Suicidal attempts and self–harm are essential predictors of suicide in future and intolerable suffering. Assessment in this domain is challenging as no agreement is observed to conceptualize the behaviors. Our study aims to assess the prevalence rate of suicidal attempts and thoughts and self–harm thoughts in young adults. This cross-sectional study was conducted at Shaheed Mohtarma Benazir Bhutto Medical University for 6 months. An online questionnaire was prepared and circulated among young adults. Informed consent was obtained from all participants. The questionnaire included many demographic questions, including age, gender, living status, and relationship status. The data was analyzed using SPSS 24.0. For all qualitative variables, frequencies and percentages were measured. A total of 401 responses were collected. The number of ...

Fibroblast growth factor 21 (FGF-21) is a secreted protein, which is involved in glucose homeostasis. We speculate that the recombinant canine FGF-21 (cFGF-21) has the potential to become a powerful therapeutics to treat canine diabetes. The cFGF-21 gene was cloned and expressed in E. coli Rosetta (DE3). After purification, a cFGF-21 protein with the purity exceeding 95% was obtained. Mouse 3T3-L1 adipocytes and type 1 diabetic mice/dogs induced by STZ were used to examine the biological activity of cFGF-21 in vitro and in vivo, respectively. Results showed that cFGF-21 stimulated glucose uptake in adipocytes significantly in a dose-dependent manner, and reduced plasma glucose significantly in diabetic mice/dogs. After treatment with cFGF-21, the serum insulin level, glycosylated hemoglobin (HbA1c) level and the expressions of the hepatic gluconeogenesis genes (glucose-6-phosphatase, G6Pase and phosphoenolpyruvate carboxykinase, P...

However, little is known about the therapeutical effects of FGF21 on DN and underlying mechanism. In this study, FGF21 significantly ameliorated blood glucose, HbAlc, insulin resistance, renal function and histopathological change in DN mice (BKS-Leprem2Cd479/Gpt), which develop abnormalities in renal morphology and function. Our results showed that administration of FGF21 upregulated the autophagy related genes LC3Ⅱ and BCL-1 mRNA and protein expression levels. D-glucose was used for high glucose (HG) model in mesangial cells. The results showed that treatment with FGF21 reduced the levels of ROS, AGEs and inflammatory cytokines and significantly downregulated the protein expression of PCNA. Meanwhile, FGF21 significantly enhanced the expression of LC3Ⅱ and BCL-1. Besides, Our studies showed that administration of FGF21 significantly upregulated the phosphorylation...

The immune stimulatory cytokine interleukin-15 (IL-15) was recognized as one of the most promising drugs for cancer treatment and is currently applied in clinical trials alone or as an adjuvant for certain types of metastatic solid tumors. But the administration of IL-15 alone has faced some hurdles that made its antitumor effects not very satisfying, here comes the need to establish new approaches for demonstrating IL-15 in a more effective way that can overcome these obstacles and get maximum benefit from its effects in vivo. One of the alternative approaches is the use of viruses as carriers to deliver foreign genes (virotherapy) which has been used almost two decades ago. Since the 1990s when the first genetically engineered virus was described to selectively replicate in tumor cells, several preclinical and cli...

FGF21 is a key regulating endocrine, lipid metabolism, glucose metabolism. Fibroblast growth factor-21 improves glucose tolerance in obese mice, lowers serum free fatty acids (FFA) & results in weight loss. The FGF-21 in obese & diabetes rodent models create confirmatory metabolic variations which consist of dyslipidemia, hyperglycemia, and a decrease in body weight achieved by insulin sensitivity, glucose uptake in peripheral tissues, rise in energy consumption, fat usage, reducing the production of glucagon on the islet of alpha cells. However, FGF-21 rectifies several metabolic complications and has a promising strategic role in therapeutic aim for treatment of obesity, T2DM, dyslipidemia and other related metabolic

Notably, Lactobacillus brevis is the most common species of LAB that produces γ-aminobutyric acid (GABA). The aim of this study is to clarify the effect of time, strain types, antibiotic concentrations, different levels of pH, and intestinal juices in aerobic or anaerobic conditions and the effect of interactions between these factors on the potential properties of KLDS 1.0727 and KLDS 1.0373, furthermore, antagonistic activity against foodborne pathogens. Moreover, another aim is to study the capability of KLDS 1.0727 and KLDS 1.0373 strains as gad gene carriers to express GABA that reduce the risk of type 1 diabetes in C57BL/6 mice as diabetic models. The obtained results exhibited the surprising tolerance of Lactobacillus brevis strains in vitro digestion models mimicking the conditions of the gastrointestinal tract, further, large antagonistic activity against foodborne pa...

DTF consisted of an anti-IL-17A single chain variable fragment, a soluble TNF receptor 1, and an Fc fragment. Both clinical and histopathological evaluations suggest that DTF and etanercept can ameliorate collagen induced arthritis. However, the arthritis severity score of DTF-treated mice was lower than that of etanercept-treated mice. In addition, DTF was more potent than etanercept in decreasing the ratio of RANKL/OPG in the serum and rebalancing the population ratio of Treg/Th17 cells in the spleens. In vitro, IL-17A and TNF-α had synergistic effects in inducing the expression of inflammatory cytokines in fibroblast-like synoviocyte from RA patients (RA-FLS), human leukemia (THP-1), and rheumatoid synovial fibroblast (MH7A). IL-17A and TNF-α also had synergistic...

In this paper, we show that FGF-21 has a potential role in anti-inflammation and immunoregulation. In vivo, treatment with exogenous FGF-21 can alleviate LPS-induced inflammation. In vitro, FGF-21 inhibited LPS-induced IL-1β expression in THP-1 cells. Furthermore, besides the NF-κB pathway, the mechanism of action of FGF-21 was observed to involve the elevation of IL-10 in the ERK1/2 pathway. This study clearly indicates that FGF21 can be used as an attractive target for the management of inflammatory disorders. This piece of research indicates that FGF-21 could have much value in the management of inflammatory disorders.
