El momento y la forma en que se produjo el poblamiento inicial del continente americano forma par... more El momento y la forma en que se produjo el poblamiento inicial del continente americano forma parte de numerosas investigaciones científicas desde hace más de un siglo; aún hoy es un tema de estudio de interés principal para la arqueología, situándose en el centro de acalorados debates a nivel internacional (Graf et al. 2013). Temas como la cronología de la llegada de las sociedades humanas a Sudamérica, su ruta de entrada, el tempo del poblamiento, entre otros, han formado parte de este debate desde los comienzos de la disciplina (Ameghino 1918; Borrero 1989, 2016; Miotti 2006; Borrero & Miotti 2007; Boeda et al. 2013; Prates et al. 2013; Politis et al. 2016). El estudio de la variabilidad humana en América ha sido abordado aplicando técnicas de la antropo... more El estudio de la variabilidad humana en América ha sido abordado aplicando técnicas de la antropometría, la lingüística, la etnografía y la historia. Otra técnica para evaluar similitudes y diferencias entre grupos humanos proviene de la biología molecular. El ADN mitocondrial (ADNmt) debido a su herencia exclusivamente materna no sufre recombinación y por ello permite la reconstrucción de filogenias a nivel subespecífico. Los estudios de ADNmt en poblaciones humanas han apoyado la hipótesis del origen africano de los humanos modernos y han contribuido a reconstruir las rutas migratorias de poblamiento. Los datos referidos a América confirman el origen asiático de sus primeros pobladores y contribuyen a discutir el tiempo y modo de dispersión. El estudio de mutaciones diagnósticas de haplogrupos del ADN mitocondrial es una herramienta de u... more El estudio de mutaciones diagnósticas de haplogrupos del ADN mitocondrial es una herramienta de uso corriente en estudios antropológicos. Presentamos aquí un método de tipificación simplificada de haplogrupos que permite discriminar el origen continental de los linajes maternos presentes en poblaciones americanas utilizando sólo dos reacciones de PCR y electroforesis en gel de acrilamida. Los linajes maternos presentes en la actual población argentina derivan de tres fuentes principales: a aquellos derivados de las poblaciones indígenas (haplogrupos A-D) se sumaron los aportados por inmigrantes europeos y de Medio Oriente (asignables casi exclusivamente al super-haplogrupo N) y por las poblaciones de origen africano sub-sahareano (miembros del para-haplogrupo "L", Lx[M,N]). La determinación de los haplogrupos mitocondriales suele realizarse mediante secuenciación y/o la tipificación de una batería de mutaciones diganósticas. Con el objetivo de agilizar y abaratar la caracterización del origen continental de los linajes maternos presentes en poblaciones cosmopolitas argentinas adaptamos un protocolo desarrollado por Umetsu (2005) para la co-amplificación alelo-específica de posiciones variables diagnósticas de haplogrupos mitocondriales humanos. Cada posición variable es analizada con un trío de cebadores, dos de los cuales son alelo-específicos y difieren en su longitud, permitiendo obtener fragmentos distinguibles por 3-7 pb según esté presente la base ancestral o la derivada. Seleccionamos cinco tríos de cebadores, específicos para los haplogrupos A, M8 (al que pertenece C), D, M y N, que permiten obtener amplicones de tamaños no superpuestos. Las cinco posiciones son analizadas simultáneamente mediante amplificación en una única reacción de PCR y los fragmentos, de entre 52 y 149 pb, son resueltos en geles de poliacrilamida al 10%. La deleción de 9 pb es analizada mediante PCR-AFLP en forma independiente. All populations from Alaska to the MagellanÕs