Artificial upwelling of nutrient-rich waters and the corresponding boost in primary productivity harbor the potential to enhance marine fishery yields and strengthen the biological pump for sequestration of atmospheric CO2. There is increasing urgency to understand this technology as a “ocean-based solution” for counteracting two major challenges of the 21st century—climate change and overfishing. Yet, little is known about the actual efficacy and/or possible side effects of artificial upwelling. We conducted a large-scale off-shore mesocosm study (∼44 m3) in the oligotrophic waters of the Canary Islands to identify the community-level effects of artificial upwelling on a natural oligotrophic plankton community. Four upwelling intensities were simulated (approx. 1.5/3/5.7/10 μmol L–1 of nitrate plus phosphate and silicate) via two different upwelling modes (a singular deep-water pulse vs. recurring supply every 4 days) for 37 days. Here we present results on the response of net community production (NCP), metabolic balance and phytoplankton community composition (<250 μm). Higher upwelling intensities yielded higher cumulative NCP. Following upwelling onset, the phytoplankton community became dominated by diatoms in all treatments, but other taxa such as Coccolithophores increased later in the experiment. The magnitude of effects on the metabolic balance scaled with the amount of added deep water, leading to (i) a balanced to net-heterotrophic system in the singular and (ii) a net-autotrophic system in the recurring upwelling treatments. Accordingly, the mode in which nutrients are supplied to an oligotrophic system plays a crucial role in the ecosystem response, with recurring upwelling leading to higher long-term positive NCP than singular upwelling. These results highlight the importance of empirically measured local responses to upwelling such as community structure and metabolism, with major implications for the potential employment of artificial upwelling as an ocean-based solution to generate (primary) production.

Obesity and ageing place a tremendous strain on the global healthcare system. Age-related sarcopenia is characterized by decreased muscular strength, decreased muscle quantity, quality, and decreased functional performance. Sarcopenic obesity (SO) is a condition that combines sarcopenia and obesity and has a substantial influence on the older adults’ health. Because of the complicated pathophysiology, there are disagreements and challenges in identifying and diagnosing SO. Recently, it has become clear that dysbiosis may play a role in the onset and progression of sarcopenia and SO. Skeletal muscle secretes myokines during contraction, which play an important role in controlling muscle growth, function, and metabolic balance. Myokine dysfunction can cause and aggravate obesity, sarcopenia, and SO. The only ways to prevent and slow the progression of sarcopenia, particularly sarcopenic obesity, are physical activity and correct nutritional support. While exercise cannot completely prevent sarcopenia and age-related loss in muscular function, it can certainly delay development and slow down the rate of sarcopenia. The purpose of this review was to discuss potential pathways to muscle deterioration in obese individuals. We also want to present the current understanding of the role of various factors, including microbiota and myokines, in the process of sarcopenia and SO.

Intraoperative stress is common to patients undergoing carotid endarterectomy (CEA); thus, impaired oxygen and metabolic balance may appear. In this study, we aimed to identify new markers of intraoperative cerebral ischemia, with predictive value on postoperative complications during CEA, performed in regional anesthesia. A total of 54 patients with significant carotid stenosis were recruited and submitted to CEA. Jugular and arterial blood samples were taken four times during operation, to measure the jugulo-arterial carbon dioxide partial pressure difference (P(j-a)CO2), and cortisol, S100B, L-arginine, and lactate levels. A positive correlation was found between preoperative cortisol levels and all S100B concentrations. In addition, they are positively correlated with P(j-a)CO2 values. Conversely, postoperative cortisol inversely correlates with P(j-a)CO2 and postoperative S100B values. A negative correlation was observed between maximum systolic and pulse pressures and P(j-a)CO2 after carotid clamp and before the release of clamp. Our data suggest that preoperative cortisol, S100B, L-arginine reflect patients’ frailty, while these parameters postoperatively are influenced by intraoperative stress and injury. As a novelty, P(j-a)CO2 might be an emerging indicator of cerebral blood flow during CEA.

Abstract. Although atmospheric dust fluxes from arid as well as human-impacted areas represent a significant source of nutrients to surface waters of the Mediterranean Sea, studies focusing on the evolution of the metabolic balance of the plankton community following a dust deposition event are scarce, and none were conducted in the context of projected future levels of temperature and pH. Moreover, most of the experiments took place in coastal areas. In the framework of the PEACETIME project, three dust-addition perturbation experiments were conducted in 300 L tanks filled with surface seawater collected in the Tyrrhenian Sea (TYR), Ionian Sea (ION) and Algerian basin (FAST) on board the R/V Pourquoi Pas? in late spring 2017. For each experiment, six tanks were used to follow the evolution of chemical and biological stocks, biological activity and particle export. The impacts of a dust deposition event simulated at their surface were followed under present environmental conditions and under a realistic climate change scenario for 2100 (ca. +3 ∘C and −0.3 pH units). The tested waters were all typical of stratified oligotrophic conditions encountered in the open Mediterranean Sea at this period of the year, with low rates of primary production and a metabolic balance towards net heterotrophy. The release of nutrients after dust seeding had very contrasting impacts on the metabolism of the communities, depending on the station investigated. At TYR, the release of new nutrients was followed by a negative impact on both particulate and dissolved 14C-based production rates, while heterotrophic bacterial production strongly increased, driving the community to an even more heterotrophic state. At ION and FAST, the efficiency of organic matter export due to mineral/organic aggregation processes was lower than at TYR and likely related to a lower quantity/age of dissolved organic matter present at the time of the seeding and a smaller production of DOM following dust addition. This was also reflected by lower initial concentrations in transparent exopolymer particles (TEPs) and a lower increase in TEP concentrations following the dust addition, as compared to TYR. At ION and FAST, both the autotrophic and heterotrophic community benefited from dust addition, with a stronger relative increase in autotrophic processes observed at FAST. Our study showed that the potential positive impact of dust deposition on primary production depends on the initial composition and metabolic state of the investigated community. This impact is constrained by the quantity of nutrients added in order to sustain both the fast response of heterotrophic prokaryotes and the delayed one of primary producers. Finally, under future environmental conditions, heterotrophic metabolism was overall more impacted than primary production, with the consequence that all integrated net community production rates decreased with no detectable impact on carbon export, therefore reducing the capacity of surface waters to sequester anthropogenic CO2.

