Salivary Oxidative -Antioxidant Profile Following Adjunctive Gaseous Ozone Administration in Non-Surgical Periodontal Treatment: A Randomized Controlled Trial

Background: Periodontitis is associated with increased oxidative stress, which may impair treatment outcomes. Ozone therapy has shown promise in reducing oxidative stress and improving periodontal health. This study examined the impact of adjunctive gaseous ozone administration on salivary oxidative stress markers in patients with periodontitis stages II–IV and grades A–C undergoing non-surgical periodontal treatment (NSPT). Methods: Ninety patients with periodontitis were randomly allocated to either the test group (NSPT with gaseous ozone administration) or the control group (NSPT alone) using computer-generated randomization. The OzoneDTA system was used to deliver ozone at 2100 ppm for 60 s per site once weekly for 4 weeks. Clinical periodontal parameters (probing depth [PD], clinical attachment level [CAL], plaque index [PI], gingival index [GI]) and salivary oxidative stress markers (malondialdehyde [MDA], total antioxidant capacity [TAC], superoxide dismutase [SOD]) were assessed by blinded examiners at baseline, 3, and 6 months post-treatment. Results: Mixed ANOVA revealed significant three-way interactions between time, treatment, and stage or grade for clinical and biochemical measures (p < 0.001). The test group exhibited significant improvements in TAC (mean difference: 0.45 ± 0.12 mmol/L, p = 0.002), MDA (−0.38 ± 0.09 nmol/mL, p = 0.001), and SOD (65 ± 18 U/mL, p < 0.001) compared with the control group, with more pronounced effects in stages III and IV. Large effect sizes (Cohen’s d > 0.8) were observed for the test group between baseline and 6 months for all markers. Conclusions: Gaseous ozone administration as an adjunct to NSPT can effectively improve clinical periodontal parameters and salivary oxidative stress markers, particularly in stages III and IV periodontitis. The enhanced outcomes may be attributed to ozone’s antimicrobial and immunomodulatory properties, which synergistically reduce oxidative stress and promote periodontal healing. Full article

This study aimed to assess the effects of the timing of administering botulinum neurotoxin A (BoNT/A) on nerve regeneration in rats. Sixty 6-week-old rats with a sciatic nerve injury were randomly divided into four groups: the immediately treated (IT) group (BoNT/A injection administered immediately post-injury), the delay-treated (DT) group (BoNT/A injection administered one week post-injury), the control group (saline administered one week post-injury), and the sham group (only skin and muscle incisions made). Nerve regeneration was assessed 3, 6, and 9 weeks post-injury using various techniques. The levels of glial fibrillary acid protein (GFAP), astroglial calcium-binding protein S100β (S100β), growth-associated protein 43 (GAP43), neurofilament 200 (NF200), and brain-derived neurotrophic factor (BDNF) in the IT and DT groups were higher. ELISA revealed the highest levels of these proteins in the IT group, followed by the DT and control groups. Toluidine blue staining revealed that the average area and myelin thickness were higher in the IT group. Electrophysiological studies revealed that the CMAP in the IT group was significantly higher than that in the control group, with the DT group exhibiting significant differences starting from week 8. The findings of the sciatic functional index analysis mirrored these results. Thus, administering BoNT/A injections immediately after a nerve injury is most effective for neural recovery. However, injections administered one week post-injury also significantly enhanced recovery. BoNT/A should be administered promptly after nerve damage; however, its administration during the non-acute phase is also beneficial. Full article

(1) Background: Heat stress detrimentally restricted economic growth in dairy production. In particular, the cooling mechanism of the spraying system effectively reduced both environmental and shell temperatures. This study was designed to investigate the underlying modulatory mechanism of an automatic cooling system in alleviating heat-stressed dairy cows. (2) Methods: A total of 1208 multiparous dairy cows was randomly allocated into six barns, three of which were equipped with automatic sprinklers (SPs), while the other three were considered the controls (CONs). Each barn was considered a replicate. (3) Results: Body temperatures and milk somatic cell counts significantly decreased, while DMI, milk yield, and milk fat content significantly increased under SP treatment. Rumen fermentability was enhanced, embodied by the increased levels of total VFA, acetate, propionate, and butyrate after SP treatment. The rumen microbiota results showed the relative abundances of fiber-degrading bacteria, including the Fibrobacters, Saccharofermentans, Lachnospira, Pseudobutyrivibrio, Selenomonas, and Succinivibrio, which significantly increased after receiving the SP treatment. (4) Conclusions: This study demonstrated that SP effectively alleviated heat stress and improved production performances and milk quality through modulating the rumen microbiota composition and fermentation function of dairy cows. Exploring the Relationship between Ecosystem Services and Sustainable Development Goals for Ecological Conservation: A Case Study in the Hehuang Valley of Qinghai-Tibet Plateau

