    A clinical trial pilot study, double-blinded, randomized, and controlled with a placebo to assess the effectiveness of oral doxycycline (200 mg, single dose) in preventing leptospirosis after high exposure to potentially contaminated water was performed in São Paulo, SP, Brazil. Confirmed cases were defined as those with leptospira IgM antibody and symptoms; asymptomatic cases were those presenting with IgM antibodies but no symptoms; and suspected cases were individuals with symptoms but no IgM antibody. Forty subjects were given doxycycline and 42 were given placebo. In the drug-treated group there were 2 confirmed cases, 11 asymptomatic cases, and 6 suspected cases. In the placebo group there were 5 confirmed, 6 symptomatic, and 5 suspected cases. Even though we found a protective association of doxycycline for confirmed leptospirosis cases (RR = 2.3) and seroconversion only (RR = 2.0), the association was not statistically significant because of the small number of individuals enrolled in this pilot study. We observed that the 22% of the volunteers already had IgM antibodies to leptospirosis at the first sampling. Finally, the attack rate to confirmed, asymptomatic, and suspected cases of Leptospirosis was 8.5%, 22%, and 13%, respectively, in this population.

    Suspicion of Brazilian spotted fever (BSF) should occur in endemic regions upon surveillance of the acute febrile icteric hemorrhagic syndrome (AFIHS). However, limitations associated with currently available laboratory tests pose a challenge to early diagnosis, especially in fatal cases. Two real-time PCR (qPCR) protocols were evaluated to diagnose BSF in 110 fatal AFIHS cases, collected in BSF-endemic regions in 2009-2010. Of these, 24 were positive and 86 negative by indirect immunofluorescence (IFA) assay (cut-off IgG and/or IgM ≥ 128). DNA from these samples was used in the qPCR protocols: one to detect Rickettsia spp. (citrate synthase gene) and another to determine spotted fever group (SFG) Rickettsia species (OmpA gene). Of the 24 IFA-positive samples, 5 (21%) were positive for OmpA and 9 (38%) for citrate synthase. In the IFA-negative group (n=86), OmpA and citrate synthase were positive in 23 (27%) and 27 (31%), respectively. These results showed that the 2 qPCR protocols were about twice as sensitive as the IFA test alone (93% concordance). In conclusion, qPCR is a sensitive method for the diagnosis of fatal BSF cases and should be considered for routine surveillance of AFIHS in places like Brazil, where spotted fever-related lethality is high and other endemic diseases like dengue and leptospirosis can mislead diagnosis.

    Utilizaram-se 153.963 controles mensais de produção de leite e 13.273 primeiras lactações de vacas da raça Holandesa, com partos entre 1989 a 1998, com o objetivo de estimar parâmetros genéticos, fenotípicos e de meio ambiente para produção de leite no dia do controle (PLDC) e produção até 305 dias de lactação (P305), bem como verificar a conveniência de se utilizar a PLDC em avaliações genéticas, em substituição à P305.     Under an agreement of scientific cooperation between Brazil and Japan, a prototype detector of cosmic ray muons has been operating since March 2001 at Southern Space Observatory - SSO located at Sao Martinho da Serra (29oS, 53oW), Brazil, in order to observe cosmic ray precursors of geomagnetic storms. This detector plays a key role in the prototype network of muon observations together with two larger detectors operating in Japan and Australia. The planned extension of the detector in its size will complete the global coverage of our muon detector network. The prototype network has already discovered cosmic ray precursors of several magnetic storms, as reported by Munakata et al. (2000, 2001). We have also observed the Forbush Decreases (FD's), as well as the precursory enhancements of cosmic ray anisotropy preceding the onsets of geomagnetic storms. This report presents the description of the network and some results obtained since the prototype detector implementation.

    ABSTRACT This study evaluated the performance of two carbon analyzer catalyzers, one with high sensitivity and another with regular sensitivity, used in the determination of carbon concentration in a total organic carbon analyzer – TOC. Its applicability was then evaluated in an environmental monitoring study. They have been constructed two calibration curves with 5 points each, first in the interval of 2 mgL-1 at 10 mgL-1 and the second in the interval of 10 mgL-1 at 100 mgL-1. Both calibration curves showed a high level of linearity that can be observed by R2 values that were 0.9996 for the concentration range of 2 mgL-1 to 10 mgL-1, 0.9999 for the concentration range of 10 mgL-1 to 100 mgL-1, and 0.9999 for the concentration range of 100 mgL-1 to 1.000 mgL-1. The limits of detection and quantification for inorganic carbon were, respectively, 0.003 mgL-1 and 0.009 mgL-1. The limits of detection and quantification for the different catalyzers were also evaluated and showed the following values: regular sensitivity: 0.015 mgL-1 and 0.050 mgL-1, respectively, and high sensitivity; 0.004 mgL-1 and 0.013 mgL-1, respectively. The environmental monitoring was performed in two points (PB1 and PB2) in the Garcia stream. Carbon concentrations were monitored during 24 hours with 2-hour intervals between sampling. It was observed that the concentrations of carbon remained constant throughout the experiment, with the exception of the period between 10:00 am and 04:00 pm, probably due to the input of carbonates and bicarbonates derived from textile companies in the region or still had to the launching of urban sewers. Comparing the average carbon concentration, values of point PB2 were always higher than those observed at point PB1. In high concentration analysis, only irrelevant variations were observed with the use of both catalyzers.

