Received: Revised: Accepted: Published online: September 07, 2015 October 06, 2015 November 01, 2015 January 22, 2016 The aim of this study was to evaluate the changes that occur in the acute phase response (APR) of experimentally-induced salmonellosis in buffalo calves and to identify potential indicators of infection. Five buffalo calves received 10 colonyforming units of Salmonella typhimurium, orally. Another five animals were used as control group. Clinical examinations were performed and venous blood was sampled before and throughout 7 days after inoculation. Red blood cell count (RBC), packed cell volume (PCV), hemoglobin (Hb), plasma fibrinogen, serum haptoglobin, ceruloplasmin, transferrin and iron were measured. Severe diarrhea started 72-108 h after inoculation, in all animals. The infectious stimulus induced severe APR, characterized by pyrexia, lymphopenia (4.99±1.73x10 cells/L), increase in fibrinogen (24.7±4.91 μmol/L), haptoglobin (21.5±8.65 μmol/L) and ceruloplasmin...

To determine whether platelets become activated and form platelet-platelet or platelet-neutrophil aggregates, or both, when subjected to shear. Blood obtained from 3 Thoroughbreds. Blood, with PCV adjusted to 32 (low hematocrit) or 60 (high hematocrit)%, was subjected to shear rates of 11.25, 22.5, 45, 90, 225, and 750/s for 3 minutes by use of a cone-plate viscometer. Flow cytometric techniques were used to identify activated platelets, platelet-platelet aggregates, and platelet-neutrophil aggregates. Shear resulted in decreased platelet count, increased mean platelet volume, platelet activation, and formation of platelet-platelet and platelet-neutrophil aggregates. These changes occurred at lower shear rates in blood with high hematocrit. Platelet-neutrophil aggregate formation was inhibited by blocking P-selectin, but not CD11/CD18 receptors. Shear-induced platelet activation and aggregate formation occur at physiologic shear rates. Shear-induced platelet activation may explain t...

To determine whether plasma protein concentrations were altered in ponies with alimentary laminitis. 12 adult ponies. Acute laminitis was induced in 6 ponies by oral administration of carbohydrate (85% corn starch, 15% wood flour); the other 6 ponies were used as controls. A physical examination was performed and blood samples were collected immediately before and 4, 8, 12, 24, and 28 hours after administration of carbohydrate. Plasma protein concentrations were determined by means of sodium dodecyl sulphate-polyacrylamide gel electrophoresis. 19 plasma proteins ranging from a molecular weight of 24,000 to a molecular weight of 350,000 were identified in all 12 ponies. Plasma concentrations of proteins with molecular weights of 350,000 (fibrinogen), 130,000 (ceruloplasmin), 118,000 (c-reactive protein), 67,000 (alpha1-antitrypsin I), 65,000 (alpha1-antitrypsin II), 50,000 (haptoglobulin), and 45,000 (acid glycoprotein) were significantly increased in ponies with laminitis, compared with concentrations in control ponies. Changes in plasma protein concentrations are detectable within 4 hours after the onset of alimentary laminitis in ponies. Clinical Relevance-Measurement of plasma protein concentrations may be useful in monitoring the progression of laminitis in ponies.

ABSTRACT: In face of the few reports found in national literature analyzing the potential influence of parturition number in serum proteinogram and biochemical profile in the peripartum period of high yielding dairy cows, the aim of the present study was to comparatively evaluate the dynamics of these serum constituents’ concentrations in blood samples obtained from primiparous and multiparous Holstein cows, 60 and 30 days prepartum and in the day of parturition. Data were analyzed by repeated measures variance analysis (ANOVA) and differences between groups and moments were analyzed by Tukey’s test. Results were considered significant when P<0.05. Parity influenced levels of total protein, albumin, globulins, magnesium, cholesterol, which were higher in multiparous cows, as well as concentrations of ceruloplasmin, total calcium, chloride and alkaline phosphatase activity, which were higher in primiparous cows. Parturition influenced serum concentrations of ceruloplasmin (+58%), ...

The efficacy of intravenous heparin administration (100UI/kg/8h, from 24 to 48h after carbohydrate administration) in the control of carbohydrate overload-induced equine laminitis (17.6g of corn starch/kg live weight) was evaluated. Fifteen horses were allocated into three experimental groups: GI (control group), GII (laminitis group), and GIII (laminitis+heparin group). These animals were submitted to physical and laboratorial examination during 48h. After that time, they were euthanized with intravenous administration of 5ml of acepromazine followed by 1g of thiopental sodium and 1 liter of saturated solution of KCl to obtain hoof tissues samples for histological examination. GII and GIII horses developed laminitis, showing lameness 12 and 24h after carbohydrate administration, respectively, as well increased heart rate and capillary refill time. The histological alterations, similar in GII and GIII, were degenerative lesions, as thinness of epidermal lamina, retraction, flattenin...

Early diagnosis of canine ehrlichiosis favors prompt institution of treatment and improves the prognosis for the animal, since this disease causes mortality among dogs. Studies have shown that determining the concentration of acute-phase proteins (APPs) may contribute towards early detection of disease and aid in predicting the prognosis. This study aimed to evaluate the APP profile in dogs experimentally infected with Ehrlichia canis, at the start of the infection and after treatment. It also investigated whether any correlation between APP levels and the clinical and laboratory alterations over the course of the disease would be possible. The results obtained showed abnormal levels of all the APPs on the third day after infection (D3), with the highest levels being reached on D18, with the exception of ceruloplasmin and acid glycoprotein, which presented their peaks on D6 and D12 respectively. We concluded that assessment of APP levels could contribute towards establishing an earl...

Hemograms and acute-phase proteins in adult male New Zealand White rabbits that had been experimentally infected orally with sporulated oocysts of Eimeria stiedai were evaluated over a 28-day period. Fifty animals were used, divided into two groups: group A infected with 1 × 10(4) sporulated oocysts of E. stiedai and group B inoculated with distilled water. On the seventh day after infection, the infected animals presented anemia and leukocytosis with neutrophilia and monocytosis. Protein fractionation by means of electrophoresis identified 19 acute-phase proteins with molecular weights ranging from 24 to 238 kD. Ceruloplasmin, transferrin and haptoglobin showed high levels on the seventh day after infection, with gradual increases in their concentrations until the end of the experimental period. Thus, from the data of the present study, E. stiedai is considered to be a pyogenic etiological agent for which the infection level can be monitored through the leukocyte count and serum co...

Passive immunity transfer (PIT) evaluation is an essential tool for the maintenance of healthy calves during the first months of life. Since lactation number and breed have been proven to influence immunoglobulin levels in colostrum, the aim of this study was to evaluate PIT from primiparous and multiparous Canchim cows to their calves. Blood samples were collected from the calves before colostrum intake and 1, 2, 7, 15 and 30 days thereafter, while colostrum samples from the cows were taken immediately after parturition. Activities of gamma-glutamyl transferase (GGT), alkaline phosphatase (ALP), and concentrations of total protein, albumin, globulins, immunoglobulin A (IgA), immunoglobulin G (IgG), total and ionized calcium, inorganic phosphorus, magnesium, sodium and potassium were evaluated in calves' serum and activities of GGT and ALP and concentrations of total protein, IgA and IgG were assessed in…
