Methods: Among the seventy-nine patients included in the VeriStrat® trial (evaluating the role of pretreatment circulating tumor markers as predictors of prognosis), we selected patients submitted to FDG-PET/CT for clinical reasons just before the inclusion in the trial. For each patient a peripheral blood sample was collected at baseline for evaluation of CTCs and cfDNA. CTCs were isolated by size using a filtration-based device (ScreenCell) and then identified and enumerated; cfDNA was isolated from plasma and quantified by qPCR method using human telomerase reverse transcriptase (hTERT). The following FDG-PET/CT-derived parameters were computed: maximum diameter (dmax) of the primary lesion (T), of the grea...

We applied a computational quantification software application to hybrid FDG PET/CT images to assess activity and distribution of the hematopoietic system throughout the whole skeleton of recently transplanted patients. 34 patients underwent PET/CT 30 days after either adult stem cells (ACT, n=18) or cord blood transplantation (CBT, n=16). Our software automatically recognized compact (CBV) and trabecular bone (IBV) in CT slices. Within IBV, co-registered PET data were extracted to identify the active BM (ABM) from the inactive tissue. Patients were compared with 34 matched controls, chosen among a published normalcy database. Whole body ABM increased in ACT and CBT when compared to controls (12.4±3 and 12.8±6.8 vs 8.1±2.6 ml/Kg of ideal body weight, IBW, p<0.001). In long bones, ABM was three- and six-fold increased in CBT and ACT, respectively, in comparison with controls (...

The selective breakdown of brain networks during the progression from the normal aging (NA) to Alzheimer's Disease (AD) has also been observed. We implemented Independent Component Analysis (ICA) on 18F-FDG-PET data in five groups of subjects with cognitive state ranging from NA to AD dementia, including mild cognitive impairment patients not converting (ncMCI) and converting (MCI) to mild AD dementia, in order to disclose the spatial distribution of the independent components (ICs) in each cognitive state, and their accuracy in discriminating the groups. We could identify spatially distinct ICs in each group with an increasing generation of new local circuits proportional to the severity of the disease. AD-specific ICs appeared from the late MCI stage and could discriminate MCI and AD dementia from ncMCI with an accuracy of 8...