Strait present this lineage in frequencies higher than 60%. This lineage was considered the ÒfounderÓ, but further information shows that M3 occurred in America, or Siberia shortly before migration to America and so it is actually considered autochthonous. It belongs to the Q haplogroup, shows derivate state for M242, M346, and M3 polymorphisms, and is named subhaplogroup Q1a3a. Another lineage, paragroup Q1a3*, entered America showing derived states for M242 and M346 polymorphisms, and ancestral for M3. It is present in Eurasia and is more frequent in North than in South America. Though found at low frequency, it has not been possible to rule out a genetic bottleneck occurrence during the migration to South America.Fil: Ramallo, Virginia. Objective: This study aims to contribute to the recovery of Indigenous evolutionary history in th... more Objective: This study aims to contribute to the recovery of Indigenous evolutionary history in the Southern Pampas region of Argentina through an analysis of ancient complete mitochondrial genomes. Materials and Methods: We generated DNA data for nine complete mitogenomes from the Southern Pampas, dated to between 2531 and 723 cal BP. In combination with previously published ancient mitogenomes from the region and from throughout South America, we documented instances of extra-regional lineage-sharing, and estimated coalescent ages for local lineages using a Bayesian method with tip calibrations in a phylogenetic analysis. Results: We identified a novel mitochondrial haplogroup, B2b16, and two recently defined haplogroups, A2ay and B2ak1, as well as three local haplotypes within founder haplogroups C1b and C1d. El Nordeste argentino se caracteriza por la presencia de diversidad étnica, cultural y lingüístic... more El Nordeste argentino se caracteriza por la presencia de diversidad étnica, cultural y lingüística; y por una historia de movimientos poblacionales ligada a variaciones geopolíticas. El objetivo de este trabajo es presentar los primeros resultados de una investigación, estructurada en tres ejes (local, regional y sudamericano), que busca explorar la historia poblacional de la región a través de la caracterización de los linajes maternos y paternos de las ciudades de Corrientes (CRR, N=151) y Formosa (FOR, N=274). Se detallan las técnicas y métodos de análisis, así como las preguntas de investigación abordadas en cada eje, y se presentan los resultados del primer eje. Se estimaron los aportes de origen nativo americano, del oeste de Eurasia y de África subsahariana. Se obtuvieron frecuencias mayoritarias de linajes nativos americanos por vía materna (FOR=85%, CRR=89,4%). El actual territorio de Trujillo, una ciudad costera en el Norte de Perú, fue ocupado por diversa... more El actual territorio de Trujillo, una ciudad costera en el Norte de Perú, fue ocupado por diversas culturas precolombinas a lo largo de la historia. Con la llegada de los conquistadores europeos en el siglo XVI, se inició un proceso de mestizaje que continúa hasta la actualidad. Los objetivos de este trabajo son identificar y caracterizar linajes autóctonos y alóctonos del cromosoma Y en la población de Trujillo y evidenciar fenómenos del poblamiento y similitudes entre la población en estudio y poblaciones de Argentina. Se analizaron los linajes paternos a través de la región no recombinante del cromosoma Y por medio de un método de amplificación alelo-específica para la identificación marcadores bialélicos (SNPs) que determinan haplogrupos autóctonos y alóctonos para América en reacciones PCR-AFLP multiplex. Tal como se esperaba, esta población presentó una ancestralidad predominantemente nativa, con una alta influencia de linajes europeos.
