The yields of chemical reactions are highly dependent on the mixing pattern between reactants. Herein, we report the modification of a meso-micromixing interaction reaction model which is applied in batch reactors by leveraging the flow characteristics in the continuous reactors. Both experimental and model-predicted yields were compared using the classical Villermaux–Dushman method in a self-designed split and recombination reactor. This modified model significantly reduced the error in predicted product yields from approximately 15% to within 3%, compared to a model containing the micromixing term only. The effects of flow rates and reactor structure parameters on mixing performance were analyzed. We found that increasing flow rates and the degree of twist in the mixing element’s grooves, as well as decreasing the cross-sectional area of grooves, improved mixing performance. The optimization of reactor flow rates and structural parameters was achieved by combining Gaussian process regression and Bayesian optimization with the modified model. This approach provided higher target product yields for consecutive reactions, while simultaneously achieving a lower pressure drop in the reactor. Corresponding combinations of reactor parameters were also identified during this process. Our modified model-based optimization methodology can be applied to a diversity of reactors, serving as a reference for the selection of their structure and operational parameters. Full article

A novel passive micromixer based on multiple baffles and a submergence scheme was designed, and its mixing performance was simulated over a wide range of Reynolds numbers ranging from 0.1 to 80. The degree of mixing (DOM) at the outlet and the pressure drop between the inlets and outlet were used to assess the mixing performance of the present micromixer. The mixing performance of the present micromixer showed a significant enhancement over a wide range of Reynolds numbers (0.1 ≤ Re ≤ 80). The DOM was further enhanced by using a specific submergence scheme. At low Reynolds numbers (Re < 5), submergence scheme Sub24 produced the highest DOM, approximately 0.57, which was 1.38 times higher than the case with no submergence. This enhancement was due to the fluid flowing from or toward the submerged space, creating strong upward or downward flow at the cross-section. At high Reynolds numbers (Re > 10), the DOM of Sub1234 became the highest, reaching approximately 0.93 for Re = 20, which was 2.75 times higher than the case with no submergence. This enhancement was caused by a large vortex formed across the whole cross-section, causing vigorous mixing between the two fluids. The large vortex dragged the interface between the two fluids along the vortex perimeter, elongating the interface. The amount of submergence was optimized in terms of DOM, and it was independent of the number of mixing units. The optimum submergence values were 90 μm for Sub24 and Re = 1, 100 μm for Sub234 and Re = 5, and 70 μm for Sub1234 and Re = 20. Full article

We developed a highly efficient passive mixing device based on a split-and-recombine (SAR) configuration. This micromixer was constructed by simply bonding two identical microfluidic periodical open-trench patterns face to face. The structure parameters of periodical units were optimized through numerical simulation to facilitate the mixing efficiency. Despite the simplicity in design and fabrication, it provided rapid mixing performance in both experiment and simulation conditions. To better illustrate the mixing mechanism, we developed a novel scheme to achieve high-resolution confocal imaging of serial channel cross-sections to accurately characterize the mixing details and performance after each SAR cycle. Using fluorescent IgG as an indicator, nearly complete mixing was achieved using only four SAR cycles in an aqueous solution within a device’s length of less than 10 mm for fluids with a Péclet number up to 8.7 × 104. Trajectory analysis revealed that each SAR cycle transforms the input fluids using three synergetic effects: rotation, combination, and stretching to increase the interfaces exponentially. Furthermore, we identified that the pressure gradients in the parallel plane of the curved channel induced vertical convection, which is believed to be the driving force underlying these effects to accelerate the mixing process. Cryopreservation is the final step of stem cell production before the cryostorage of the product. Conventional methods of adding cryoprotecting agents (CPA) into the cells can be manual or automated with robotic arms. However, challenging issues with these methods at industrial-scale production are the insufficient mixing of cells and CPA, leading to damage of cells, discontinuous feeding, the batch-to-batch difference in products, and, occasionally, cross-contamination. Therefore, the current study proposes an alternative way to overcome the abovementioned challenges; a highly efficient micromixer for low-cost, continuous, labour-free, and automated mixing of stem cells with CPA solutions. Our results show that our micromixer provides a more homogenous mixing of cells and CPA compared to the manual mixing method, while the cell properties, including surface markers, differentiation potential, proliferation, morphology, and therapeutic potential, are well preserved. Micromixers are one of the critical components in microfluidic devices. They significantly affect the efficiency and sensitivity of microfluidics-based lab-on-a-chip systems. This study introduces an efficient micromixer with a simple geometrical feature that enables easy incorporation in a microchannel network without compromising the original design of microfluidic devices. The study proposes a newly designed planar passive micromixer, termed a planar asymmetric contraction-and-expansion (P-ACE) micromixer, with asymmetric vertical obstacle structures. Numerical simulation and experimental investigation revealed that the optimally designed P-ACE micromixer exhibited a high mixing efficiency of 80% or more within a microchannel length of 10 mm over a wide range of Reynolds numbers (0.13 ≤ Re ≤ 13), eventually attaining approximately 90% mixing efficiency within a 20 mm microchannel length. The highly asymmetric geometric features of the P-ACE micromixers enhance mixing because of their synergistic effects. The flow velocities and directions of the two fluids change differently while alternately crossing the longitudinal centerline of the microchannel, with the obstacle structures asymmetrically arranged on both sidewalls of the rectangular microchannel. This flow behavior increases the interfacial contact area between the two fluids, thus promoting effective mixing in the P-ACE micromixer. Further, the pressure drops in the P-ACE micromixers were experimentally investigated and compared with those in a serpentine micromixer with a perfectly symmetric mixing unit. Full article