    The objective of this study was to investigate the frequency of HIV infection among female sex workers in the port area of Imbituba (State of Santa Catarina), and to identify the viral subtype and its susceptibility to antiretroviral medications. Ninety women were interviewed between December 2003 and February 2004. Six (6.7%) were HIV-positive. Genotyping for HIV, performed on four samples, detected subtype C in three of them, which is predominant in Africa and Asia, and subtype B in one of them, which is prevalent in Brazil, USA and Europe. The results suggest that the Port of Imbituba may be one of the gateways for HIV-1 subtype C to enter Brazil, and for its dissemination to the rest of the country and the Mercosul area, along the highway BR-101. This points towards the need for preventive work to reduce the introduction and dissemination of HIV subtype C in Brazil.

    Here a young patient (< 21 years of age) with a history of infective dermatitis is described. The patient was diagnosed with myelopathy associated with HTLV-1/tropical spastic paraparesis and treated with interferon beta-1a. The disease was clinically established as HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP), and laboratory tests confirmed the presence of antibodies to HTLV-1 in the cerebrospinal fluid (CSF). Mumps, cytomegalovirus, Epstein-Barr virus, schistosomiasis, herpes virus 1 and 2, rubella, measles, varicella-zoster toxoplasmosis, hepatitis, HIV, and syphilis were excluded by serology. The patient was diagnosed with neurogenic bladder and presented with nocturia, urinary urgency, paresthesia of the lower left limb, a marked reduction of muscle strength in the lower limbs, and a slight reduction in upper limb strength. During the fourth week of treatment with interferon beta-1a, urinary urgency and paresthesia disappeared and clinical motor skills improved.

    Infection of non-adherent TG180 murine sarcoma cells with Toxoplasma gondii was compared, at the ultrastructural level, in both in vivo and in vitro conditions. Suspensions of 3.0 x 10(6) TG180 cells infected in vitro with 1.0 x 10(6) parasites of the RH strain were harvested between the first and 6th day post-infection and processed for transmission electron microscopy. In vivo infection was made by intraperitoneal inoculation in mice of 1.0 x 10(6) TG180 cells, that were co-inoculated with a parasite suspension at the same cell concentration. Cells were harvested 10, 20, 30 min and 24, 48 h post-inoculation and processed for transmission electron microscopy at the same conditions of the in vitro culture. It was observed TG180 murine sarcoma cells with intense and equivalent intracellular parasitism in both conditions. Host cells with parasitophorous vacuoles containing up to 16 parasites, as well as parasites undergoing mitoses or presenting a bradyzoite-like morphology, were frequently seen in both culture methods.

    Linkage disequilibrium (LD) across the genome is critical information for association studies and genomic selection because it determines the number of SNP that should be used for a successful association analysis and genomic selection. Linkage disequilibrium also influences the accuracy of genomic breeding values. Some studies have demonstrated that SNP in strong LD are organized into discrete blocks of haplotypes, which are separated by possibly hot spots of recombination. To reduce the number of markers needed to be genotyped for association mapping, a set of SNP can be selected that labels all haplotype blocks. We estimated the LD, calculated the average haplotype block size for 6 pig lines, and compared the block size between lines. Six commercial pig lines were genotyped using the Illumina PorcineSNP60 (number of markers M = 62,163) Genotyping BeadChip (Illumina Inc.); on average, a panel of 37,623 SNP with an average minor allelic frequency (MAF) of 0.283 was included in the analysis. The LD declined as a function of distance. All pig lines had an average r(2) above 0.3 for markers 100 to 150 apart. The estimated average block size was 394.885 kb, and blocks between 100 and 400 kb were most prominent (49.96%) in all lines. These results showed that the extent of LD in pigs is much larger than in the cattle population, in accordance with the genetic map length of pigs, which is much shorter than cattle. The evaluated lines have 2,640 to 3,037 blocks, covering 45% of the pig genome, on average. Differences in haplotype block size between lines were observed for some chromosomes (i.e., SSC 3, 5, 7, 13, 14, and 18), which provide a direction for future studies of haplotype block conservation or divergence across lines.