En este trabajo se presenta la secuencia completa de ADN mitocondrial, obtenida a partir de resto... more En este trabajo se presenta la secuencia completa de ADN mitocondrial, obtenida a partir de restos óseos de un hombre adulto, hallado en el sitio Cañadón Misioneros (provincia de Santa Cruz, Argentina), con una antigüedad de 70 ± 30 años antes del presente. La secuencia corresponde al haplogrupo (hg) D4h3a5, nativo de América y exclusivo del sur de Patagonia, donde ha sido descripto tanto en muestras antiguas como actuales. Esta secuencia constituye el primer dato de ADN mitocondrial en la costa atlántica de Patagonia con la resolución suficiente para definir a nivel de subhaplogrupo. Tradicionalmente se agrupó a los linajes del ADN mitocondrial nativos de América dentro de los ci... more Tradicionalmente se agrupó a los linajes del ADN mitocondrial nativos de América dentro de los cinco haplogrupos A, B, C, D y X. En forma posterior, el avance en las técnicas moleculares permitió distinguir la existencia de, al menos, trece linajes que habrían ingresado a América previamente diferenciados. En la actualidad se está avanzando en la definición de sublinajes con distribución geográfica acotada. En este trabajo se analizan 743 secuencias de la Región Control (RC) completa, obtenidas en nueve poblaciones actuales de Argentina en el contexto de una recopilación de más de 6000 secuencias de linajes nativos de Sudamérica. Se identificaron grupos potencialmente monofiléticos (linajes) sobre la base de la presencia de mutación(es) compartida(s) en la RC, de los cuales se analiza su distribución geográfica. El análisis del genoma de individuos de origen arqueológico revela una continuidad genética durante 6.600 años y sugiere que los movimientos poblacionales no explican la apari-ción de la adaptación marina, adquirida por desarrollo local o por transmisión cultural. Sin embargo, dos even-tos de migración posteriores estarían correlacionados con cambios vinculados a tecnologías líticas. Durante el año 2005, se realizaron dos viajes de campaña a comunidades Wichí cercanas a las local... more Durante el año 2005, se realizaron dos viajes de campaña a comunidades Wichí cercanas a las localidades de Ingeniero Juárez y Laguna Yema (provincia de Formosa, Argentina), como parte del proyecto multidisciplinario “De las historias étnicas a la prehistoria en el Gran Chaco”. Partiendo del planteo metodológico de unidad poblacional, se obtuvieron datos genéticos en 93 muestras utilizando marcadores binarios y microsatélites del cromosoma Y, determinando haplogrupos y haplotipos masculinos. El haplogrupo Q1a3a, natural del continente americano, resultó mayoritario en ambas localidades (72,7 % y 81,6 %). Los linajes moleculares se compararon con la diversidad de apellidos registrada y las posibles vinculaciones entre las comunidades Wichís se analizaron por redes “median joining”, encontrando una variabilidad de linajes coherente con la distribución de las parcialidades del “complejo étnico Wichí” propuesto por Braunstein.

Estos nuevos desarrollos han permitido mejorar las comunicaciones y el acceso a la información. En estas condiciones, aparece una corriente global, el data sharing, que aboga por la libre puesta en disposición de los datos producto de las investigaciones científicas. Esta tendencia no está desarrollada en el área de Patagonia Austral. Sin embargo, antes de intercambiar datos, es importante conocer qué tipo de información se puede o pretende compartir. Con el fin de acercarnos a la situación en la que se encuentra la información disponible en la región, se realizó un análisis bibliográfico de las principales revistas donde se publican artículos del ámbito de la bioantropología en Patagonia Austral. El Nordeste argentino se caracteriza por la presencia de diversidad étnica, cultural y lingüístic... more El Nordeste argentino se caracteriza por la presencia de diversidad étnica, cultural y lingüística; y por una historia de movimientos poblacionales ligada a variaciones geopolíticas. El objetivo de este trabajo es presentar los primeros resultados de una investigación, estructurada en tres ejes (local, regional y sudamericano), que busca explorar la historia poblacional de la región a través de la caracterización de los linajes maternos y paternos de las ciudades de Corrientes (CRR, N=151) y Formosa (FOR, N=274). Se detallan las técnicas y métodos de análisis, así como las preguntas de investigación abordadas en cada eje, y se presentan los resultados del primer eje. Se estimaron los aportes de origen nativo americano, del oeste de Eurasia y de África subsahariana. Se obtuvieron frecuencias mayoritarias de linajes nativos americanos por vía materna (FOR=85%, CRR=89,4%). Con la llegada de los conquistadores europeos en el siglo XVI, se inició un proceso de mestizaje que continúa hasta la actualidad. Los objetivos de este trabajo son identificar y caracterizar linajes autóctonos y alóctonos del cromosoma Y en la población de Trujillo y evidenciar fenómenos del poblamiento y similitudes entre la población en estudio y poblaciones de Argentina. Se analizaron los linajes paternos a través de la región no recombinante del cromosoma Y por medio de un método de amplificación alelo-específica para la identificación marcadores bialélicos (SNPs) que determinan haplogrupos autóctonos y alóctonos para América en reacciones PCR-AFLP multiplex. Tal como se esperaba, esta población presentó una ancestralidad predominantemente nativa, con una alta influencia de linajes europeos.
