This study was conducted to assess the post thaw quality of Kundhi buffalo semen diluted in tris based diluents. After collection the ejaculates were pooled and checked for color, volume, pH, mass activity, progressive linear motility% (PLM), and intactness of cell membrane. The semen qualifying these tests was divided into four aliquots A, B, C and D, diluted with tris based diluents; the last three diluents containing sugar types glucose, fructose and lactose respectively. Aliquot A was without sugar and severed as control. These were frozen, stored for 24 hrs, thawed and incubated at 35 0 C for 5 hrs. The incubated semen aliquots were examined for PLM and membrane integrity of sperm cells with phase-contrast microscopy and HOST procedures. It was observed that all the ejaculates were creamy white in color. Seminal plasma transaminases (GOT and GPT) were assessed in diluted (Tris-egg yolk-citric acid-fructoseglycerol extender 1:4) and thawed (40 degree C for 15 seconds) semen of Sirohi bucks (control group). Diluted, cooled (from 25 degree C up to 5 degree C) and
equilibrated (at 5 degree C for 2 hours) semen straws were frozen @ 15, 20, 25 and 30 degree C/minute for M1, M2, M3 and M4 groups respectively. These frozen straws were stored separately in LN2. Effect of freezing rates on leakage of tranferases was studied by assessing GOT and GPT in seminal plasma of thawed semen from M1, M2, M3 and M4 groups. The Mean + S.E. (C.V.%) values of transferases (GOT : GPT) in M1, M2, M3, M4 and control group were 313.00 + 8.40 (6.58) : 20.5 + 0.43 (5.12), 272.5 + 6.68 (6.01) :
16.33 + 1.02 (15.33), 255.00 + 10.1 (9.7) : 14.5 + 0.67 (11.33), 292.0 + 9.67 (8.09) : 19.67 + 0.70 (8.99) and 147.67 + 10.96 (18.17) ; 9.5 + 0.62 (15.96) units per 0.92 x 109 spermatozoa respectively. Least square analysis of variance revealed highly significant (P < 0.01) rise in the seminal plasma GOT enzyme levels in frozen thawed semen as compared to that in fresh diluted, cooled and equilibrated semen. Among the freezing mode groups lowest GOT and GPT values in seminal plasma was observed in M3 followed by M2, M4 and M1 groups in increasing order.

The  study  was  conducted  to  determine  the  effect  of  subacute
chlorpyrifos  exposure  on  some  hormonal  and  semen  characteristics
and,  the  ameliorative  effects  of  melatonin.  Twenty  four  adult  male
Wistar rats divided into 4 groups of 6 animals eachwere used for the
study. Rats in group I (S/oil) received soya oil (2ml/kg) while those
in group II (Mel) were given melatonin (0.5 mg/kg).Rats in group III
(CPF)  were  exposed  to  CPF  only  (8.5  mg/kg  ~  1/10

while rats in group IV (Mel + CPF) were pretreated  with melatonin
(0.5 mg/kg) and then exposed to CPF (8.5 mg/kg), 10minutes later.
The regimens were administered by gavage once dailyfor a period of
28 days. At the end of the study period, the rats were sacrificed, and
sera obtained from the blood samples were analyzed for testosterone,
follicle  stimulating  hormone  (FSH)  and  luteinizing  hormone  (LH).
Testicular  weight,  testicular  acetylcholinesterase  (AChE)  activity,
epididymal sperm count and sperm motility were alsoevaluated. The
results showed that pretreatment with melatonin ameliorated the CPF
induced  alterations  in  epididymal  sperm  count,  sperm  motility,
testicular  weight  and  levels  of  testosterone,  FSH  and  LH.  In
conclusion,  subacute  CPF-induced  alterations  in  sex hormones  and
semen characteristics were ameliorated by melatoninprobably due to
its antioxidant property.
