Se la coneix pels seus poemes, que tracten temes com la igualtat, el feminisme, la justícia social o el judaisme. De marcada tendència progressista, va cobrir com a corresponsal el cas dels i va treballar per a la . El 1936 era a Barcelona per cobrir l'Olimpíada Popular i va viure de primera mà el Cop d'estat del juliol del 1936 a Barcelona, fets que van impactar la seva obra. Combinava poesia i ciència en obres com Theory of Flight (1935) o U.S.I. (1938). El 1938 va publicar The Book of the Dead, que documentava els detalls del . Kenneth Rexroth said that she was the greatest poet of her "exact generation." One of her most powerful pieces was a group of poems titled The Book of the Dead (1938), documenting the details of the Hawk's Nest incident, an industrial disaster in which hundreds of miners died of silicosis. Her poem "To be a Jew in the Twentieth Century" (1944), on the theme of Judaism as a gift, was adopted by the American Reform and Reconstructionist movements for their prayer books, something Rukeyser said "astonished" her, as she had remained distant from Judaism throughout her early life.
