Eriobotrya japonica (loquat) is an evergreen fruit tree of the apple tribe in Rosaceae with high edible and medicinal value. The yield and quality of loquat fruits are significantly influenced by environmental stress, particularly heat stress during fruit ripening. In this study, thirty EjHSFs were identified in the loquat genome. Twenty-nine EjHSFs were unevenly distributed across sixteen chromosomes, except Chr-6. A synteny analysis revealed that twenty-six EjHSF genes had undergone duplication events. Twenty-nine EjHSF genes were found to be in sync with HSF in apples while also diverging with other Rosaceae species. A phylogenetic analysis revealed that EjHSFs could be divided into three categories, including eighteen EjHSF-A, ten EjHSF-B, and two EjHSF-C. Twenty-nine members of the EjHSF family showed high homology to those of Malus domestica and Gillenia trifoliate. A promoter analysis retrieved thirty-three CAREs that were functionally relevant and connected to the expression of HSFs. Among these, the promoters of twenty-three EjHSF genes possessed at least one STRE element that could be activated by heat shock. Eleven of these EjHSFs were highly expressed in loquat tissues, namely EjHSF-B4a, EjHSF-A4a, EjHSF-A4d, and EjHSF-C1a in roots; EjHSF-B4b in roots and inflorescence; EjHSF-C1b in stems and roots; EjHSF-A2a in three tissues; EjHSF-A8b in four tissues; and EjHSF-A4c, EjHSF-B1a, and EjHSF-B2b in six tissues. Moreover, fifteen EjHSFs were differentially expressed (eleven upregulated and four downregulated) in fruits under heat stress treatment in the color-changing period. Among them, EjHSF-A2a and EjHSF-A2b upregulated transcriptional abundance over 300 times after heat treatment; EjHSF-B2b consistently displayed an increasing trend over time; and EjHSF-B1a was significantly downregulated. Hence, these results suggest that EjHSFs take part in loquat tissue development and may be involved in the fruit’s heat stress response. These findings enhance the understanding of EjHSFs’ role within loquats and the heat stress response of its fruit and provide target genes for heat stress improvement. Full article

It is well known that the widespread utilization of fossil fuels contributes to climate change, so exploring new sustainable energy sources is more important than ever for energy transition pathways. The variability and intermittency of solar and wind sources are of concern. Hydrogen (H2) utilization as an energy carrier can address this issue. The technology for producing hydrogen from biomass gasification has not yet reached a high level of technological maturity. The main novelty of this work is to evaluate the state of the art of the technologies for producing H2 from solid biomass, taking into account technological, economic, and environmental indicators and the results of a bibliometric study, and also the calculation of the technical potential for hydrogen production through biomass gasification on a worldwide and Brazilian scale. The most frequently mentioned technology to boost H2 production efficiency is the addition of catalysts to the gasifier. Primary catalyst utilized in biomass gasification for hydrogen enhancing enhancement, such as olivine, CaO, and CeO2-Ni-CaO are reviewed in the article. As a result, the syngas had an H2 content rise of 511%, 659.6%, and 853.4%, respectively. According to the reviewed literature, the levelized cost of hydrogen production can reach an average value of USD3.15/kg of H2, and the average yield is 0.1 kg-H2/kg-biomass. The worldwide potential for hydrogen production from solid biomass in an optimal trends scenario for 2050 is estimated to be 45.03 EJ, and Brazil’s potential is 6.5 EJ. Full article

