The HUMACLOT VA coagulometer is a completely automatic instrument created to

determinate the main parameters of the plasma coagulation methods:

! Prothrombin Time or Quick Time (PT)

! Activated Partial Thromboplastin Time (APTT)
! Fibrinogen Concentration (FB)
! Thrombin Time (T) and factors studies

The user should prepare the samples and reagents and place them in the instrument,
introduce the patient’s data and chose the essays to be run.
HUMACLOT VA makes the dilutions required and respects the incubation times for every
step of the process and perform the samples in a completely automatic way.
At the end of the analyses, the results are printed out and displayed.

The aspiration and dispensation of samples and reagents in the reaction cups is automatic,
to obtain a completely standardisation of the methods.

The system detects the clot formation by means of a continuous reading of the optical
density. The timer and the mixing system are activated when a high variation of the optical
density takes place, which permits start the measuring when the sample contacts the
reagent, and also to stop when the clot is formed.

The permanent mixing permits a perfect homogenisation and facilitates the measuring of the
Fibrinogen concentration at low concentrations due to the fibrin filaments agglutinate in the
exe of the optical path.

The software allows to adapt the instrument to all needs and uses of every laboratory.

MODULAR DESCRIPTION OF THE INSTRUMENT:
The HUMACLOT VA has the following elements:

! Automatic pipette to intake and dispense the samples and reagents. There is a
position to place a cuvette with deproteinisant solution.
! Automatic cleaning zone.
! Automatic dilutor.
! Plate for 32 sample cuvettes and 32 reaction cups.
! 6 positions for reagents, one of them with continuous mixing.
! Compartment to place the pipette cleaning solution bottle.
! Waste tube connected to the waste reservoir.
! Keypad.
! Printer.

The HUMACLOT VA has three fundamental parts: one mechanic, one hydraulic and another
electrical.

! The heart of the mechanical part is a metallic rotor with capacity for 32 samples and
32 reaction cups. 5 of these positions are always at 37ºC. This plate moves thanks to
a step by step motor of high features.
The probe has a conductimetric level detection system.
The mechanical part includes the compartment for the 6 reagents vials.

capillary for aspiration and samples and reagents dispensation. Both movements are
done by means of a step by step precision motor.

 ! The hydraulic part includes all the liquid circuit that allows the samples and reagents
dilution preparations. The reproducibility of the dilutions is assured by means of the
microsyringe controlled by a step by step motor of high precision.
To guarantee the absence of contamination, the pipette has a cleaning station that
complements the washing with a deproteinisant solution, that eliminates the possible
fibrin rest.

! The electronic part has been accurately selected to give the maximum accuracy and
output. It has a modular composition.
The software is fruit of a rigorous study of the principal laboratory requirements. It is
very easy to use, being also very attractive to the user.

To correctly install the HUMACLOT VA, the following instructions should be followed:

HUMACLOT VA should be stored in a dry place without extreme temperatures.

During the unpacking process be sure that the instrument and all accessories are in good
conditions and have no defects due to the transportation.
The accessories that are included with the instrument are:

! Fuse
! Thermal roll paper
! Reaction cups
! Sample cuvettes + stirring bars

In case any anomaly is detected, please fulfil the Return Tag sheet included in the
instrument, and send it to your HUMAN distributor.

a) Requirements of the place:
HUMACLOT VA should be installed in a place without any vibration nor direct light,
because its detection system is based on variation of light absorbance and all external
interference’s must be avoided.
Moreover, the room temperature should be between 15 and 25ºC, and the humidity
mustn’t be excessive.
It should be placed in a place to allow the sufficient ventilation of the instrument and the
correct wasting of the washing system.

b) Technical requirements:
The current connection must be done by means of a 3 plugs socket with ground
connection for 220 VAC. It is indispensable a good and stable ground connection. If the
current oscillates more than 10% over the nominal value, a stabiliser should be placed.

In the main menu 4 functions appear:

1. METHODS
2. WORKLIST
3. PROFILES
4. UTILITIES

1. Methods: it is used to program a work season of several samples for one kind of
method (ex.: 30 PT). The identification of the first sample is introduced, the rest are
introduced sequentially. HUMACLOT VA prints out all the results after every
determination.
2. Worklist: it is used to program several methods of several samples. It is the most
useful method in most of the laboratories.
The identification of the sample is introduced and then the methods to be done are
chosen. After programming all the samples press OK, and the instrument indicates
the position where the reagents must be placed . After finishing the essays the results
are printed out.
If during the process all cuvettes are used, an alarm sounds and it indicates that the
cuvettes must be changed by new ones. Then the analyse can go ahead.
3. Profiles: it is used to program the same group of methods (ex.: PT and APTT) to
several samples.
4. Utilities: this option allows to get into the UTILITIES menu, which includes the SET
UP, the programming and control of the equipment to adapt it to the user
requirements.

deviation for Quality Control, etc).

5. See QC. It is used to see the data stored in the QC program.

6. Results file: HUMACLOT VA allows to store the last two working seasons.
