During a six-year period (from January 2009 to December 2014), specimens collected from 344 cases... more During a six-year period (from January 2009 to December 2014), specimens collected from 344 cases of suspected organophosphorus and carbamate pesticide poisonings in wildlife, including birds, were submitted to the Toxicology Laboratory (ARC-OVI) for analysis. A positive diagnosis was made in 135 (39%) of these cases. The majority of cases were from birds, which included Cape vultures (Gyps coprotheres) and African white-backed vultures (Gyps africanus) and bateleur eagles (Terathopius ecaudatus). In one incident 49 vultures were killed when a farmer intentionally laced carcasses with carbofuran in an attempt to control jackal predation. There were 22 incidents of poisoning in helmeted guineafowl (Numida meleagris). On nine different occasions blue cranes (Anthropoides paradiseus) were poisoned, in one incident 14 birds were reported to have been killed. Over the period of investigation, there were 20 cases of poisoning involving mammalian species, the majority being vervet monkeys (Chlorocebus pygerythrus). The carbamate pesticides were responsible for 57 incidents of poisoning. Aldicarb, carbofuran and methomyl were detected in 26, 18 and 12 cases respectively. The majority of organophosphorus pesticide poisonings were caused by diazinon (n = 19), monocrotophos (n = 13) and methamidophos (n = 10).
In the present study, the developmental (including fertility) and endocrine-disruptive effects in... more In the present study, the developmental (including fertility) and endocrine-disruptive effects in relation to chemical burden in male and female Nile crocodiles (Crocodylus niloticus), from a commercial crocodile farm in the Brits district, South Africa, exposed to various anthropogenic aquatic contaminants from the natural environment was investigated. Hepatic transcript levels for vitellogenin (Vtg), zona pellucida (ZP) and ERα (also in gonads) were analyzed using real-time PCR. Plasma estradiol-17β (E2), testosterone (T) and 11-ketotestosterone (11-KT) were analyzed using enzyme immunoassay. Gonadal aromatase and hepatic testosterone metabolism (6β-hydroxylase (6β-OHase)) were analyzed using biochemical methods. Overall, there is high and abnormal number (%) of infertile and banded eggs during the studied reproductive seasons, showing up to 57 and 34% of infertile eggs in the 2009/2010 and 2013/2014 seasons, respectively. In addition, the percentage of banded eggs ranged between 10 and 19% during the period of 2009-2014 seasons. While hepatic ERα, Vtg, ZP mRNA and testosterone 6β-OHase, were equally expressed in female and male crocodiles, gonadal ERα mRNA and aromatase activity were significantly higher in females compared to male crocodiles. On the other hand, plasma T and 11-KT levels were significantly higher in males, compared to female crocodiles. Principal component analysis (PCA) produced significant grouping that revealed correlative relationships between reproductive/endocrine-disruptive variables and liver contaminant burden, that further relates to measured contaminants in the natural environment. The overall results suggest that these captive pre-slaughter farm crocodiles exhibited responses to anthropogenic aquatic contaminants with potentially relevant consequences on key reproductive and endocrine pathways and these responses may be established as relevant species endocrine disruptor biomarkers of exposure and effects in this threatened species.

The cattle tested were heifers and cows over 18 months of age (n = 3,374) which grazed on communal tribal lands under extensive ranching conditions. Fifty (1.48%) animals were tested positive with a CFT titre of 30iu ml-1 or higher.
An epidemiological investigation into an "illthrift&... more An epidemiological investigation into an "illthrift" problem occurring on a dairy farm adjacent to an alloy-processing unit, established that the probable cause of the problem was chronic vanadium poisoning. The disease manifested initially in animals 4-18 months old which showed emaciation, chronic diarrhoea and, in some cases, rhinitis, conjunctivitis and recumbency followed by death. Post-mortem (n = 17) and clinical-pathology findings (n = 60) indicated that malabsorption and immunosuppression were the basis of the pathogenesis in affected animals. Eight months after the commencement of the investigation, adult cows began showing evidence of emaciation, reduced milk production and an apparent increase in the number of abortions, stillbirths and dystocias. Over a 2-year period, 134 surface-soil samples, 134 subsoil samples and 134 grass samples from the farm were analysed for various fractions of vanadium. Thirty-four of each of these samples were collected at different time intervals (autumn 1990, summer 1991 and winter 1991) and at varying distances and directions from the processing unit, in order to gauge the magnitude of the problem, and the distribution pattern of vanadium, and to identify possible seasonal trends. The remaining 100 of each of these samples were taken at 100-m intervals over an area of approximately 1,140,000 m2 directly adjacent to the processing unit so that concentration isolines for vanadium could be drawn and the source more conclusively identified. The levels of vanadium were found to be highest closest to the mine, and surface-soil levels were consistently higher than subsoil levels, suggesting aerial pollution, which was confirmed by air sampling.(ABSTRACT TRUNCATED AT 250 WORDS)