En este trabajo se presenta la secuencia completa de ADN mitocondrial, obtenida a partir de resto... more En este trabajo se presenta la secuencia completa de ADN mitocondrial, obtenida a partir de restos óseos de un hombre adulto, hallado en el sitio Cañadón Misioneros (provincia de Santa Cruz, Argentina), con una antigüedad de 70 ± 30 años antes del presente. La secuencia corresponde al haplogrupo (hg) D4h3a5, nativo de América y exclusivo del sur de Patagonia, donde ha sido descripto tanto en muestras antiguas como actuales. Esta secuencia constituye el primer dato de ADN mitocondrial en la costa atlántica de Patagonia con la resolución suficiente para definir a nivel de subhaplogrupo. Tradicionalmente se agrupó a los linajes del ADN mitocondrial nativos de América dentro de los ci... more Tradicionalmente se agrupó a los linajes del ADN mitocondrial nativos de América dentro de los cinco haplogrupos A, B, C, D y X. En forma posterior, el avance en las técnicas moleculares permitió distinguir la existencia de, al menos, trece linajes que habrían ingresado a América previamente diferenciados. En la actualidad se está avanzando en la definición de sublinajes con distribución geográfica acotada. En este trabajo se analizan 743 secuencias de la Región Control (RC) completa, obtenidas en nueve poblaciones actuales de Argentina en el contexto de una recopilación de más de 6000 secuencias de linajes nativos de Sudamérica. El análisis del genoma de individuos de origen arqueológico revela una continuidad genética durante 6.600 años y sugiere que los movimientos poblacionales no explican la apari-ción de la adaptación marina, adquirida por desarrollo local o por transmisión cultural. Sin embargo, dos even-tos de migración posteriores estarían correlacionados con cambios vinculados a tecnologías líticas. Durante el año 2005, se realizaron dos viajes de campaña a comunidades Wichí cercanas a las local... more Durante el año 2005, se realizaron dos viajes de campaña a comunidades Wichí cercanas a las localidades de Ingeniero Juárez y Laguna Yema (provincia de Formosa, Argentina), como parte del proyecto multidisciplinario “De las historias étnicas a la prehistoria en el Gran Chaco”. Partiendo del planteo metodológico de unidad poblacional, se obtuvieron datos genéticos en 93 muestras utilizando marcadores binarios y microsatélites del cromosoma Y, determinando haplogrupos y haplotipos masculinos. El haplogrupo Q1a3a, natural del continente americano, resultó mayoritario en ambas localidades (72,7 % y 81,6 %). Los linajes moleculares se compararon con la diversidad de apellidos registrada y las posibles vinculaciones entre las comunidades Wichís se analizaron por redes “median joining”, encontrando una variabilidad de linajes coherente con la distribución de las parcialidades del “complejo étnico Wichí” propuesto por Braunstein.

Estos nuevos desarrollos han permitido mejorar las comunicaciones y el acceso a la información. En estas condiciones, aparece una corriente global, el data sharing, que aboga por la libre puesta en disposición de los datos producto de las investigaciones científicas. Esta tendencia no está desarrollada en el área de Patagonia Austral. Sin embargo, antes de intercambiar datos, es importante conocer qué tipo de información se puede o pretende compartir. Con el fin de acercarnos a la situación en la que se encuentra la información disponible en la región, se realizó un análisis bibliográfico de las principales revistas donde se publican artículos del ámbito de la bioantropología en Patagonia Austral.
