The pelC gene, which encodes one of the five major pectate lyase (PL) isoenzymes in Erwinia chrys... more The pelC gene, which encodes one of the five major pectate lyase (PL) isoenzymes in Erwinia chrysanthemi 3937, designated PLc, was subcloned from a hybrid lambda phage into a pBR322 derivative and mutagenized with a mini-Mu-lacZ transposable element able to form fusions to the lacZ gene. One plasmid (pAD1) which had an inactivated pelC gene and a Lac+ phenotype was selected in Escherichia coli. This plasmid was introduced into Erwinia chrysanthemi, and the pelC::mini-Mu insertion was substituted for the chromosomal allele by homologous recombination. This strain lacks the PLc isoenzyme. This Erwinia chrysanthemi strain has a Lac+ phenotype that is inducible by polygalacturonate, as are the wild-type PL activities.

Erwinia chrysanthemi strains 3937 and B374 were shown to secrete 5 major pectate lyase (PL) isozy... more Erwinia chrysanthemi strains 3937 and B374 were shown to secrete 5 major pectate lyase (PL) isozymes with isoelectric points ranging from 4.4 to more than 9.5. To determine the number of pel genes encoding the PL and to study them, two gene libraries were constructed in Escherichia coli, using the Lambda vector L47-1 and the broad host range mobilizable cosmid pMMB33. These gene libraries provided several clones producing the PL. The pel genes of the strain B374 were also cloned by an in vivo procedure promoted by the pULB113 plasmid. Subcloning experiments have shown the existence of 5 pel genes, each with its own promoter, clustered in two regions on the chromosome of E. chrysanthemi. The restriction maps of these regions from 3937 and B374 were similar. In addition, a pectin methylesterase gene of 3937 was found closely linked to one pel gene. Different pel genes were mutagenized in E. coli using a mini-Mu phage able to promote translational fusions with lacZ. One fusion in the pelC gene was introduced by marker exchange into the chromosome of E. chrysanthemi 3937, giving rise to a strain lacking the PLc isozyme and allowing study of the regulation of pelC.

This technique has been used after nondenaturing blotting transfer in order to detect depolymerizing enzyme activities once analytical protein separation (e.g., by electrophoresis, electrofocusing, or titration curves) has been completed. This rapid and very sensitive method was successfully applied to the enzymes polygalacturonate lyase, polygalacturonate hydrolase, endoglucanase, and xylan hydrolase. Other possible applications are presented.

Cet ouvrage est destiné à faire le point sur les enzymes qui catalysent l'hydrolyse ou la dépolym... more Cet ouvrage est destiné à faire le point sur les enzymes qui catalysent l'hydrolyse ou la dépolymérisation des molécules d'origine biologique. Il rassemble les textes d'une série de conférences prononcées au cours d'un séminaire animé par Annette Mouranche et Claude Costes au Centre de Grignon de l'Institut national agronomique. Lorsqu'il s'agit de macromoléeules, comme par exemple l'amidon, la cellulose ou les caséines, leur utilisation industrielle implique souvent une simplification chimique préalable. Cette simplification est souvent à l'origine l'hydrolyse d'une fonction ester ou amide. Cette réaction hydrolytique est à la base de bien des processus technologiques dans les industries alimentaires: fromagerie, industrie des viandes, industrie des jus de fruits, brasserie, amidonnerie. De même la valorisation de la biomasse lignocellulosique commence par une série de réactions de dépolymérisation. L'ensemble de ces réactions est le plus souvent réalisé par voie enzymatique. Une meilleure connaissance des enzymes concernées permettra un élargissement des technologies alimentaires, un renouvellement de certaines stratégies phytosanitaires, une exploitation plus rationnelle des déchets animaux et végétaux ou la fabrication de substances nouvelles par hémisynthèse industrielle. Quelle est l'origine biologique de ces enzymes? Que sait-on sur leur structure, leurs propriétés et leur mode d'action? Comment les met-on en œuvre ou comment peut-on élargir leur utilisation industrielle? Cet ouvrage essaie de présenter une analyse de ces problèmes et propose des perspectives nouvelles. La vaste catégorie des nucléases n'a pas été abordée dans ce travail qui s'adresse principalement à tous ceux qui, étudiants, ingénieurs, responsables d'entreprise ou enseignants, sont concernés par la transformation industrielle de la production agricole, végétale ou animale.