A passive micromixer combined with two different mixing units was designed by submerging planar structures, and its mixing performance was simulated over a wider range of the Reynolds numbers from 0.1 to 80. The two submerged structures are a Norman window and rectangular baffles. The mixing performance was evaluated in terms of the degree of mixing (DOM) at the outlet and the required pressure load between inlet and outlet. The amount of submergence was varied from 30 μm to 70 μm, corresponding to 25% to 58% of the micromixer depth. The enhancement of mixing performance is noticeable over a wide range of the Reynolds numbers. When the Reynolds number is 10, the DOM is improved by 182% from that of no submergence case, and the required pressure load is reduced by 44%. The amount of submergence is shown to be optimized in terms of the DOM, and the optimum value is about 40 μm. This corresponds to a third of the micromixer depth. The effects of the submerged structure are most significant in the mixing regime of convection dominance from Re = 5 to 80. In a circular passage along the Norman window, one of the two Dean vortices burst into the submerged space, promoting mixing in the cross-flow direction. The submerged baffles in the semi-circular mixing units generate a vortex behind the baffles that contributes to the mixing enhancement as well as reducing the required pressure load. Full article

A new passive micro-mixer with mixing units stacked in the cross flow direction was proposed, and its performance was evaluated numerically. The present micro-mixer consisted of eight mixing units. Each mixing unit had four baffles, and they were arranged alternatively in the cross flow and transverse direction. The mixing units were stacked in four different ways: one step, two step, four step, and eight step stacking. A numerical study was carried out for the Reynolds numbers from 0.5 to 50. The corresponding volume flow rate ranged from 6.33 μL/min to 633 μL/min. The mixing performance was analyzed in terms of the degree of mixing (DOM) and relative mixing energy cost (MEC). The numerical results showed a noticeable enhancement of the mixing performance compared with other micromixers. The mixing enhancement was achieved by two flow characteristics: baffle wall impingement by a stream of high concentration and swirl motion within the mixing unit. The baffle wall impingement by a stream of high concentration was observed throughout all Reynolds numbers. The swirl motion inside the mixing unit was observed in the cross flow direction, and became significant as the Reynolds number increased to larger than about five. The eight step stacking showed the best performance for Reynolds numbers larger than about two, while the two step stacking was better for Reynolds numbers less than about two. Full article

