Purpose To provide guidelines for environmental monitoring of product exposure and production areas.

Responsibility 3.1 3.2 Quality Control Manager is responsible to ensure that this SOP is implemented and strictly followed. All QC personnel must clearly understand and strictly follow this SOP .
4.0
Sampling Procedure The number of sampling points per given room shall be determined by the size of that room as well as its classification of cleanliness. 4.1 Surface Monitoring
4.1.1 Rodac Plates - Suitable for quantitative analysis for environmental procedures. For irregular surfaces, use swab test ( 4.1.2). a. Frequency of sampling shall be performed before, during and after aseptic fill and/or as baseline for GMP production. b. Procedure: Label Rodac plates with pertinent building number, sample point, and date. Remov e the lid of the Rodac and firmly press the surface of the agar on the area to be assayed. Hold for approximately five seconds, and replace lids. c. Incubate plate according to duration and temperature recommended by manufacturer. d. Follow decontamination procedure after sampling, to remove residual agar. e. Count incubated plates using an approved colony counter or other accepted means.

f. Record the date, time, colony forming units (CFU) in the Environmental Monitoring Logbook. 4.1.2 Swabs test - Suitable for quantitative analysis for environmental procedures : a. Frequency of sampling shall be performed before, during, and after aseptic fill and/or as baseline for GMP production. b. Procedure: Identify Tryptic Soy Broth ( TSB ) tubes with location and date of assay. Pull down sleeve of swab package halfway. Remove the swab ( DO NOT TOUCH SWAB TIP ) Dip swab in sterile, normal saline or water and rub over 4 square surface to be tested. Break off swab into 10 ml Trypticase Soy Broth tube. Include one uninoculated control TSB tube and one swab direct form package to TSB. Incubate all TSB tubes for 48 hours at 37C 1C. Record the date, time, results in the Environmental Monitoring Logbook. NOTE: The test is positive when the sample broth is turbid while the uninoculated control TSB and the TSB with the control swab remain clear. When results, are positive in baseline sampling, categorization and speciation must be performed using an automated ID system.

Air Monitoring 4.2.1 Viable Counts - Gel membrane filtration sampler is used when viable count is required for an area. One sampler shall be dedicated for the use within the aseptic operation room. a. Frequency of sampling shall be performed before, during and after aseptic fill and/or as baseline for GMP production. b. Record the date, time, viable count in the Environmental Monitoring Logbook. 4.2.2 Non-viable Particulate Count - Calibrated Laser Particle Counters used to determine the concentration (number of particles per cubic foot) of airborne particles of predetermined sizes at a given location. One particle counter shall be dedicated for use within the aseptic operation room. a. Frequency of sampling shall be performed during and after production in product exposure areas. b. Record the date, time, number of particles per cubic foot in the Environmental Monitoring Logbook. 4.2.3 Temperature and Humidity Counter - An accessory to the Laser Particle Counter - a temperature relative humidity probe will be used to monitor temperature and relative humidity as non-viable particulate counts are taken. a. Frequency of sampling shall be performed during and after production in product exposure areas b. Record the date, time, number of particles per cubic foot in the Environmental Monitoring Logbook.
4.3
Personnel Monitoring 4.3.1 Gloved, finger-tip Rodac samples will be taken upon leaving the aseptic processing area as well as during each operation in the critical processing zones. 4.3.2 Rodac plate samples will be taken of each person s gown/suit on the surfaces to the left and right of the chest, upon completion of specific operations in the critical processing zones, as well as when leaving the aseptic area.

Alert limits for a given site shall be established relative to one or more of the following : a. Classification of that area as per national standard. b. Nature of operation and/or product. c. Repeated documentation and subsequent trending of data pertaining to that site. 5.1.1 The Alert Limit will correspond to the 95% upper confidence level of historic data collected over the duration of baseline monitoring of a given area/room. 5.1.2 When an Action Limit is attained, i.e., the air quality level is poorer than the alert limit, corrective action must be implemented. When microbiological viable count exceeds alert limit, colony forming units will be categorized and speculated.

Reporting Results 6.1 6.2 6.3 The QA Manager will review the results of environmental monitoring. When values are within specified limits, Section 7.0 will be followed. Where established Alert or Action Limits are exceeded, the following will be pursued. 6.3.1 QC technician will note any abnormal occurrence or activities prior to, or during, monitoring, if any for that area or zone. 6.3.2 Appropriate corrective action will be recorded and forwarded to the QC Manager.
7.0
Corrective Action Corrective action performed is documented on the appropriate form by the individual performing the corrective actions and approved by the QC Manager.
