Objectives:
1. Name the four biological macromolecules and their building blocks
2. Test food samples for the presence of lipids, proteins, and simple and complex sugars
Materials
Procedure:
Testing for carbohydrates
Testing for the presence of starch (complex sugar)
Lugol's reagent (iodine solution) changes from yellowish-brown to dark purple/black.
1. Demonstrate how to perform this test.
2. Put 2-3 ml of starch solution in a test tube
3. Add 3-4 drops of iodine
4. A bluish black color indicates a positive test for starch
5. Testing for simple carbohydrates (monosaccharides and some disaccharides)
Benedict's solution is used to test for simple carbohydrates. Benedict's solution is a blue colored
liquid that contains copper ions. When Benedict's solution and simple carbohydrates are heated, the
solution changes to orange red/ brick red.
1. Put 2-3 ml of corn syrup in a test tube
2. Add 1ml of the Benedict Reagent, the solution will look blue
3. Put the tube in a gently boiling water bath. Wait a few minutes.
4. The glucose present in the solution reacts with the copper sulfate in the Benedict reagent and
makes copper oxide that is an orange to red-brick precipitate.
5. The intensity of the color depends on the concentration of glucose present in the sample.