Background: the reference method for detection of myositis-specific and myositis-associated antibodies (MSAs and MAAs) is considered immunoprecipitation (IP), but it is routinely replaced by semi-automated methods, like lineblot (LB). Few data are available on the consistency with clinical diagnoses; thus, we aim at analysing these aspects. Methods: sixty-nine patients with idiopathic inflammatory myopathies (IIM) were studied via LB (Myositis Antigens Profile 3 EUROLINE, Euroimmun) and IP (RNA and protein antigens). The degree of concordance between methods was calculated using Cohen’s coefficient. Results: a substantial concordance was found for anti-Ku and anti-PM/Scl and a moderate concordance was found for anti-Jo1 and anti–Mi-2, while a fair concordance was found for anti-EJ, anti-SRP, and anti-Ro52 antibodies. The concordance could not be calculated for anti-OJ, anti-PL-7, anti-PL-12, anti-NXP2, anti-TIF1ɣ, and anti-MDA5, because they were only detected with one method. Multiple MSAs were found only with LB in 2/69 sera. Anti-MDA5, TIF1ɣ, NXP2 (detected via IP), and anti-Jo1 in anti-synthetase syndrome (both LB and IP) had the best concordance with clinical diagnosis. Conclusions: LB and IP show substantial concordance for PM/Scl and Ku, and moderate concordance for Jo1 and Mi-2, with a good concordance with clinical diagnoses. IP shows a high performance for DM-associated MSAs. LB seems to be more sensitive in detecting anti-Ro52 antibodies, but it identified multiple MSAs, unlike IP. Full article

Although urban greenspace enhances ecological functioning and human well-being through ecosystem services (ES), it is oftentimes inequitably distributed. Environmental justice (EJ) encompasses aspects of distributive, procedural, and interactive justice related to accessibility and allocation of environmental benefits. Vacant land in shrinking cities has the potential to address greenspace inequalities and provide ES. This study investigated the perceptions of residents regarding urban ES and EJ in their communities in St. Louis (MO, USA)—a shrinking city that was undergoing green development, through semi-structured interviews. Altogether, 27 residents were selected from socio-economically disadvantaged neighborhoods characterized by high levels of vacancy due to legacies of redlining and systemic racism. Interview analysis revealed four themes: green benefits (including recreation opportunities), green costs (e.g., concerns for increased crime and nuisance animals), injustice issues (e.g., access to community greenspaces), and changes in the community (e.g., higher property taxes). Results revealed that residents perceived ES as closely connected with EJ when it comes to urban greening projects in their city. This study helps inform the process of urban greening projects, particularly in shrinking cities at risk of inequities. Full article

The yellow-fleshed loquat is abundant in carotenoids, which determine the fruit’s color, provide vitamin A, and offer anti-inflammatory and anti-cancer health benefits. In this research, the impact of abscisic acid (ABA), a plant hormone, on carotenoid metabolism and flesh pigmentation in ripening loquat fruits was determined. Results revealed that ABA treatment enhanced the overall content of carotenoids in loquat fruit, including major components like β-cryptoxanthin, lutein, and β-carotene, linked to the upregulation of most genes in the carotenoid biosynthesis pathway. Furthermore, a transcription factor, EjWRKY6, whose expression was induced by ABA, was identified and was thought to play a role in ABA-induced carotenoid acceleration. Transient overexpression of EjWRKY6 in Nicotiana benthamiana and stable genetic transformation in Nicotiana tabacum with EjWRKY6 indicated that both carotenoid production and genes related to carotenoid biosynthesis could be upregulated in transgenic plants. A dual-luciferase assay proposed a probable transcriptional control between EjWRKY6 and promoters of genes associated with carotenoid production. To sum up, pre-harvest ABA application could lead to carotenoid biosynthesis in loquat fruit through the EjWRKY6-induced carotenoid biosynthesis pathway. Full article