With the increase in human activities and the acceleration of urbanization, over-exploitation of natural resources has led to a decline in ecosystem services (ESs), subsequently affecting the achievement of sustainable development goals (SDGs). As the key ecological zone of Qinghai-Tibet Plateau, the stability and enhancement of ESs in the Hehuang Valley are crucial for achieving SDGs and biodiversity conservation. This study quantifies nine SDGs for the Hehuang Valley in the last twenty years. Four ecological models were utilized to compute key ESs: net primary productivity (NPP), water yield, soil retention, and sand fixation. Panel data were analyzed using a coupling coordination model to quantify the relationship between ESs and sustainable development level (SDL) in each county. Additionally, the Geographically and Temporally Weighted Regression (GTWR) model was employed to examine the correlation between ESs and SDL. The results indicate the following: (1) During the period, NPP and water yield first increased and then decreased. The capacity for soil retention and sand fixation showed an overall increase, highlighting substantial variability among counties in their ability to deliver these ESs. (2) The SDL of counties in the Hehuang Valley increased, with Xining City showing slightly higher SDL than other counties. (3) The overall coupling coordination degree among NPP, water yield, soil retention, sand fixation, and SDL in the Hehuang Valley exhibited an upward trend in the last twenty years. SDL demonstrated the highest coordination degree with NPP, followed by soil retention, water yield, and sand fixation. (4) Most counties in the Hehuang Valley exhibited a lag in SDL relative to NPP, water yield, and soil retention in the last twenty years. In the early stage, sand fixation and SDL were primarily lagging in SDL, while in the late stages, sand fixation lagged behind SDL. (5) During the period, there was an increasing negative correlation observed between the four ESs and SDL. The positive contribution of NPP and sand fixation in some counties gradually shifted to a negative effect, and the negative effect of water yield and soil retention on SDL intensified. The impact of human activities on ecosystem function hindered local SDL. This study offers scientific theoretical backing and practical recommendations for promoting SDL and biodiversity conservation in the Hehuang Valley. Full article

A novel tool utilizing machine learning techniques was designed to forecast ap index values for the next three consecutive days (24 values). The tool employs time series data from the 3 h ap index of solar cycles 23 and 24 to train the Long Short-Term Memory (LSTM) model, predicting ap index values for the next 72 h at three-hour intervals. During periods of quiet geomagnetic activity, the LSTM model’s performance is sufficient to yield favorable outcomes. Nevertheless, during geomagnetically disturbed conditions, such as geomagnetic storms of different levels, the model needs to be adapted in order to provide accurate ap index results. In particular, when coronal mass ejections occur, the ap Prediction tool is modulated by inserting predominant features of coronal mass ejections such as the date of the event, the estimated time of arrival and the linear speed. Sphaerococcenol A is a cytotoxic bromoditerpene biosynthesized by the red alga Sphaerococcus coronopifolius. A series of its analogues (16) was designed and semi-synthesized using thiol-Michael additions and enone reduction, and the structures of these analogues were characterized by spectroscopic methods. Cytotoxic analyses (1–100 µM; 24 h) were accomplished on A549, DU-145, and MCF-7 cells. The six novel sphaerococcenol A analogues displayed an IC50 range between 14.31 and 70.11 µM on A549, DU-145, and MCF-7 malignant cells. Compound 1, resulting from the chemical addition of 4-methoxybenzenethiol, exhibited the smallest IC50 values on the A549 (18.70 µM) and DU-145 (15.82 µM) cell lines, and compound 3, resulting from the chemical addition of propanethiol, exhibited the smallest IC50 value (14.31 µM) on MCF-7 cells. The highest IC50 values were exhibited by compound 4, suggesting that the chemical addition of benzylthiol led to a loss of cytotoxic activity. The remaining chemical modifications were not able to potentiate the cytotoxicity of the original compounds. Regarding A549 cell viability, analogue 1 exhibited a marked effect on mitochondrial function, which was accompanied by an increase in ROS levels, Caspase-3 activation, and DNA fragmentation and condensation. This study opens new avenues for research by exploring sphaerococcenol A as a scaffold for the synthesis of novel bioactive molecules. Full article

