The evaluation factors were the sucrose concentration in the culture medium and the frequency immersion. There was a set of five pairs of RITA® vessels with liquid culture medium containing sucrose at 0,15, 30, 45, 60 g/L. A half-strength MS medium plus vitamins, supplemented with vitamins and thidiazuron (5 mg/L) was used. The experiment had two stages, each lasting two months. Each stage had a one minute immersion. The first stage had a four hour immersion frequency and the second one had an eight hour immersion frequency. Results showed that the best proliferating response was reached in a 15 g/L of sucrose medium with one minute of immersion time every four hours, resulting in 8,2 adventitious protocorms per protocorm per month. Factores tales como la cepa bacteriana, el tiempo de precultivo de los embriones y el tiempo de c... more Factores tales como la cepa bacteriana, el tiempo de precultivo de los embriones y el tiempo de cocultivo con la bacteria fueron evaluados en terminos de la expresion transiente del gen Gus en embriones somaticos de Heliconia stricta . Los resultados mostraron que la cepa bacteriana y el tiempo de cocultivo tienen efecto significativo en el porcentaje de embriones con expresion transiente, lo que no ocurre con el tiempo de precultivo. La cepa LBA4404, que porta el plasmido pBI121, con tres dias de cocultivo mostro el mejor resultado en la expresion transitoria de Gus. Sin embargo, el efecto de algunos antibioticos y tiempos de cocultivo prolongados sobre procesos posteriores a la transformacion deben ser evaluados con el fin de optimizar un protocolo que permita la transformacion estable de los embriones somaticos de esta especie.

Besides knowing the difficulties in propagation by seed (due to the low rate of germination and high seedling premature death) and by vegetative (slow growth), the present study evaluated the potential for propagation by using direct somatic embryogenesis. Young leaves segments from plants cultivated in a greenhouse were disinfected and planted in MS medium (Murashige y Skoog), supplemented with different concentrations of growth regulators. IBA and BAP combinations at 1 and 2 mg / L and 2 and 1 mg / L, respectively, shown to be effective in the formation of somatic embryos in this species. The results showed that the bacteria strain and coculture time had significant effects on the percentage of embryos with transient expression, but not the preculture time. The strain LBA4404 with the plasmid pBI121, with three days of coculture, showed the best transient expression of the gene Gus in this specie. However, it will be necessary to evaluate the effect of certain antibiotics and longer coculture times on the process after transformation, in order to optimize a protocol that allows for the stable transformation of the embryos.

The effect of different combinations of auxin (2.4-D and NAA) and cytokinins (KIN, BAP and TDZ) was evaluated on the formation of callus and shoot regeneration (direct or indirect), using MS (MB1) basal medium and B5 (MB2) basal modified medium. It grows on rocks in clear-water rivers and well-protected zones, similar to the waters in certain watersheds of the Antioquia Department, Colombia. Loss of habitat has threatened this promising ornamental plant species, which is also important because of its role in the ecosystem. In vitro tissue culture is considered an efficient alternative for the propagation of endangered species with the aim of establishing short-, medium- and long-term conservation programs. In the present research, in vitro introduction and shoot induction from A. antioquiense seedlings were performed. The highest production of shoots was obtained in a ½ MS (half-salt content) medium with 1 mg L-1 of BAP, which attained a 23.7 shoots/explant per month multiplication rate.
