The spreadsheet allows the user to perform various common HPLC calculations such as column equilibration time, void volume, pressure conversion, mobile phase quantity, and method development parameters.

The spreadsheet allows calculation of column equilibration time, void volume, pressure conversion between units, mobile phase buffer strength, mobile phase quantity for isocratic and gradient methods, and suggestions for method development parameters based on chromatographic data.

To calculate the void volume, the internal diameter and length of the column as well as the flow rate must be input. The void volume is then calculated based on these parameters.

HPLC CALCULATOR
Column Equilibration
Insert your column dimensions, as indicated below, and the flow
rate required for your method to obtain recommended
equilibration times. Then click on Enter.
INPUTS
Internal diameter of column in mm
Length of column in cm
Flow rate of method in mL/min

4.6 mm
10 cm
1.5 mL/min

OUTPUTS
Equilibration time for:
10 column volumes
20 column volumes
30 column volumes

7.8 min
15.5 min
23.3 min

10 column volumes is recommended for re-equilibration after a

gradient injection has been run, 10 to 20 column volumes are
recommended for initial equilibration when the column is installed
on the system, and 30 column volumes is a recommended starting
point for methods which require more equilibration, such as ionpairing.

HPLC CALCULATOR
Mobile Phase Quantity - Isocratic Methods
To calculate the quantity of mobile phase required for an isocratic
HPLC method simply input the flow rate, runtime and number of
injections in your analysis below. An overage of mobile phase is
required for pre-analysis equilibration, for contingencies and because
the complete volume cannot be delivered from the mobile phase
reservoir. An overage of 15% is included but you can specify the
overage that you want as a percentage, or simply add a suitable
amount, based on your own experience. The value calculated for the
'Amount to prepare' should be rounded to a suitable amount.
