Seven extenders, egg-yolk citrate, urea egg-yolk, 0.9% NaCl, Kurokura-2, Ma and Mb and four cryoprotectants viz. DMSO, glycerol, methanol and ethanol were used to find out the suitable cryodiluent. Among the cryodiluents egg-yolk citrate with DMSO yielded (mean±SD) highest post-thaw motility (83%±5.70) that was closely followed by egg-yolk citrate with methanol (81%±6.51) and urea egg-yolk with DMSO (79%±6.47). Six dilution ratios such as 1: 2, 1: 4, 1: 7, 1: 10, 1: 15, and 1: 20 (milt:cryodiluent) were used to determine the suitable milt dilution and found that sperm motility percentage increased roughly with the increase of dilution ratio. Different dilution ratios exhibited marked differences in the post-thaw spermatozoan motility (P=0.000) and revealed that it has an interaction effect with the extenders (P<0.05) but not with the cryoprotectants (P>0.05)....

The whole aquaculture production system is now hampering due to pandemic COVID-19 and the value chain in this sector is quite vulnerable. I conducted a wireless survey through cell phone collected information from different stakeholders of different agro-ecological zones of Bangladesh to assess the impact of the pandemic situations in the aquaculture sector. Survey results revealed that pandemic COVID-19 has been affected in different areas of the aquaculture sector including reduction of consumption, export order cancellation, reduction in price, delay in summer stocking, lack of technical support, and transport crisis.

Several studies showed that among the various natural marine sources of antioxidants, seaweeds have become a potential source of antioxidants because of their bioactive compounds. Most of the metabolic diseases are caused by oxidative stress. It is very well known that antioxidants have a pivotal role in the treatment of those diseases. Recent researches have revealed the potential activity of seaweeds as complementary medicine, which have therapeutic properties for health and disease management. Among the seaweeds, brown seaweeds (Phaeophyta) and their derived bioactive substances showed excellent antioxidant properties than other seaweeds. This research was designed to find out the effect of different factors such as influence of lunar... more This research was designed to find out the effect of different factors such as influence of lunar cycle, harvesting interval, rope type and seeding gap on the production of G. tenuistipitata in coast of Cox’s Bazar. Duration of these experiments were sixty days and all the parameters were recorded fortnightly. Monitoring of water quality parameters indicated that salinity, temperature, transparency, pH and DO were suitable for seaweed cultivation. In determining lunar cycle effect, results envisaged that fresh yield was 14.43% increased when seeding and harvesting time was selected considering the moon cycle. Regarding the selection of harvesting interval, it was found that T3 (30 days interval) was the best to harvest the seaweed whereas T4 (40 days interval) showed decreasing trend in production. Manual separation of male and female tilapia is one of the methods for obtaining single sex. In Bangladesh, abundance of sex reversed GIFT male is very limited, therefore, the experiment was conducted to compare the yield of manually separated 1 1 GIFT with others. Highest yield (5285.3±289.9 kg haG) and gross return (358343.7±23555.6 tk haG) were found from manually separated male GIFT, which was significantly different from others. Even manually separated female GIFT gave the better production than culture of both sexes together and of carps also.
The aim of this study was to characterise the structural features of glycoprotein isolated from t... more The aim of this study was to characterise the structural features of glycoprotein isolated from the brown alga Saccharina japonica (SJGP) and to evaluate its effects on the probiotic properties of Lactobacillus plantarum in Caco-2 cells. The amino acid profile, protein backbone and monosaccharides were analysed by amino acid analysis, Fourier transform infrared spectroscopy (FT-IR) and high-performance liquid chromatography (HPLC), respectively. The pretreatment of L. plantarum with SJGP increased cell adhesion, auto-aggregation and growth in Caco-2 cells, as observed by plate counting, light microscopy and scanning electron microscopy. Auto-aggregation and cell surface hydrophobicity of L. plantarum was also increased following pretreatment with SJGP. SJGP was shown to improve adhesiveness through its lipase inhibitory activity. Adhesion-related genes of L. plantarum showed upregulated expression in the presence of SJGP confirmed by reverse transcriptase-PCR assay. In summary, SJGP could be used as a bioactive compound to improve the probiotic properties of L. plantarum and could be relevant to the preparation of functional foods.