This study aimed to confirm the mediating effects of symptom experiences on the relationship between the body image and quality of life of hemodialysis patients. Data were collected from 153 patients who were diagnosed with ESRD at a tertiary general hospital and receiving regular hemodialysis in Korea. The data were collected between 20 July and 11 August 2023. The following statistical analyses were conducted: t-test, ANOVA, Scheffé test, Pearson’s correlation coefficient analysis, and Hayes’ Process Macro Model 4 (to test the mediating effect). The factors influencing quality of life were body image (β = 0.46, p < 0.001), monthly family income (over KRW 3 million) (β = 0.22, p = 0.002), and symptom experiences (β = −0.20, p = 0.001). The mediation analysis indicated that the symptom experiences mediated the relationship between body image and quality of life. Based on the results, education, counseling, and symptom management programs that can improve body image and reduce symptom experience should be developed, and customized programs that reflect the characteristics of a target population, such as economic level, should be developed and provided. Knowing the spatial structure of traditional villages is required to promote and preserve these villages. These traditional villages are an essential part of China’s farming legacy and hold substantial historical and cultural significance. Therefore, this article analyzed 30 nationally recognized traditional villages in Liaoning Province, selected from the 6819 traditional villages in the province, as samples. These were divided into three types based on elevation: plain-type (below 200 m above sea level), hilly-type (200–500 m), and mountain-type (above 500 m) villages. Two villages of each type were selected for a total of six villages as the study objects; for these, quantitative comparative research on the spatial structure of these villages was carried out. The results of the study show that: (1) plain-type traditional villages are little affected by the terrain, the overall presentation of the surface space, the village traffic is well developed, able to form a commercial street as the core of the road interruptions in the head of the road more; (2) hilly-type traditional villages are influenced by mountains and water systems, forming a linear space with main roads as the core and crossroads, their core areas are more remote and lack space for public activities, and the villages rely on religious venues or the former residences of celebrities to attract tourists; (3) mountain-type villages are greatly influenced by the mountains, making it difficult to form a commercial area, the distribution of each natural town is relatively scattered and forms a point-like space, each point is developed with public space as the core, and there is a lack of characteristics within the village. The above quantitative characteristics are compared and three targeted conservation strategies for national-level traditional villages in Liaoning are proposed. Full article

Cucumber (Cucumis sativus L.) and celery (Apium graveolens L.) are among the most widely cultivated vegetable crops, belonging to the melon and leafy vegetable categories, respectively. This study aims to provide predictions for the quality formation of greenhouse cucumber and celery, as well as technical support for intelligent vegetable production management. Based on the light and temperature response characteristics of cucumber and celery growth and development and using the experimental varieties ‘Jinsheng 206’ for cucumber and ‘Juventus’ for celery, the observational data from a five-cropping season trial over 2 years were utilized. By analyzing the relationship between the growth and development of vegetables and key meteorological factors (radiation and temperature), simulation models for quality formation were constructed using the day–night temperature difference method (ATD) and the product of thermal effectiveness and the photosynthetically active method (TEP) as independent variables. The model parameters were determined and the model was validated using independent trial data. The results showed the following: (1) There exist quantifiable relationships between different quality indicators and ATD or TEP. These relationships are mainly presented through linear functions, exponential functions, logarithmic function, and logical functions. (2) The normalized root mean square error (NRMSE) of the cucumber quality model ranges from 1.13% to 29.53%, and the NRMSE of the celery quality model ranges from 1.63% to 31.47%. (3) Based on two kinds of normalization methods, the average NRMSE of the VQSM model is 13.72%, demonstrating a relatively high level of accuracy in simulation. These results demonstrate that the proposed model can dynamically and accurately simulate the quality formation process of vegetables, providing a theoretical basis and data support for the research on productivity and the economic benefits of greenhouse crops. In fish, increasing the crude lipid level of feed can save protein and improve feed utilization. Mirror carp (Cyprinus carpio) is one of the most widely farmed fish species in the world. In this study, mirror carp larvae were fed isonitrogenous diets with different lipid levels (3%, 5%, 7%, 9%, 11%, and 13%). The rearing trial lasted for eight weeks. The results revealed that when the fat content was 9%, the AWGR, WGR, and FCR were highest, whereas FCR was lowest. The AWGR was correlated with the dietary lipid level, and the regression equation was y = −2.312x2 + 45.01x + 214.49. Compared with those in the control group, the T-CHO and TG contents were significantly greater in the 13% lipid content groups and significantly lower in the 9% lipid content groups (p < 0.05). In terms of muscle quality, the contents of MUFAs, PUFAs, and DHA + EPA were significantly greater than those in the other experimental groups (p < 0.05). Oil red O staining revealed a lipid content of 13% with severe fat deposition. In addition, the results of the analysis of antioxidant enzyme activity revealed that the activities of GSH, CAT and T-AOC were significantly greater at the 9% lipid content, and that the MDA content was significantly greater at the 13% lipid content (p < 0.05). Similarly, the mRNA levels of GH, IGF-I, FAS, and LPL were significantly highest at a lipid level of 9% (p < 0.05). The above results revealed that the optimal dietary lipid requirement for the fast growth of mirror carp (6.86 ± 0.95 g) was 9.74% on the basis of nonlinear regression analysis of the AWGR. The dietary lipid level (9%) improved the growth, stress resistance, and lipid utilization of mirror carp to a certain extent. Full article

