrocarbons (PAHs) and heavy metals can influence smoked fish quality and safety. Concentrations of 16 PAHs and heavy metals from smoked herring from three governorates in Egypt [Al-Behera (CF1), Cairo (CF2), and Alexandria (CF3)] were investigated. The mean concentration of individual PAHs ranged from 0.0007 μg/g of benzo[g,h,i]perylene (BgP) in Cairo to 0.3446 μg/g of benzo[a]anthracene (BaA) in Alexandria with the highest concentrations of the total PAHs found in Alexandria, followed by Al-Behera and then Cairo. Fluorene at CF3 had the highest mean concentration (0.2856 μg/g) which comprised 17.86% of total noncarcinogenic PAHs. Among the genotoxic PAHs (PAH8), BgP showed the lowest mean concentration (0.0007 μg/g) in Alexandria which accounted for 0.2% of ΣPAHs. Benz(a)anthracene had the highest mean values (0.3446 μg/g, 21.55%), followed by chrysene (0.2644 μg/g, 16.54%) at CF3. Mean concentrations of benzo(a)pyrene (BaP) were 0.0186, 0.0045, and 0.0254 μg/g for Al-Behera, Cairo, and Alexandria, respectively. The mean concentrations for BaP were higher than the maximum acceptable levels (5 μg/kg) established by European Union, which were classified as carcinogen. Among heavy metals, lead had the highest concentration (6.96 μg/g), which was higher than the maximum permissible levels set by World Health Organization, while nickel and arsenic were not detected.
Les travaux decrits dans ce memoire ont ete conduits dans le but d'apporter notre contributio... more Les travaux decrits dans ce memoire ont ete conduits dans le but d'apporter notre contribution a l'etude de la toxicite de divers contaminants (le cuivre ; l'isoproturon ; le fenhexamide et le chitosan sous forme de chito-gel), deja presents ou susceptibles de l'etre dans l'environnement champardenais. Le modele experimental utilise est un invertebre d'eau douce " Tubifex tubifex " (Annelide, Oligochete). Apres avoir analyse le comportement des produits dans l'eau et les vers, les concentrations letales ont ete determinees. Le Cu, l'isoproturon et le fenhexamide s'accumulent dans le ver. Le classement des produits selon leur toxicite est le suivant : Cu > fenhexamide > isoproturon > chito-gel, le cuivre etant le plus toxique. Les vers ont ensuite ete exposes pendant des durees variant de 2 a 14 jours a des concentrations subletales des contaminants ; en reponse a ces expositions la reponse du contenu en proteines totales depend du contaminant utilise. Les vers s'amputent de leur partie posterieure en presence des 4 contaminants. Dans la plupart des cas les systemes de defenses antioxydantes et le metabolisme de phase II (catalase, glutathion-S-transferase, glutathion-reductase et glutathion) sont stimules. De plus, le contenu en metallothioneines (MTs) augmente, non seulement en reponse au cuivre, mais aussi aux trois pesticides organiques. Les perturbations physiologiques induites par les contaminants constituent autant de biomarqueurs potentiels qui devront etre valides sur le terrain. L'induction des metallothioneides en reponse aux pesticides organiques est discutee en relation avec le stress oxydant.
