The text discusses how temperature and pH impact the rate of enzyme reactions, with every enzyme having an optimal temperature and pH range.

The data shows that the enzyme lactase works best at 40 degrees Celsius, and outside of this optimal temperature the reaction rate decreases significantly.

The data indicates that lactase works best at a pH of 7, while outside of this pH range the reaction rate is much lower.

Lactase Enzyme Activity with Data Analysis

Simulation at Jon Darkow

Pre-Lab:
1. Read the Background section and view the
diagram to complete the following. Define the
following and explain how each relates to the
lactase chemical reaction (see diagram for help
as well).

Term Definition and Relation to the Lactase Chemical Reaction

Enzyme a catalyst that brings a chemical reaction. it digests the lactase

Substrate a molecule upon which an enzyme acts

Active Site where the chemical reaction occurs

Reaction Rate at what speed the chemical reaction occurs

Activation the energy used to undergo a certain chemical reaction

Energy

2. What type of biomolecule is an enzyme?

proteins

3. What is the monomer of an enzyme?

amino acids

4. What is another name for an enzyme?

catalyst

5. How do enzymes speed up chemical reactions?

they lower the activation energy for reactions

Simulation Option 1: Temperature

Start with the initial lactose at 500 mg/dL and the pH at 7.

Do not vary these parameters while testing for temperature,
they are environmental controls. The temperature is in
degrees Celsius. Vary the temperature by 20oC.

Run each experiment 3 times (for reliability in the results.)

** Click on the 20 minute mark to get your run # see photo

Temperature Data table:

0oC 20oC 40oC 60oC 80oC

Run 1 0 470 493 115 0

Run 2 0 471 497 140 0

Run 3 0 472 496 119 0

Average: 0 471 495 124 0

Temperature Analysis:

1. What temperature did the enzyme lactase work best? This is considered the optimal temperature.
Explain your answer using data from your simulation.

40 degrees. at that temperature there was the highest data compared to all the other temperatures
data

2. Explain what happened to the rate of reaction when lactase was placed out of its optimal temperature.
Why does this happen?

the data came out as 0 each run

Simulation Option 2: pH
Start with the initial lactose at 500 mg/dL and the
temperature at 25 degrees Celsius. Do not vary these
parameters while testing for pH, they are environmental
controls.

Vary by a pH of 3. Run each experiment 3 times (for

reliability in the results.)

** Click on the 20 minute mark to get your run # see photo

pH Date Table:

pH 1 4 7 10 13

Run 1 0 237 489 128 5.82

Run 2 0 218 496 105 6.05

Run 3 0 230 494 101 6.42

Average 0 228 493 111 6

pH Analysis:

1. What pH did the enzyme lactase work best? This is considered the optimal pH. Explain your answer
using data from your simulation.

a pH of 7 worked best. my answer is proven through my data, it is clear that when the pH was 7 it was
a lot higher numbers in the data than other pH levels

2. Explain what happened to the rate of reaction when lactase was placed out of its optimal pH. Why
does this happen?

the reaction rate was very low, either 0 or below ten

Conclusion: Write a CER paragraph explaining the results. Be sure to include the following in your
CER.

● Summarizes data used to draw conclusions.

● Conclusions follow data (no wild guesses or leaps of logic).
● Hypothesis is rejected or accepted based on the data.
● Discusses possible experimental errors.
● Discusses applications or real world connections.

Claim: The claim should be a complete sentence that answers the question:
How does temperature and pH affect enzyme controlled reactions?

there is an optimal zone for pH, and for temperature that affects the enzyme reaction rates.

Evidence:
- You must include 3 pieces of evidence from the Virtual Lab – data that supports your claim!

in my temperature data, when the temperature was too high or too low the data came out as 0. this
similarly occurred with my pH data. for both there is a clear optimal zone for the enzyme reaction rates
to thrive

Reasoning:
-Connect your evidence to your understanding of how enzymes affect the rate of chemical reactions.
--You need to discuss each piece of evidence. You should not discuss any data that you did not highlight in your
evidence section.
