The reaction was catalyzed in the presence of silica-H2SO4(+6). The derivatives computational ADMET profile was calculated. The study shows that most active derivatives have optimal logP, higher anti-inflammatory activity score, and poor metabolism at the sight of P450-3A4 and 2D6. The entire series of derivatives were further evaluated for anti-tubercular (against Mycobacterium tuberculosis H37Rv (Resistant strain)) and anti-inflammatory activity (in-vivo assay using Wistar rat). The result showed that derivatives 4c, 4h, and 4m have significant potency against tested M. tuberculosis. However, derivatives 4i and 4j found significantly active against inflammation. In vitro COX inhibition assay also supported the result in favor of selectivity and efficacy of derivatives.

The recognition of the plant growth hormone, heteroauxin, the significant amino acids, tryptamine and tryptophan and anti-inflammatory drug, indomethacine are the imperative derivatives of indole which have added stimulus to this review work. Isatin (1H-indole-2,3-dione), an indole derivative of plant origin. Although it is a naturally occurring compound, but was synthesized by Erdmann and Laurent in 1840 before it was found in nature. Isatin is a versatile precursor for many biologically active molecules and its diversified nature makes it a versatile substrate for further modifications. It is concerned in many pharmacological activities like anti-malarial, antiviral, anti-allergic, antimicrobial etc; isatin and its derivatives have been also found to demonstrate promising outcomes against various cancer cell lines. This review provides a brief overview on the recent advances and future perspectives on chemistry and biological aspects of isatin and its derivatives reported in the recent past.

In vitro assay result encouraged to further move towards in ovo anticancer evaluation using chick embryo. The series of QCT derivatives showed higher anticancer and antiangiogenic activity against HeLa and MCF-7 cell lines. In the series, synthetic molecule 8d, 8l, and 8m displayed significant activity. Further, these results substantiated by docking study on VGFR2. SAR study concluded that the potency of drugs depends on the nature of aliphatic substitution and the heterocyclic ring system.

Dried leaves and bark powder material was used for determination of ash value, extractive value, and phytochemical constituents. fifteen compounds from the whole plant of Callicarpa macrophylla were subjected to molecular properties prediction and drug-likeness by Lipinski rule of five & Molinspiration software. Phytochemical screening proved the presence of chemical constituent like tannins, alkaloids, proteins, starch, flavanoids, and glycoside. 12 compounds of the plant fulfill the requirements of Drug likeness were taken for biological activity calculation with the help of Molinspiration software and compared with standard drug Silibinin. Calliterpenone monoacetate almost fulfills the Lipinski rule of five and showed good bioactivity score than Silibinin. Out of 15 compounds Calliterpenone, Ursolic acid, Crategolic acid, β-Sitosterol, Betulinic acid, Luteolin-7-O-glucuronide, Apigenin-7-O-glucuronide and Calliterpenone monoacetate has good bioactivity score as compared to Silibinin which is potent hepatoprotective drug. So these compounds can be considered as lead compounds with hepatoprotective activity from Callicarpa macrophylla.