Appropriate concentration of androgens is necessary for general men’s health, especially for development and maintain reproductive and sexual function, as well as metabolic balance. Depending on the function of the testicles, production of androgens and sperm is also conditioned on hypothalamus-pituitary axis, metabolic status and age of the individual. Considering that, hypogonadism of primary, secondary, functional or late-onset origin may be recognised. As it concerns susceptible matter of sexual and reproductive health, it requires special interest of differently clinicians, i.e. endocrinologists, andrologists or urologists.

Glutamate is the main excitatory neurotransmitter in the brain, and its excitatory neurotoxicity is closely related to the occurrence and development of Alzheimer’s disease. However, increasing evidence shows that in the process of Alzheimer’s disease, glutamate is not only limited to its excitotoxicity as a neurotransmitter but also related to the disorder of its metabolic balance. The balance of glutamate metabolism in the brain is an important determinant of central nervous system health, and the maintenance of this balance is closely related to glutamate uptake, glutamate circulation, intracellular mitochondrial transport, and mitochondrial metabolism. In this paper, we intend to elaborate the key role of mitochondrial glutamate metabolism in the pathogenesis of Alzheimer’s disease and review glutamate metabolism in mitochondria as a potential target in the treatment of Alzheimer’s disease.

Solid sago waste is a potential source of producing renewable energy in the form of biogas. This study investigated the effects of solid sago waste particle size, biological pretreatment using a microbial consortium of lignocelluloses, pretreatment with NaOH, and the ratio between solid sago waste and cow rumen based on the biogas production rate. Several variations of these conditions were used to achieve this. The anaerobic digestion process was conducted over two months at 30.42 °C ± 0.05 °C, and the biogas production rate was measured every two days. The 1:1 ratio showed better results compared to the 2:1, because it allows the bacteria to achieve metabolic balance. The highest cumulative biogas production (27.91 mL/g TS) was generated when the sago waste underwent milling (±1 mm), pretreatment with 4% NaOH g/g TS, and treatment with microbial consortium 5% v/v at a 1:1 ratio of solid sago waste to the rumen.

Evidence indicates that patients with Alzheimer’s dementia (AD) show signs of copper (Cu) dyshomeostasis. This study aimed at evaluating the potential of Cu dysregulation as an AD susceptibility factor. We performed a meta-analysis of 56 studies investigating Cu biomarkers in brain specimens (pooled total of 182 AD and 166 healthy controls, HC) and in serum/plasma (pooled total of 2929 AD and 3547 HC). We also completed a replication study of serum Cu biomarkers in 97 AD patients and 70 HC screened for rs732774 and rs1061472 ATP7B, the gene encoding for the Cu transporter ATPase7B. Our meta-analysis showed decreased Cu in AD brain specimens, increased Cu and nonbound ceruloplasmin (Non-Cp) Cu in serum/plasma samples, and unchanged ceruloplasmin. Serum/plasma Cu excess was associated with a three to fourfold increase in the risk of having AD. Our replication study confirmed meta-analysis results and showed that carriers of the ATP7B AG haplotype were significantly more frequent in the AD group. Overall, our study shows that AD patients fail to maintain a Cu metabolic balance and reveals the presence of a percentage of AD patients carrying ATP7B AG haplotype and presenting Non-Cp Cu excess, which suggest that a subset of AD subjects is prone to Cu imbalance. This AD subtype can be the target of precision medicine-based strategies tackling Cu dysregulation.

Accumulation of visceral fat causes a lipotoxic effect on the hypothalamus. Hypothalamic inflammation closes a vicious circle, aggravating the disturbance of neuroendocrine mechanisms to regulate metabolic balance, including coordination of energy consumption (during basic metabolism, physical activity, thermogenesis) and proper feeding behavior, the study group included 62 individuals (women) with obesity. Each patient met the diagnostic criteria for idiopathic hypothalamic syndrome – the neuroendocrine form with obesity.The current study shows that obesity of hypothalamic origin is associated with a dyslipidemia that progresses simultaneously with the increase of the body mass index.