The present study was performed to optimise the extraction of carrageenan from Hypnea musciformis collected from Saint Martin Island, Bay of Bengal, Bangladesh, and characterisation of its chemical, rheological properties and antioxidant activities. Aqueous- and alkali-treated carrageenan from H. musciformis was extracted using conventional method and ultrasonic-assisted extraction (UAE) because this technique allows rapid extraction, which is important to avoid degradation of labile compounds. The resulting carrageenan was investigated in terms of yield, sulphate, 3,6-anhydrogalactose (AG), galactose (Gal) contents and structural properties using Fourier transform infrared (FT-IR) and 1H NMR spectroscopy. The carrageenan yield was higher using the novel UAE method and was comparable to that using the conventional technique. In vitro results confirmed a slight variation in sulphate, AG and Gal contents depending on the extraction method used. FT-IR and 1H NMR spectrum showed that carrageenan extracted using either the conventional method or UAE possesses typical analytical characteristics of κ-carrageenan. The rheological properties of carrageenan were evaluated using differential scanning calorimetery (DSC) and viscosity measurements and found to be similar to those of carrageenan extracted from same species of red alga. The antioxidant activity of carrageenan was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH), OH radical scavenging, reducing power and phosphomolybdenum assays. The results indicated that carrageenan possesses antioxidant activity, the degree of which depends on its structural composition and the extraction method used.

Two months aged experimental fishes were collected from two different regions namely Noakhali and BFRI, Mymensingh. Five enzymes namely LDH, SOD, MDH, ODH and MDH (NADP+) were used in TC-1 buffer system to analyze the genetic variation. LDH and SOD provided clear resolution for muscle tissue. Two presumptive loci were identified where Ldh-1 * loci was found polymorphic in both populations. GIFT populations showed same mean proportion of polymorphic loci which was 50 % whereas GIFU population showed highest mean number of alleles per locus which was 2.0. The highest mean number of effective alleles per locus was 1.47 and the mean proportion of heterozygous loci per individual was 24.31% which was found in GIFT population. Probiotics are living beneficial microorganisms which confer positive health benefits to the host... more Probiotics are living beneficial microorganisms which confer positive health benefits to the host that has been used in aquaculture in Bangladesh since last decades. However, no comprehensive information on the commercial probiotic products has been found in the literature. Therefore, this study was carried out to isolate and characterize of commercial probiotics being used for fish and shrimp culture in Bangladesh. A total of 31 probiotic products were collected from aqua product sellers and stored of which 27 products were in powder form and 4 in liquid state. All the products were imported from foreign countries like Japan, Thailand, Vietnam and India and in most cases no details label information were found. For isolating the microorganisms, eight different types of selective media such as MRS, LB, NB, PDA, BHI, TSB, Aspergillus differentiated, antibiotic resistance etc. were used. A total of 122 isolates were isolated from twenty-two companies commercial probiotics and stored in 30% glycerol at -220C. For molecular identification of isolates, DNA sequence was performed. The results revealed that diversified genus including 59% Enterobacter, 15% Aeromonas, 13% Enterococcus, 8% Streptococcus and 2% Bacillus, Rhodobacter, Moraxella were identified. The preliminary identification of the selected 122 bacterial isolates were carried out based on morphological and biochemical tests such as gram staining, catalase and oxidase test. Regarding biochemical characterization, 80/122 isolates were found gram positive and 42/122 gram negative, 56/122 oxidase positive and 66/122 oxidase negative, 115/122 catalase positive and 7/122 catalase negative. In case of probiotic properties, different in-vitro assay such as pH tolerance, bile tolerance and auto-aggregation were performed. Most of the tested isolates showed moderate tolerance in acidic condition whereas highly resistant in bile salt. In case of auto-aggregation, most of the isolates showed low auto-aggregation capacity. In addition to that antibiotic resistance and hemolytic assay of different isolates were tested. A total of 56 isolates were screened for antibiotic resistance and showed different level of resistance in response to different antibiotics. To determine the pathogenicity of isolates, a total of 122 isolates were screened out by hemolytic assay and found that 48 isolates showed β hemolysis indicating complete lysis of blood cell. Based on the findings it can be concluded that imported probiotic products are in question in terms of label information, contamination with pathogenic microbes, antibiotic susceptibilities as well as other probiotic properties which ultimately hamper the food safety and quality. More comprehensive study is needed to confirm these properties of commercial probiotic products and emphasis should be given on the evaluation of efficacy of different commercial probiotic products according to their instructions of uses.
