Chemistry Lab #1

Title: States of Matter

Aim: To investigate osmosis in living tissue

Materials/Apparatus:
Potato strips
Distilled water
Common salt
Beakers
Spatula
Labels
Stopwatch
Razor blade
Ruler
Balance
Petri dish

Procedure:
1. Using a blade produce 9 potato strips. Ensure that the skin has
been removed from all and that they are each 6cm long.
2. Measure 100cm3 of distilled water and place into the three
identical beakers, labeled A, B and C.
3. Using a petri dish weigh 1g of common salt and add to the beaker
labeled B. Repeat this step weighing 5g of salt instead and add to
beaker C.
4. Place three of the strips of potato tissue into each beaker. Note the
time.
5. Leave the strips of tissue in the liquid for at least thirty minutes.
6. After the time has elapsed, remove the strips from the liquids.
Feel and record their texture.
7. Measure the length of each strip. Record your observations in the
table below.

Observations:
Beaker A B C
Initial
length /cm
Final

Texture of
strips after
30 mins

Analysis and Interpretation:

1. Explain what you observed; account for the differences in texture
2. What is the semi-permeable membrane in this experiment?
3. What do you think different solutions were used?
Distilled Salt solution Salt solution
water 1g/100mL 5g/100mL

4. What is osmosis?

Conclusion
Limitations/Sources of Error/Variables:

Explanation
 Osmosis is a special type of diffusion that applies to water and
other solvents; it is the movement of water from a level of high
concentration to low concentration or across a water potential
gradient.
Note: Water potential refers to the potential energy of water in
relationship to water in pure conditions. This potential refers to
the ability of water to move from one region to another due to
gravity, osmosis, pressure, or any other type of action.
 A perfect example of a solvent with a high water potential/high
concentration of water is pure water. Sea salt water, on the other
hand is an example of one with a low concentration of water
molecules as we have NaCl in its composition. (Ref. mixtures)
 If a partially permeable membrane separating two water samples
there will be a net movement of water from the place with a high
conc. to low.
 In the lab the cell membrane of the potato strips acted as the semi
permeable membrane separating the water in the living tissue
from the water in the beaker.
 In the living tissue the water concentration is greater than it is
outside and so there is a net movement of water from the tissue
out making it flaccid and lose length as it shrivels.
 In the pure water, the water moves into the potato strips making
it turgid and increase length.
 The different solutions have varying water potential. Varying
concentrations of water.
Resoures: Message Jeff, Nazelle and Britny or go visit when you get the
chance. You need cork borers (can be used on site and returned) and
distilled water.
Use the one period of the double period to explain, and the other to do
lab.
