Herein, we present a simple simultaneous method for the fabrication of poly(ethylene glycol) diac... more Herein, we present a simple simultaneous method for the fabrication of poly(ethylene glycol) diacrylate/alginate (PEGDA/ALG) hydrogels as three-dimensional (3D) scaffolds for ciprofloxacin (CIP) encapsulation using gamma-irradiation at an irradiation dose of 15 kGy. The absorption capacity of the hydrogels studies demonstrated that the PEGDA/ALG hydrogels showed a small absorption capacity at an acidic pH (pH = 2) and a high absorption capacity at a basic pH (pH = 10). The morphology and functional groups of the PEGDA/ALG hydrogels were successfully determined using scanning electron microscopy (SEM) and fourier transform infrared spectroscopy (FT-IR), respectively. SEM images observed a decrease in the pore size of the hydrogels with increasing alginate concentration, indicating successful alginate incorporation. FT-IR spectra of the hydrogels indicated that the PEGDA and alginate were crosslinked. The antimicrobial experiment showed that all the PEGDA/ALG hydrogels possessed good antimicrobial activity against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli). These results demonstrate the practicality and efficacy of gamma-irradiation as a one-step method for the simultaneous fabrication of PEGDA/ALG hydrogels and encapsulation of CIP without compromising the bioactivity of the encapsulated CIP.

Due to the limitations of daily used oral hygiene products, there is an unmet need for new, effective, safe, and economic oral products. We have recently demonstrated that N-(2(2,6-diaminohexanamide)-chitosan (CS3H Lys) has enhanced antibacterial properties against Streptococcus mutans, the main cariogenic bacterium, and here we investigated the effect of fluoridation of this polymer (CS3H Lys F) on its antibacterial properties and the ability to protect teeth from acid demineralization. We further formulated this polymer into mouthwash preparations and studied their cytocompatibility and physicochemical stability over 6 months. CS3H Lys F was 1.6-fold more effective than the highest tested oral NaF dose in preventing acid demineralization. But those products in the market have not been processed hygienically so contaminated by bacteria, fungi and mold that have the maximum microbial contamination. Pasteurization radiation dose up to 10 kGy can reduce the total plate count bacteria of Curcuma mangga Val and Zijp until 3 – 4 logarithms. In terms of microbiology, pasteurization radiation very advantageous, but the retrieval of information about the activities of irradiated C. mangga are still limited. This study was conducted to determine the effect of pasteurization radiation doses of 0, 5 and 10 kGy for curcuminoid compound and antidiabetic activity of C.mangga in in vitro (inhibition enzyme α-glucosidase) and in vivo (in mice induced alloxan). C. mangga was extracted by maceration eith ethanol. In vitro test result showed that iradiation 5 kGy could increased the curcuminoid compound while C. mangga 0, 5 and 10 kGy did not have inhibitory activity of the enzyme α-glucosidase, with value of percentage of α-glucosidase inhibition consecutive 9.89 ± 0.68; 8.44 ± 1.21 and 16.95 ± 2.05 at a concentration of 50 ppm. In vivo result test, on the 15th day of C. mangga 0, 5 and 10 kGy reduced blood glucose levels relative of 46.53% ± 18.41; 54.66 % ± 19.94 and 48.13 % ± 36.02.

Indonesia as tropical country has high temperature and humidity that can trigger bacteria and yeast/mold growth. Gamma irradiation has been used by the industries to reduce or eliminate total bacteria and yeast/mold on herbal medicine. The aim of this research was to get the dose of gamma radiation for pasteurization in C. asiatica herb so that it complies with standard pharmacopea. Other benefit from this research is supporting the regulation of the pasteurization radiation use for herb medicines. In this study, we applied doses of the gamma radiation were 0, 5, and 10 kGy. The asiaticoside quality test of C. asiatica irradiated were 1,18%; 0,98% and 1,58% while according to the Standard Herbal Pharmacopea, C. asiatica should contains ≥ 0,07 % Asiaticoside. The total microbiological tests were (1.9±1.7) x 105 cfu g-1, (8.0±2.8) x 101 cfu g-1 and < 10 cfu g-1. So that the irradiation dose up to 10 kGy can be use for decontamination of bacteria. The antioxidant capacity of irradiated C. asiatica herbs were 288.91 μg/ml, 176.64 μg/ml and 173.43 μg/ml. For studying wound healing test, extracts of C. asiatica 0, 5 and 10 kGy were found to be effective in healing wound than control group which were evident by recovery wound on C. asiatica group faster than control group. Infusion of C. asiatica non-irradiated and irradiated 5 kGy showed as high immunomodulators while extract of C. asiatica irradiated 10 kGy showed as medium immunomodulators. Results of the tests provided baseline information and established safe dose of using gamma radiation as pasteurization technique.
