Points to Consider for Aseptic Processing

P3: Personnel and Air Monitoring: How to David Hussong, PhD
Control the Most Important Variables
Aseptic Processing
• Has advantages over terminal sterilization
– One of them is Not sterility assurance
– Product stability is a significant advantage
• Requires manipulation of sterile components
to produce a sealed, sterile product
3
Aseptic Manipulation
• Manipulation – operating with or as if with
the hands or by mechanical means especially
in a skillful manner (from Webster)
• Aseptic ‐ free of or freed from pathogenic
microorganisms (from Webster)
• But wait, there’s more! 4

P3: Personnel and Air Monitoring: How to David Hussong, PhD
Control the Most Important Variables
Personnel
• Necessary for cleaning, disinfection and set‐up
• Operate the filling equipment
• Must be trained to work near critical areas
Operators – The Caveat
• The human factor
– Hard to control
– Necessary to perform
interventions
– Source of >99% microbial
contamination (Jim Agalloco)

Control the Most Important Variables
Controlling the Human Factor
• Authorized Personnel
Only (Qualification)
• Keep people out of
the cleanroom
(Automation)
7
Qualifications and Training
• What is the process to qualify personnel to
work in (or gain access to) the aseptic
processing area (Grade A/B)?
– Written training program
– Program successfully completed
– Records of training
8

Control the Most Important Variables
How to Control Personnel ‐ Access
• Control of access to the aseptic processing
area should be in place
– Procedures (understood from training and
supervision)
– Physical controls (doors and security provisions
such as access cards) 9
Access Control Record
• Should be maintained of all persons entering
and leaving the aseptic processing area
• Usually part of batch record
• Always review
10

Control the Most Important Variables
Environmental Monitoring Personnel
• May be QC or production personnel (QC may
reduce appearance of bias)
– random verification sampling or audits
• Trained and qualified in good sampling
practices
11
Monitoring Personnel ‐ Gloves
• “After an intervention where inappropriate
contact with product contact surfaces may have
occurred…
• “…as defined by internal procedures”
• “Upon exiting from the sterile area”
• “As deemed necessary…”

Control the Most Important Variables
Microbiological Personnel Monitoring
• Monitor as part of qualification
– Gowning qualification
– Process simulation
• Never sample personnel before they enter the
cleanroom
13
Personnel Qualification
• Initial
• Periodic (e.g., annual)
• After an exception
– Long absence
– Supervisory recommendation (e.g., observed
unacceptable practice, CAPA, failed media fill)
• Exceptional entry by nonqualified personnel (e.g., safety
issue) 14
7

Control the Most Important Variables
Supervision
• “… performed by individuals who are trained and
qualified
• “… degree of oversight needed may depend on
the level of physical separation
• Required for cleaning, maintenance, production
• Responsibility of the quality unit
PDA PtC Part 1 (2015) 15
Sterile Gown Usage
• Each entry needs a new gown
• Change if gown integrity is in doubt
• Have procedures documented
• Train personnel and document training
• Review

Control the Most Important Variables
Personnel in Aseptic Processing Area
• More is not better
• Establish how many are needed
– Classification and activity dependent
• Only necessary personnel should be present
• Design process for minimum personnel
17
Designing the Aseptic Process
• Goal is to minimize
personnel activity in critical
areas
• No personnel is ideal
– RABS
– Isolators
– Robotics
• Not always practical 18
9
P3: Personnel and Air Monitoring: How to David Hussong, PhD
Control the Most Important Variables
Personnel Hygienic Practices
• No jewelry
• Appropriate garments
under gowns
• Overall hygiene
• No open wounds
• No illnesses (flu) 19
Personnel & Contamination
• “It is useful to assume that the operator is always
contaminated while operating in the aseptic area.
If the procedures are viewed from this
perspective, those practices which are exposing
the product to contamination are more easily
identified.” Hank Avallone (1988)

Process Evolution
• We have come a long
way, and will continue.