Mixing at the microscale is of great importance for various applications ranging from biological and chemical synthesis to drug delivery. Among the numerous types of micromixers that have been developed, planar passive spiral micromixers have gained considerable interest due to their ease of fabrication and integration into complex miniaturized systems. However, less attention has been paid to non-planar spiral micromixers with various cross-sections and the effects of these cross-sections on the total performance of the micromixer. Here, mixing performance in a spiral micromixer with different channel cross-sections is evaluated experimentally and numerically in the Re range of 0.001 to 50. The accuracy of the 3D-finite element model was first verified at different flow rates by tracking the mixing index across the loops, which were directly proportional to the spiral radius and were hence also proportional to the Dean flow. It is shown that higher flow rates induce stronger vortices compared to lower flow rates; thus, fewer loops are required for efficient mixing. The numerical study revealed that a large-angle outward trapezoidal cross-section provides the highest mixing performance, reaching efficiencies of up to 95%. Moreover, the velocity/vorticity along the channel length was analyzed and discussed to evaluate channel mixing performance. A relatively low pressure drop (<130 kPa) makes these passive spiral micromixers ideal candidates for various lab-on-chip applications. Full article

Optimal Combination of Mixing Units Using the Design of Experiments Method

A passive micromixer was designed by combining two mixing units: the cross-channel split and recombined (CC-SAR) and a mixing cell with baffles (MC-B). The passive micromixer was comprised of eight mixing slots that corresponded to four combination units; two mixing slots were grouped as one combination unit. The combination of the two mixing units was based on four combination schemes: (A) first mixing unit, (B) first combination unit, (C) first combination module, and (D) second combination module. The statistical significance of the four combination schemes was analyzed using analysis of variance (ANOVA) in terms of the degree of mixing (DOM) and mixing energy cost (MEC). The DOM and MEC were simulated numerically for three Reynolds numbers (Re = 0.5, 2, and 50), representing three mixing regimes. The combination scheme (B), using different mixing units in the first two mixing slots, was significant for Re = 2 and 50. The four combination schemes had little effect on the mixing performance of a passive micromixer operating in the mixing regime of molecular dominance. The combination scheme (B) was generalized to arbitrary mixing slots, and its significance was analyzed for Re = 2 and 50. The general combination scheme meant two different mixing units in two consecutive mixing slots. The numerical simulation results showed that the general combination scheme was statistically significant in the first three combination units for Re = 2, and significant in the first two combination units for Re = 50. The combined micromixer based on the general combination scheme throughout the entire micromixer showed the best mixing performance over a wide range of Reynolds numbers, compared to other micromixers that did not adopt completely the general combination scheme. The most significant enhancement due to the general combination scheme was observed in the transition mixing scheme and was negligible in the molecular dominance scheme. Droplet-based micromixers have shown great prospects in chemical synthesis, pharmacology, biologics, and diagnostics. When compared with the active method, passive micromixer is widely used because it relies on the droplet movement in the microchannel without extra energy, which is more concise and easier to operate. Here we present a droplet rotation-based microfluidic mixer that allows rapid mixing within individual droplets efficiently. PDMS deformation is used to construct subsidence on the roof of the microchannel, which can deviate the trajectory of droplets. Thus, the droplet shows a rotation behavior due to the non-uniform distribution of the flow field, which can introduce turbulence and induce cross-flow enhancing 3D mixing inside the droplet, achieving rapid and homogenous fluid mixing. In order to evaluate the performance of the droplet rotation-based microfluidic mixer, droplets with highly viscous fluid (60% w/w PEGDA solution) were generated, half of which was seeded with fluorescent dye for imaging. Mixing efficiency was quantified using the mixing index (MI), which shows as high as 92% mixing index was achieved within 12 mm traveling. Here in this work, it has been demonstrated that the microfluidic mixing method based on the droplet rotation has shown the advantages of low-cost, easy to operate, and high mixing efficiency. It is expected to find wide applications in the field of pharmaceutics, chemical synthesis, and biologics. Full article