This study aimed to develop a population pharmacokinetic (PK) model for meropenem in healthy adults and explore optimal dosing regimens for patients with normal renal function. PK samples were obtained from 12 healthy participants, which were analyzed using noncompartmental analysis and nonlinear mixed-effect modeling. The PK profiles of meropenem were characterized using a two-compartment model, and serum creatinine level was identified as a significant covariate affecting total clearance. Monte Carlo simulations were conducted using this model to inform dosing recommendations. The target index for meropenem efficacy was defined as the cumulative percentage over 24 h during which free (f) drug concentration exceeded the minimum inhibitory concentration (MIC) under steady state conditions (fT>MIC). These simulations indicated that the current dosage regimen of 1 g for 30 min infusions every 8 h achieved a 90% probability of target attainment (PTA) for 40%fT>MIC when the MIC was <2 mg/L. However, to achieve more stringent therapeutic targets, such as a 90%PTA for 100%fT>MIC or a 90%PTA for 100%fT>4MIC, higher doses administered as 3 h extended infusions or as continuous infusions may be necessary. These results highlight the need for model-informed precision dosing to enhance the efficacy of meropenem therapy across various MIC levels and therapeutic targets. Full article

UTRs and Ago-2/miR-335 Complex Restricts Amylin Translation in Insulinoma and Human Pancreatic β-Cells

Amylin promoter and transcriptional factors are well-established, inducible factors in the production of the main amyloidogenic pancreatic hormone, human islet amyloid peptide (hIAPP) or amylin. However, posttranscriptional mechanisms driving hIAPP expression in pancreas remain enigmatic, and hence were explored here. The translational assay revealed that both 5′ and 3′ untranslated regions (UTRs) of hIAPP restricted expression of the luciferase constructs only in constructs driven by the hIAPP promoter. Bioinformatics analysis revealed several putative seed sequences for a dozen micro RNAs (miRNAs) in hIAPP’s 3′ UTR. miR-182, miR-335, and miR-495 were the most downregulated miRNAs in stressed human islets exposed to endoplasmic reticulum (ER) or metabolic stressors, thapsigargin (TG) or high glucose (HG). Correspondingly, miR-335 mimics alone or in combination with miR-495 and miR-182 mimics significantly and potently (>3-fold) reduced hIAPP protein expression in HG-treated cultured human islets. siRNA-mediated silencing of Ago2 but not Ago1 significantly stimulated hIAPP expression and secretion from transfected, HG-treated human islets. Conversely, ectopic expression of Ago2 in hIAPP-expressing RIN-m5F cell line driven by CMV promoter reduced hIAPP intracellular protein levels. Collectively, the results point to a novel and synergistic role for hIAPP promoter, 5/3′ UTRs and Ago-2/miR-335 complex in post-transcriptional regulation of hIAPP gene expression in normal and metabolically active β-cells. Early Biological Response to Poly(ε-Caprolactone)/Alumina-Toughened Zirconia Composites Obtained by 3D Printing for Peri-Implant Application

