Murashige and Skoog Medium (MS) was originally VITAMINS
formulated by Murashige and Skoog in 1962 to optimize myo-Inositol 100.000
tobacco callus bioassay system for facilitating the study Nicotinic acid (free acid) 1.000
of cytokinins. Since then, it is widely used for micro Pyridoxine HCl 1.000
propagation, organ culture, callus culture and suspension Thiamine hydrochloride 10.000
culture. The formulation is a nutrient blend of inorganic Total(gms/litre) 4.4
salts and vitamins.

Murashige and Skoog Medium (MS) provides all Material required but not provided :
essential macroelements and microelements. Potassium • Autoclaved distilled water
dihydrogen phosphate serves as a source of • Gelling agents like Agar (PCT0901) or CleriGel (PCT0903)
phosphate. Microelements like Boron, Manganese, • Plant growth regulators
Molybdenum, Copper, Iron and Zinc play vital • 1N NaOH/HCl
role in plant metabolism. Boron plays a key role • Sucrose (PCT0607)
in carbohydrate metabolism. Thiamine, pyridoxine,
nicotinic acid act as enzymatic cofactors in universal Precautions :
pathways including glycolysis and TCA cycle along
• Ensure appropriate pH of the medium before addition of
with primary and secondary metabolism in the plants.
gelling agent as acidic pH will lead to decreased gelation
resulting in semi solid flowing gel while alkaline pH will lead
The product is plant tissue culture tested but it is the sole to formation of hardened gel.
responsibility of the user to ensure the suitability of the • Use of Distilled water / Tissue culture grade water is
medium for individual species. recommended for media preparation as tap water or lower
grade water may lead to salt precipitation and improper
Composition : gelation.
• Avoid preparation of concentrated solutions, as it will lead
Ingredients mg/L to precipitation of salts.
MACROELEMENTS
Ammonium nitrate 1650.000
Calcium chloride 332.200
Directions :
Magnesium sulphate 180.690 • Reconstitute medium by adding required quantity of
Potassium nitrate 1900.000 powder in two-third of total volume with constant, gentle
Potassium phosphate monobasic 170.000 stirring till the medium gets completely dissolved.
MICROELEMENTS • Add heat stable supplements prior to autoclaving.
Boric acid 6.200 • Make up the final volume with distilled water.
Cobalt chloride hexahydrate 0.025 • Adjust the pH of the medium to 5.75 ± 0.5 using 1N NaOH/
Copper sulphate pentahydrate 0.025 HCl.
EDTA disodium salt dihydrate 37.300 • Add gelling agent and heat the medium to boiling till
Ferrous sulphate heptahydrate 27.800 complete dissolution of gelling agent.
Manganese sulphate monohydrate 16.900 • Sterilize the medium by autoclaving at 15 lbs and 121°C
Molybdic acid (sodium salt) 0.213 for 15 min.

Please refer disclaimer overleaf

• Cool the autoclaved medium to about 45°C before adding
heat labile supplements.
• Aseptically dispense the desired amount of medium under
a laminar airflow unit in sterile culture vessels.

Plant Tissue Culture Test
The growth promoting properties of medium is assessed
by providing plant cultures with relative humidity of about
60%±2%,temperature 22ºC±2ºC and photoperiod of about
16:8.The plant species showed actively growing callus and
shoots with no structural, necrotic and toxic deformity.