The impact of global warming is increasing and thus exacerbating environmental stresses that affect plant yield and distribution, including the Eriobotrya japonica Lindl (Loquat tree). Eriobotrya japonica, a member of the Rosaceae family, is valued not only for its nutritious fruit but also for its medicinal purposes, landscape uses, and other pharmacological benefits. Nonetheless, the productivity of Eriobotrya japonica has raised a lot of concern in the wake of adverse environmental conditions. Understanding the characteristics of the LRR-RLK gene family in loquat is crucial, as these genes play vital roles in plant stress responses. In this study, 283 LRR-RLK genes were identified in the genome of E. japonica that were randomly positioned on 17 chromosomes and 24 contigs. The 283 EjLRR-RLK proteins clustered into 21 classes and subclasses in the phylogenetic analysis based on domain and protein arrangements. Further explorations in the promoter regions of the EjLRR-RLK genes showed an abundance of cis-regulatory elements that functioned in growth and development, phytohormone, and biotic and abiotic responses. Most cis-elements were present in the biotic and abiotic responses suggesting that the EjLRR-RLK genes are invested in regulating both biotic and abiotic stresses. Additional investigations into the responses of EjLRR-RLK genes to abiotic stress using the RT-qPCR revealed that EjLRR-RLK genes respond to abiotic stress, especially heat and salt stresses. Particularly, EjapXI-1.6 and EjapI-2.5 exhibited constant upregulation in all stresses analyzed, indicating that these may take an active role in regulating abiotic stresses. Our findings suggest the pivotal functions of EjLRR-RLK genes although additional research is still required. This research aims to provide useful information relating to the characterization of EjLRR-RLK genes and their responses to environmental stresses, establishing a concrete base for the following research. Over the last two decades, substantial studies have been conducted to assess the feasibility of a multi-pollutant strategy for managing air quality in the United States. Given the inherent complexity of multi-pollutant air quality challenges, including fine particulate matter (PM2.5), ozone (O3), and air toxics, this paper undertook a multi-pollutant analysis at both national and local levels. Our analysis incorporated O3 and PM2.5 concentrations, air toxics that increase the risk of cancer, environmental justice (EJ) data, emissions data, and monitoring data. Initially, we identified counties across the continental U.S. with heightened multi-pollutant exposures and EJ concerns. Subsequently, a case study within the Detroit metropolitan area was conducted, revealing a clear overlap between multi-pollutant and EJ issues, underscoring the disproportionate burden on disadvantaged communities. The analysis of detailed emissions data unveiled potential co-control benefits in this region. Lastly, employing a proximity analysis method, we assessed environmental issues surrounding points of interest such as monitoring sites and emissions sectors, in the Detroit metropolitan area. The results demonstrated that monitoring sites with the highest monitoring value, alongside top-ranked emissions sectors such as electric utilities, coke ovens, and iron and steel production, were likely to exhibit elevated air pollutant concentrations/risks and associated EJ concerns in their vicinity. Loquat is an evergreen fruit crop which blooms from autumn–winter, and supports human beings with juicy fruit from late spring to early summer. However, the most traditional cultivars of this crop produce small fruit and bear a much lower yield than its relatives like apple, pear and peach. Large-size cultivars have long been a cherished aim of breeders for improving the production yield of loquat. Agronomic practices like panicle thinning, fruit thinning, growth regulator application, fertilization and so on are easier and more accessible ways for growers to produce large-size loquat fruit on existing production trees. Here, we develop a novel pruning method with an annual double back-cut, which provides vigorous shoot with more leaves and thicker branches for bearing much larger loquat fruit. Cellular observation determined that the vigorous shoot training method motivated cell division to produce larger loquat fruit, and that most of these cell layers were proliferated before the appearance of flower blossoms. Gene expression data of four development stages showed that EjFWL1 and EjFWL2 were notably downregulated in flower buds of the vigorously pruned tree. The data here further confirmed that the cell division capacity during flower development greatly influenced both the flower and fruit size of loquat. More importantly, we developed a novel pruning method to inhibit cell division repressors, promote cell proliferation and enlarge fruit size in loquat. Full article