The improvement of the mucosal sealing around the implant represents a challenge, one that prompted research into novel materials. To this purpose, a printable poly(ε-caprolactone) (PCL)-based composite loaded with alumina-toughened zirconia (ATZ) at increasing rates of 10, 20, and 40 wt.% was prepared, using a solvent casting method with chloroform. Disks were produced by 3D printing; surface roughness, free energy and optical contact angle were measured. Oral fibroblasts (PF) and epithelial cell (SG) tests were utilized to determine the biocompatibility of the materials through cell viability assay and adhesion and spreading evaluations. The highest level of ATZ resulted in an increase in the average roughness (Sa), while the maximum height (Sz) was higher for all composites than that of the unmixed PCL, regardless of their ATZ content. Surface free energy was significantly lower on PCL/ATZ 80/20 and PCL/ATZ 60/40, compared to PCL and PCL/ATZ 90/10. The contact angle was inversely related to the quantity of ATZ in the material. PF grew without variations among the different specimens at 1 and 3 days. After 7 days, PF grew significantly less on PCL/ATZ 60/40 and PCL/ATZ 80/20 compared to unmixed PCL and PCL 90/10. Conversely, ATZ affected and improved the growth of SG. By increasing the filler amount, PF cell adhesion and spreading augmented, while PCL/ATZ 80/20 was the best for SG adhesion. Overall, PCL/ATZ 80/20 emerged as the best composite for both cell types; hence, it is a promising candidate for the manufacture of custom made transmucosal dental implant components. Full article

Evaluating Smart Community Development in China from the Perspective of Residents’ Sense of Safety: An Analysis Using Criteria Importance through Intercriteria Correlation and Fuzzy Comprehensive Evaluation Approaches

As a vital part of smart city development, smart community development is in full swing, aiming to improve residents’ sense of safety (RSS). Most research focuses on the technological innovation and infrastructure construction of smart communities; few studies have evaluated smart community development from the perspective of the RSS. Thus, this paper aims to propose a system of evaluation indicators for assessing the RSS of smart community development. After the relevant evaluation indicators were identified, an evaluation method was proposed using the CRITIC-FCE approach. To validate the feasibility of this method, 31 smart communities in China were selected in this study. The results showed the following: (1) The indicator of the highest weight was ‘cultural activities for the elderly’, while ‘overall design’ had the lowest weight, highlighting the importance of community services in enhancing the RSS of smart community development. (2) The selected cities and communities achieved a generally high level in the RSS of smart community development, and some differences were observed among them. (3) Emergency services and property services play an important role in enhancing the RSS of smart community development, with communities excelling in these areas, achieving higher overall rankings. Conversely, the score of pension service was relatively low, which is a common problem in the evaluation results, reflecting the shortcomings of the current smart community in providing pension service. Consequently, several strategies are suggested to enhance the RSS of smart community development, such as building an emergency information system based on advanced technology, establishing mobile and Internet of Things-based emergency assistance services for the elderly, and enhancing the maintenance of public facilities. This research enriches the knowledge of RSS and provides guidance for further research for the RSS of smart community development. Full article

Design and Testing of an Inter-Toothed, Self-Propelled Digger for Deep-Rhizome Chinese Herbal Medicines

To address the challenges of digging deep-rhizome Chinese herbal medicines in northwest China’s hilly terrain, including difficulty, incompleteness, and herb damage, a specialized self-propelled digger with interlocking teeth has been developed. Designed for complex topography, small fields, and resistant soil, this digger provides an efficient and precise alternative to traditional methods. The prototype features in-place reverse differential steering, 360-degree digging capability, and minimized root and soil damage to promote future planting. Key components, including the digging mechanism, vibratory system, crawler chassis, hydraulic transmission system, and worm gear rotary hydraulic reducer, were analyzed and optimized through theoretical, graphical, and simulation studies using RecurDyn. Field tests demonstrated the digger’s effectiveness, achieving depths exceeding 600 mm with minimal herb damage and loss. The digger successfully navigated steep slopes and operated within noise regulations, surpassing industry standards, with less than 1.4% herb damage and a loss rate under 3%. The digger was capable of ascending gradients over 20° with driver noise levels below 92 dB. This innovative solution offers a valuable reference for developing specialized diggers for harvesting traditional Chinese medicinal materials in challenging conditions. Full article
