Common envelope (CE) is an important phase in the evolution of many binary systems. Giant star / compact object interaction in binaries plays an important role in high-energy phenomena as well as in the evolution of their environment. Material accreted onto the compact object may form a disk and power a jet. We study analytically and through numerical simulations the interaction between the jet and the CE. We determine the conditions under which accreting material quenches the jet or allows it to propagate successfully, in which case even the envelope may be ejected. Close to the stellar core of the companion the compact object accretes at a larger rate. A jet launched from this region needs a larger accretion-to-ejection efficiency to successfully propagate through the CE compared to a jet launched far from the stellar core, and is strongly deflected by the orbital motion. The energy deposited by the jet may be larger than the binding energy of the envelope. The jet can, thus, play a fundamental role in the CE evolution. We find that the energy dissipation of the jet into the CE may stop accretion onto the disk. We expect the jet to be intermittent, unless the energy deposited is large enough to lead to the unbinding of the outer layers of the CE. Given that the energy and duration of the jet are similar to those of ultra-long GRBs, we suggest this as a new channel to produce these events.

The BeppoSAX Gamma Ray Burst Monitor (GRBM) is triggered any time a statistically significant counting excess is simultaneously revealed by at least two of its four independent detectors. Several spurious effects, including highly ionizing particles crossing two detectors, are recorded as onboard triggers. In fact, a large number of false triggers is detected, in the order of 10/day. A software code, based on an heuristic algorithm, was written to discriminate between real and false triggers. We present the results of the analysis on an homogeneous sample of GRBM triggers, thus providing an estimate of the efficiency of the GRB detection system consisting of the GRBM and the software. THE GAMMA RAY BURST MONITOR The GRBM [1,3,2,4] is a gamma-ray detection system onboard the Bep-poSAX satellite. It is the secondary function of the anticoincidence shields of the PDS experiment. It is composed of four ∼1100 cm 2 slabs of CsI(Na) scintillators operating in the 40-700 keV range. Each detector provides time series of the detected counts in the above energy range, with 1 s resolution as a continuous housekeeping and with better than 8 ms resolution upon trigger. An oboard trigger is active whenever a statistically significant counting excess is simultaneously detected in at least two of the four detectors. The human metastasis-associated gene (MTA1) is overexpressed in cell lines and tissues representing metastatic tumors. Here we report cloning of the mouse Mta1 as well as a novel structurally related mouse gene, Mta3. The mouse Mta1 protein shares 94 and 59% homology to the human MTA1 and mouse Mta3 proteins, respectively. Northern blotting analysis using an Mta1 cDNA probe revealed a prevalent 3 kb hybridization signal in all mouse tissues except the skeletal muscle while a smaller ∼1.0 kb mRNA product was also detected in the heart. Mta3 transcripts (∼2 kb) were detected in most tissues with an additional ∼6.2 kb signal detected in the brain. In vitro transcription/translation of the full-length Mta1 and Mta3 cDNAs generated products of the expected molecular masses, i.e. 80 and 60 kDa, respectively. To assess subcellular localization, green fluorescence protein (GFP)-tagged expression constructs of Mta1 and Mta3 and various deletion constructs of GFP-Mta1 were transiently expressed in Balb/MK keratinocytes. GFP-Mta1 was found exclusively in the nucleus while GFP-Mta3 was present in both the nucleus and cytoplasm. Compared to Mta3, the carboxy terminal end of Mta1 contains an additional nuclear localization signal (NLS) and a proline-rich Src homology 3 (SH3) ligand. The results of transient expression experiments of various Mta1 fragments containing these domains in different combinations indicated that nuclear localization of Mta1 depended on the presence of at least one NLS and one SH3 binding site. These SH3 ligands appeared to be functional as they facilitated interaction with the adaptor protein, Grb2, and the Src-family tyrosine kinase, Fyn.

Actinomycin D was revealed as an inhibitor of Shc/Grb2 interaction in cell lines from our recent study. Shc and Grb2 proteins are important molecules in Ras signaling pathways leading to cellular differentiation and proliferation, which require dramatic morphological changes. It was detected by transmission electron microscopy that actinomycin D induced significant changes in cellular ultrastructures of B104-1-1 cells and confirmed that the changes were due to inhibition of Shc/Grb2 interaction by actinomycin D rather than its inhibitory effect on transcription. Because actinomycin D was dispersed mainly in cytoplasm and Shc peptide (synthetic 13 amino acid tyrosine phosphorylated polypeptide) successfully displaced actinomycin D binding to its cellular targets while the other polypeptide from PDGF receptor could not. We examined the effect of actinomycin D on growth of B104-1-1 tumor xenografted in nude mice. Tumor growth was inhibited in vivo after treatment with this inhibitor. Efficacy was correlated with a reduction in the levels of Shc/Grb2 binding in excised tumors. These results suggest that actinomycin D inhibited Shc/Grb2 interaction in B104-1-1 tumor xenografted in nude mice.

Ligand stimulation of the platelet-derived growth factor (PDGF) beta-receptor leads to activation of its intrinsic tyrosine kinase and autophosphorylation of the intracellular part of the receptor. The autophosphorylated tyrosine residues mediate interactions with downstream signal transduction molecules and thereby initiate different signalling pathways. Huntington's disease (HD) is caused due to expansion of CAG repeats in the gene huntingtin (Htt). Adaptor protein Grb2, involved in Ras-MAP kinase pathway, is a known interactor of Htt. Mutant Htt-Grb2 interaction reduces Ras-MAPK signaling in HD models. In normal cells Grb2 forms Grb2-Sos1-Gab1 complex through its N-SH3 and C-SH3 domains respectively, essential for sustained activation of Ras. We found that C-SH3 of Grb2 mediates the interaction with mutant Htt and this interaction being stronger could replace Gab1, with mutant Htt becoming the preferred partner. This would have immense effect on downstream signaling events.