Introduction and objectives: To evaluate the oncological and functional outcomes of transperineal laser ablation (TPLA) as the focal therapy for localized prostate cancer (PCa) after a 12-month follow-up. Materials and methods: Patients with low- and intermediate-risk localized PCa were prospectively treated with focal TPLA between July 2021 and December 2022. The inclusion criteria were the following: clinical stage < T2b; PSA < 20 ng/mL; International Society of Urological Pathology (ISUP) grade ≤ 2; MRI-fusion biopsy-confirmed lesion classified as PI-RADS v2.1 ≥ 3. Intra-, peri-, and post-operative data were collected. Variables including age, PSA, prostate volume (PVol), Charlson’s Comorbidity Index (CCI), International Prostate Symptom Score (IPSS) with QoL score, International Index of Erectile Function (IIEF-5), International Consultation on Incontinence Questionnaire—Short Form (ICIQ-SF), and Male Sexual Health Questionnaire—Ejaculatory Dysfunction Short Form (MSHQ-EjD) were collected at baseline and at 3, 6 and 12 months after TPLA. Post-operative mpMRI was performed at 3 and 12 months. Finally, all patients underwent prostatic re-biopsy under fusion guidance at 12 months. The success of this technique was defined as no recurrence in the target treated lesion at the 12-month follow up. Results: Twenty-four patients underwent focal TPLA. Baseline features were age [median 67 years (IQR 12)], PSA [5.7 ng/mL (3.9)], PVol [49 mL (27)], CCI [0 (0)], IPSS [11 (9)], IPSS-QoL [2 (2)], IIEF-5 [21 (6)], ICIQ-SF [0 (7)], MSHQ-EjD ejaculation domain [14 (4)] and bother score [0 (2)]. Median operative time was 34 min (IQR 12). Median visual analogue scale (VAS) 6 h after TPLA was 0 (IQR 1). The post-operative course was regular for all patients, who were discharged on the second post-operative day and underwent catheter removal on the seventh post-operative day. No patient had incontinence at catheter removal. A significant reduction in PSA (p = 0.01) and an improvement in IPSS (p = 0.009), IPSS-QoL (p = 0.02) and ICIQ-SF scores (p = 0.04) compared to baseline were observed at the 3-month follow-up. Erectile and ejaculatory functions did not show any significant variation during the follow-up. No intra- and peri-operative complications were recorded. Three Clavien–Dindo post-operative complications were recorded (12%): grade 1 (two cases of urinary retention) and grade 2 (one case of urinary tract infection). At the 12-month follow-up, eight patients showed mpMRI images referable to suspicious recurrent disease (PIRADS v2.1 ≥ 3). After re-biopsy, 7/24 patients’ (29%) results were histologically confirmed as PCa, 3 of which were recurrences in the treated lesion (12.5%). The success rate was 87.5%. Conclusions: The focal TPLA oncological and functional results seemed to be encouraging. TPLA is a safe, painless, and effective technique with a good preservation of continence and sexual outcomes. Recurrence rate at 12 months was about 12.5%. Full article

Long-term cross-correlational structures are examined for pairs of sea surface temperature anomalies (SSTAs) and advective forcing parameters and sea surface height anomalies (SSHAs) and current velocity anomalies (CVAs) in the East/Japan Sea (EJS); all these satellite datasets were collected between 1993 and 2023. By utilizing newly modified detrended cross-correlation analysis algorithms, incorporating local linear trend and local fluctuation level of an SSTA, the analyses were performed on timescales of 400–3000 days. Long-term cross-correlations between SSTAs and SSHAs are strongly persistent over nearly the entire EJS; the strength of persistence is stronger during rising trends and low fluctuations of SSTAs, while anti-persistent behavior appears during high fluctuations of SSTAs. SSTA-CVA pairs show high long-term persistence only along main current pathways: the zonal currents for the Subpolar Front and the meridional currents for the east coast of Korea. SSTA-CVA pairs also show negative long-term persistent behaviors in some spots located near the coasts of Korea and Japan: the zonal currents for the eastern coast of Korea and the meridional currents for the western coast of Japan; these behaviors seem to be related to the coastal upwelling phenomena. Further, these persistent characteristics are more conspicuous in the recent decades (2008~2023) rather than in the past (1993~2008). Full article

