The study of biology often requires examination of structures/ organisms that need to be observed in their living
state. The following is a technique that provides a view of the structural details of a plant cell particularly that of an onion
(Allium cepa) skin.
I. OBJECTIVES
At the end of the activity, the students will be able to:
1. Examine and differentiate plant and animal cells using onion and cheek samples.
2. Locate and identify visible organelles in the examined cells.
3. Know about fresh specimen preparation techniques by watching a video.
4. Explain why such preparation is essential to biological studies.
II. MATERIALS
Virtual Microscope
Virtual prepared slides of the ff:
Unstained Onion Epidermal Cells
Iodine stained Onion Cells
Stained Cheek Cells
III. RESULTS
● Examine the slides listed above under the different objective lenses.
● Take a screenshot of the image seen under the microscope and paste it on the designated box below.
● Encircle or put an arrow and label to the observe organelles. CELL MEMBRANE, CYTOPLASM, and
NUCLEUS, CELL WALL, VACUOLE.
● How do the plant cells look like? How are they arranged?
Under the microscoped that we used, The plant cells are arranged in a
rectangular shapeds that are somewhat packed and forms a pattern.
Inside these rectangle shapes or the cell wall are the organelles.
● What cell parts are visible under the microscope that you used?
The cell parts that are visible under the microscope are The
nucleus,cytoplasm and the cell wall.

Since this is multicellular, the cells are grouped together. The black dot is
the nucleus of the cells.

Only can be seen under microscope is the Cell membrane and the nucleus
of the Cell.
IV. Analysis
● Create a Venn Diagram of plant and animal cells. Remember, things that they have in common go into the overlapping
area, things that are different go in the non-overlapping area.