In this paper, we present a novel microfluidic mixer with staggered virtual electrode based on light-actuated AC electroosmosis (LACE). We solve the coupled system of the flow field described by Navier–Stokes equations, the described electric field by a Laplace equation, and the concentration field described by a convection–diffusion equation via a finite-element method (FEM). Moreover, we study the distribution of the flow, electric, and concentration fields in the microchannel, and reveal the generating mechanism of the rotating vortex on the cross-section of the microchannel and the mixing mechanism of the fluid sample. We also explore the influence of several key geometric parameters such as the length, width, and spacing of the virtual electrode, and the height of the microchannel on mixing performance; the relatively optimal mixer structure is thus obtained. The current micromixer provides a favorable fluid-mixing method based on an optical virtual electrode, and could promote the comprehensive integration of functions in modern microfluidic-analysis systems. A fast mixing is critical for subsequent practical development of microfluidic devices, which are often used for assays in the detection of reagents and samples. The present work sets up computational fluid dynamics simulations to explore the flow characteristic and mixing mechanism of fluids in cross-shaped mixers within the laminar regime. First, the effects of increasing an operating parameter on local mixing quality along the microchannels are investigated. It is found that sufficient diffusion cannot occur even though the concentration gradient is large at a high Reynolds number. Meanwhile, a method for calculating local mixing efficiency is also characterized. The mixing efficiency varies exponentially with the flow distance. Second, in order to optimize the cross-shaped mixer, the effects of design parameters, namely aspect ratio, mixing angle and blockage, on mixing quality are captured and the visualization of velocity and concentration distribution are demonstrated. The results show that the aspect ratio and the blockage play an important role in accelerating the mixing process. They can improve the mixing efficiency by increasing the mass transfer area and enhancing the chaotic advection, respectively. In contrast, the inflow angle that affects dispersion length is not an effective parameter. Besides, the surface roughness, which makes the disturbance of fluid flow by roughness more obvious, is considered. Three types of rough elements bring benefits for enhancing mixing quality due to the convection induced by the lateral velocity. Full article

In this work, a comparative investigation of chaotic flow behavior inside multi-layer crossing channels was numerically carried out to select suitable micromixers. New micromixers were proposed and compared with an efficient passive mixer called a Two-Layer Crossing Channel Micromixer (TLCCM), which was investigated recently. The computational evaluation was a concern to the mixing enhancement and kinematic measurements, such as vorticity, deformation, stretching, and folding rates for various low Reynolds number regimes. The 3D continuity, momentum, and species transport equations were solved by a Fluent ANSYS CFD code. For various cases of fluid regimes (0.1 to 25 values of Reynolds number), the new configuration displayed a mixing enhancement of 40%–60% relative to that obtained in the older TLCCM in terms of kinematic measurement, which was studied recently. The results revealed that all proposed micromixers have a strong secondary flow, which significantly enhances the fluid kinematic performances at low Reynolds numbers. The visualization of mass fraction and path-lines presents that the TLCCM configuration is inefficient at low Reynolds numbers, while the new designs exhibit rapid mixing with lower pressure losses. Optimum configuration of a micromixer with two-layer crossing microstructure was performed using mixing analysis, surrogate modeling, along with an optimization algorithm. Mixing performance was used to determine the optimum designs at Reynolds number 40. A surrogate modeling method based on a radial basis neural network (RBNN) was used to approximate the value of the objective function. The optimization study was carried out with three design variables; viz., the ratio of the main channel thickness to the pitch length (H/PI), the ratio of the thickness of the diagonal channel to the pitch length (W/PI), and the ratio of the depth of the channel to the pitch length (d/PI). Through a primary parametric study, the design space was constrained. The design points surrounded by the design constraints were chosen using a well-known technique called Latin hypercube sampling (LHS). The optimal design confirmed a 32.0% enhancement of the mixing index as compared to the reference design. Full article

In this paper, we characterized an assortment of photopolymers and stereolithography processes to produce 3D-printed molds and polydimethylsiloxane (PDMS) castings of micromixing devices. Once materials and processes were screened, the validation of the soft tooling approach in microfluidic devices was carried out through a case study. An asymmetric split-and-recombine device with different cross-sections was manufactured and tested under different regime conditions (10 < Re < 70). Mixing performances between 3% and 96% were obtained depending on the flow regime and the pitch-to-depth ratio. The study shows that 3D-printed soft tooling can provide other benefits such as multiple cross-sections and other potential layouts on a single mold. Full article
