Regarded as Nature’s catalysts,
enzymes are:

highly selective
stereospecific
perform complicated reactions:
neutral pH
ambient temperatures
atmospheric pressure
aqueous environment.

A key aspect of any process is the

capability to re-use the enzyme.
Original product Enzyme reaction

New product Impurities Original product

Crystallisation Recycle
Advantages
Easy separation from reaction mixture
Re-use of enzyme for many reaction cycles
Lower cost of enzyme
Selective reactions
Increased stability of enzyme
Known engineering technology

Disadvantage
Lower activity than free enzyme
General observations

Macroreticular structure
Weakly ionic or non functionalized resins
Relatively hydrophilic surface : acrylic and phenolic
resins
Particle size important for kinetic reasons

Apart from the first point, there are exceptions to

each of the above.
Methods of fixation

Simple adsorption onto a buffered resin.

Adsorption onto a buffered resins followed by
treatment with glutaraldehyde.
Ionic bonding ( with H+ or OH- form resins).
Covalent bonding via -NH2 groups.
Via metals such as Fe, Ni, Al etc.
