d-α-Tocopherol

Content d-α-Tocopherol contains not less than 40% of total tocopherol, of which
Not less than 50 % consists of d-α-tocopherol. Its commercial products may contain
edible fats and oils.
Description d-α-Tocopherol is a slightly yellow to red-brown, clear viscous liquid,
having a characteristic odor.
Identification To 50 mg of d-α-Tocopherol, add 10 ml of absolute ethanol to
dissolve, add 2 ml of nitric acid, and heat at about 75℃ for 15 minutes. A red to orange
color develops.

Purity (1) Specific rotation [α] D : not less than +24°

Weigh accurately an amount of d-α-Tocopherol, equivalent to about 100 mg of the


total tocopherols, transfer to a separating funnel, and dissolve in 50 ml of ether.
Dissolve 2 g of potassium ferricyanide in 20 ml of sodium hydroxide solution (1→125),
add to the separating funnel, and shake for 3 minutes. Wash the ether layer with four
50-ml portions of water and take the ether layer. Dehydrate the ether layer by adding
2 g of anhydrous sodium sulfate, and filter. Evaporate ether from the filtrate,
immediately dissolve the residue in 5 ml of isooctane, and measure the optical rotation.
Calculate the specific rotation of this solution, using the number of g of the total
tocopherols in 1 ml of the solution determined in the Assay.
(2) Acid value Not more than 5.0 (Fats and Related Substances Tests).
(3) Heavy metals Not more than 20 µg/g as Pb (1.0 g, Method 2, Control
solution Lead Standard Solution 2.0 ml).
(4) Arsenic Not more than 4.0 µg/g as As2O3 (0.50 g, Method 3, Apparatus B).
Assay Weigh accurately an amount of d-α-Tocopherol, equivalent to about 50 mg
of the total tocopherols, transfer to a brown measuring flask, dissolve in n-hexane to
make exactly 100 ml. Use this solution as the test solution.
Separately, weigh accurately an amount each of the Reference Standards of d-α
-tocopherol for assay, d-β-tocopherol for assay, d-γ-tocopherol for assay, and d-δ
-tocopherol for assay, transfer each sample into a brown flask, dissolve in n-hexane to
prepare each solution of about 0.050% w/v. Mix these solutions to prepare a mix
tocopherol solution (about 0.050% w/v) so that the mixture can contain a similar
composition to that for each tocopherol in the sample, calculated from the labeled
content. Use this solutions as the standard solution.
Using 20 µl each of the test solution and the standard solution, perform Liquid
Chromatography under the operating conditions given below. Measure the peak areas
of d- α -tocopherol, d- β -tocopherol, d- γ -tocopherol, and d- δ -tocopherol for both
solutions, and express as ACα, ACβ, ACγ, and ACδ for the test solution and ASα, AS
β, ASγ, and ASδ for the standard solution, respectively. Calculate the content of
total tocopherols by the formula, and calculate the percentage of d-α-tocopherol in the
total tocopherols

Where Sα: Concentration of d-α-tocopherol in the standard solution (mg/ml),


Sβ: Concentration of d-β-tocopherol in the standard solution (mg/ml),
Sγ: Concentration of d-γ-tocopherol in the standard solution (mg/ml),
Sδ: Concentration of d-δ-tocopherol in the standard solution (mg/ml).
Operating conditions
Detector: Detector for absorbances in ultraviolet region (detection wavelength:
292 nm).
Packing material of column: 5- to 10-µm silica gel for liquid chromatography.
Column: Stainless steel pipe 3−6 mm in internal diameter and 15−25 cm in
length.
Column temperature: Constant room temperature.
Mobile phase: A mixture of n-hexane−isopropyl alcohol (200 : 1).
Flow rate: Adjust so that the retention time for d-α-tocopherol can be about 5
minutes.
