No quadro desta análise e a partir da distinção entre mediação tecnológica e mediação pedagógica, propõe-se um modelo para a mudança e inovação baseado na mediação social e cognitiva das aprendizagens para a educação na sociedade digital. A mediação para a educação aberta situa-se para além das conceções de inovação associadas à tecnologia, para se afirmar nos desafios da aprendizagem como um processo social e cognitivo sustentado pela rede. No âmbito deste enquadramento e da valorização da mudança intencional na educação em rede, a cenarização da inovação pedagógica emerge do conjunto das dimensões relacionais da rede de atores que ocorrem nos ambientes de aprendizagem e conhecimento. A partir da articulação entre os cenários de inovação no modelo pedagógico e a adequação deste ao projeto, objetivos e práticas de aprendizagem é proposto um conjunto de dimensões para a análise da usabilidade pedagógica dos ambientes de educação aberta e em rede na sociedade digital. Abstract We develop in this article a reflection on the processes that lead to the promotion of pedagogic innovation in emerging scenarios for open and online education. From this analysis and the distinction between technological and pedagogical mediation, we propose a model for change and innovation based on social and cognitive mediation of learning and education in the digital society. Mediation for open education lies beyond the conceptions of innovation related to technology, to assert the challenges of learning as a social and cognitive process supported by the network. Within this framework and the appreciation of the intentional change in education networking, the scenarios of pedagogical innovation emerges from all the relational dimensions of the actors that occur in the learning and knowledge network environments. From the articulation between the scenarios of innovation in pedagogical model and the suitability of this to the project, objectives and learning practices is proposed a set of dimensions to the analysis of the pedagogical usability of open and online education to the digital society. Multidrug resistance is often the result of the activation of drug efflux pumps able to catalyze ... more Multidrug resistance is often the result of the activation of drug efflux pumps able to catalyze the extrusion of the toxic compound to the outer medium, this activation being frequently controlled at the transcriptional level. Transcriptional regulation in the model eukaryote S. cerevisiae is the result of the interaction and cross-talk between networks of transcription factors. This is the case of the transcriptional activation of the FLR1 gene occurring in response to stress induced by the agricultural fungicide mancozeb in yeast. FLR1 up-regulation depends on the integrated action of Yap1, a key regulator of oxidative stress response, Pdr3 and Yrr1, two of the transcription factors controlling multidrug resistance, and Rpn4, a regulator of proteasome gene expression, which interplay to produce the observed transcriptional up-shift. Based on the expression profiles of FLR1, YAP1, PDR3, YRR1 and RPN4 registered during yeast adaptation to stress induced by mancozeb and using a qualitative modeling approach, a model of the FLR1 regulatory network was built, and the response of S. cerevisiae to mancozeb stress was simulated. The use of a qualitative approach is especially useful to overcome the lack of enough quantitative data on kinetic parameters and molecular concentrations, permitting the immediate focus on the qualitative behavior of the system. This Systems Biology approach allowed the identification of essential features of the early yeast response to fungicide stress. The resulting model allowed the formulation of new hypotheses, in a quick and cost effective manner, on the qualitative behavior of the system following mancozeb challenge, some of which were validated experimentally. In particular, Pdr3 and Yrr1 were shown to directly control FLR1 up-regulation in mancozeb-challenged cells, based on the analysis of the effect of the inactivation of their putative binding sites in the FLR1 promoter. Furthermore, the inter-dependent role of Yap1 and Yrr1 in the regulation of PDR3 and RPN4 was brought to light, this joint activity possibly being extensible to eight other genes involved in multidrug resistance. The FLR1 network structure was revised, based on the comparison between simulated and experimental gene expression data in the double deletion mutant strains Δyrr1Δpdr3 and Δyrr1Δrpn4, and an additional, still unidentified, transcription factor was found to be required to fully explain the behavior of the network.

OBJECTIVE: To assess endovascular treatment of thoracic aorta diseases in a national centre of an... more OBJECTIVE: To assess endovascular treatment of thoracic aorta diseases in a national centre of angiology and vascular surgery. To quantify the national registry of TEVAR's.MATERIAL AND METHODS: This unicentric and retrospective study included patients submitted to TEVAR until the end of 2012. Twenty-seven patients were considered high-risk for conventional surgery: 14 degenerative thoracic aorta aneurysms or pseudoaneurysms (10 assymptomatic), 1 ruptured thoracoabdominal aneurysm, 5 aortabronchial/aortoesophageal fistulas, 3 complicated dissections, 2 penetrating atherosclerotic ulcer/intramural hematoma, 1 traumatic laceration and 1 embolization from aortic plaque. Eighteen (67%) were emergent/urgent procedures.RESULTS: At the institutional level, immediate technical success was achieved in all cases; average follow up was 24 months (0-97). Thirty days and 24 months global mortality was, respectively, 4% (6% for emergent/urgent procedures and 0% for elective procedures) and 13%. Aortic-related mortality was similar. One case of paraplegia and 2 of case of stoke were registered. Endoleak was present in 4 patients. Survival free from aneurysmal sac expansion (aneurysm, pseudoaneurysm or dissection, n=16) was 88% at 30 days. Survival free from aortic reintervention was 93% at 30 days and 81% at 24 months. Nationally, TEVAR registries triplicated from 2007 top 2010.CONCLUSION: These results favour the actual tendency to consider TEVAR as a first-line solution for several thoracic aortic diseases.