This paper quantifies the annual dynamics of the shortwave radiation reflected from bare arable land as a result of smoothing previously plowed land located in three different agricultural subregions of the European Union and associated countries. This estimate takes into account the annual variation of the bare arable land area, obtained from Sentinel 2 satellite imagery; the spatial variability of soil units within croplands, obtained from digital soil and land-cover maps; and the laboratory spectral reflectance characteristics of these units, obtained from soil samples stored in the LUCAS soil database. The properties of the soil units, which cover an area of at least 4% of each subregion, were characterized. The highest amounts of shortwave radiation reflected under clear-sky conditions from air-dried, bare arable land surfaces—approximately 850 PJ day−1 and 1.10 EJ day−1 for land shaped by a plow (Pd) and smoothing harrow (Hs), respectively—were found in the summer around 8 August in the western subregion. However, the lowest radiation occurred in the spring on 10 April at 340 PJ day−1 for Pd and 430 PJ day−1 for Hs in the central subregion. The largest and the smallest amounts of this radiation throughout the year—only as a result of smoothing, by Hs, land that was previously treated by Pd—was estimated at 42 EJ for the western and southern subregions and 19 EJ for the central subregion, respectively. Among non-destructive testing (NDT) techniques, fast neutron radiography with a higher penetration capability has achieved rapid advancements. However, the application of the radiography detector in many fast neutron imaging systems is limited by unfavorable detection efficiency and imaging spatial resolution. In this paper, a fast neutron radiography detector was designed, which was composed of a pixelated EJ200 scintillator array, a 16 × 16 silicon photomultiplier (SiPM) array, and capacitive multiplexing network readout electronics. The main parameters of the detector were optimized using Monte Carlo simulations. In addition, the prototype of the detector was fabricated and tested under a 14 MeV D-T neutron source. The preliminary test results demonstrated that the spatial resolution of the prototype reached 1.2 mm. Moreover, the conflict between spatial resolution and detection efficiency could be mitigated by using a pixelated scintillator structure. Overall, SiPMs enabled the extensive application of the imaging system because of their excellent photon detection performance, relatively low price, and joint possibility for large areas. The therapeutic targeting of DNA repair pathways is an emerging concept in cancer treatment. Compounds that target specific DNA repair processes, such as those mending DNA double-strand breaks (DSBs), are therefore of therapeutic interest. UNC3866 is a small molecule that targets CBX4, a chromobox protein, and a SUMO E3 ligase. As a key modulator of DNA end resection—a prerequisite for DSB repair by homologous recombination (HR)—CBX4 promotes the functions of the DNA resection factor CtIP. Here, we show that treatment with UNC3866 markedly sensitises HR-deficient, NHEJ-hyperactive cancer cells to ionising radiation (IR), while it is non-toxic in selected HR-proficient cells. Consistent with UNC3866 targeting CtIP functions, it inhibits end-resection-dependent DNA repair including HR, alternative end joining (alt-EJ), and single-strand annealing (SSA). These findings raise the possibility that the UNC3866-mediated inhibition of end resection processes we define highlights a distinct vulnerability for the selective killing of HR-ineffective cancers. The recombinant structural protein described in this study was designed based on sequences derived from elastin and silk. Silk–elastin hybrid copolymers are characterized by high solubility while maintaining high product flexibility. The phase transition temperature from aqueous solution to hydrogel, as well as other physicochemical and mechanical properties of such particles, can differ significantly depending on the number of sequence repeats. We present a preliminary characterization of the EJ17zipR protein obtained in high yield in a prokaryotic expression system and efficiently purified via a multistep process. Its addition significantly improves biomaterial’s rheological and mechanical properties, especially elasticity. As a result, EJ17zipR appears to be a promising component for bioinks designed to print spatially complex structures that positively influence both shape retention and the internal transport of body fluids. The results of biological studies indicate that the addition of the studied protein creates a favorable microenvironment for cell adhesion, growth, and migration. Full article

Oxidative stress from an excess of radical compounds generally impacts apoptosis and inflammation. The use of probiotics, therefore, has emerged as a favorable tool to suppress the formation of reactive oxygen species. In the present study, we investigated the antioxidant activity of plant-derived Lactiplantibacillus (L.) plantarum MSC-5T fermented in Eriobotrya japonica (EJ) aqueous extract. In the in vitro study, the extract fermented with the MSC-5T strain markedly decreased the cell death of H2O2-induced HEK293 cells. In addition, the fermented extract showed a protective effect against fungal toxin ochratoxin A (OTA) and citrinin (CTN). Regarding the evaluation of glutathione homeostasis, it can be clearly seen that pretreatment of HEK293 cells with fermented EJ extract greatly increased glutathione (GSH) levels, while unfermented extract did not affect the cellular GSH content. Furthermore, we identified a bioactive compound as pyrocatechol, which displayed significant antioxidation activity. The extract fermented for 48 h with the MSC-5T strain in EJ extract produces 167.4 μg/mL pyrocatechol. Full article
