Brucellosis is an infectious disease that affects livestock and may be transmitted to humans. Cattle may become infected with Brucella spp. by various routes, and the pathogens induce both humoral and cellular immune responses in the host organism. The aim of this study was to determine the characteristics of the cellular immune response by using a brucellin allergen in serologically positive cows, and to differentiate cross- reactions from true positive animals, and to contribute to improvement of the overall diagnostics of bovine brucellosis in Bosnia and Herzegovina. Using the Rose Bengal Test (RBT) and Complement Fixation Test (CFT) as a combined reference standard (CRS), seropositive (n=15) and seronegative (n=14) groups were defined. Polymorphism exists in β-lactoglobulin (LGB) milk encoding gene in dairy animals, resulting in presence of different protein isoforms in milk. There is a link between genetic polymorphism in dairy proteins with technological properties of milk, milk yield and allergic reactions in humans. This study was conducted to identify LBG protein isoforms and their frequencies in population of Dubska Pramenka sheep breed. A total of 90 blood samples were genotyped using PCR-RFLP analysis. Genotypes were determined by PCR amplification followed by digestion with RsaI for identification of A and B alleles and using MspI for C allele identification. Two genetic variants (A and B) and three genotypes (AA, AB and BB) were identified. The presence of allele C in this study was not noticed. Gene frequencies of LGB A were 13; LGB B were 61; LGB AB were 61. There is no data on the genetic influence on the quality and quantity of milk from Dubska pramenka sheep, and there is need for further studies.

The aim of this study was to investigate liver function in dairy sheep during summer and consequences made by heat stress. The experiment was performed on 117 Pramenka sheep at two different locations (Travnik and Livno) during summer grazing period (July, August and September). Levels of serum enzymes were determined for alanine aminotransferase (ALT), aspartate aminotransferase (AST)), gamma-glutamyl transferase (GGT), alkaline phosphatase (AP) and lactate dehydrogenase (LDH). Enzyme activities resulted in significant (P<0.05) variation except GCT and a moderate increase followed by dcrease in activity, which is likely to be correlated with the energy balance of animals. Values of serum profile above reference range may indicate intense metabolic processesasaliver response toa negative energy balance. The results of this research couldbeused for the determination ofhealth status.

Poznati proizvodi mljekarske industrije u Bosni i Hercegovini su autohtoni Livanjski i Travnički sir koji pripadaju grupi sireva proizvedenih od sirovog, nekuhanog ovčjeg mlijeka. Livanjski i Travnički sir su specifični zbog specifičnog biljnog pokrivača na području gdje se proizvode, kao i klimatskih uvjeta i mlijeka autohtone ovce pramenke, tako da možemo očekivati razlike u sadržaju aromatskih komponenti, koje u velikoj mjeri određuju karakteristike spomenutih sireva. Sinteza isparljivih aromatskih komponenti je dinamičan proces koji započinje sazrijevanjem sira do karakterističnog okusa, koji se razvija u vrijeme sazrijevanja. The aim of this investigation is to determine changes in concentrations of biochemical and metabolic hormones in the blood of Pramenka sheep. The study included 117 sheep of Pramenka breed monitored during three different periods (July, August, September) from Livno and Travnik (Vlašic) areas. The following biochemical parameters were determined in sheep blood samples: glucose, cholesterol, triglycerides, total protein, albumin, urea, aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), gamma-glutamyltransferase (GGT), alkaline phosphatase (ALP), were performed by spectrometric analyzer (Kodak Ektachem, analyzer DT 60 II, DTSC Modul and DTE Modul). In sheep blood serum determined the following hormones by ELISA values: triiodothyronine, thyroxine, cortisol, and insulin. Hormonal status of test animals was relatively stable during periods of sampling, but significant differences have been confirmed based on locality, with the exception of cortisol. The lack of statistical significance of differences for cortisol between areas of Livno and Travnik can be due to stress caused by collecting blood samples which have equally increased cortisol levels and “concealed” all other factors that affect its level in the serum. High concentrations of thyroid hormones, particularly in sheep from the locality of Livno, can be a sign of decreased appetite, which is one of the earliest disorders in conditions of heat stress. In both areas, high insulinemia was determined as well, possibly as a result of feeding which significantly changed the hormonal regulation of metabolism in lactation process and led towards stimulation of the endocrine pancreas.

Species identification in food has become a prominent issue in recent years as the importance of consumer protection has increased. DNA-based species identification methods were developed by researchers in the last two decades, as these are reliable, accurate, and low-cost techniques for species identification in raw and processed food products as well. In our study, universal primers were designed to conserved regions of mitochondrial 12S rRNA. Amplicons were heat-denatured and a PCR single strand conformation polymorphism (SSCP) method was developed to identify cattle, buffalo, sheep, and goat DNA. Sensitivity of this technique was tested on DNA mixtures of cattle-sheep, cattle-goat, and cattle-buffalo and the threshold limit of cattle DNA was 5%, 5%, and 3%, respectively. One hundred and five cheeses were purchased and collected from Bosnian and Hungarian farmers, retails, and supermarkets to reveal fraud, 32 percent of them (34 cheeses) were found to be mislabelled by species.

The study included a total of 127 sheep milk samples from two different areas (Livno and Travnik) in summer feeding period (July, August and September). Fatty acids in milk were determined by gas chromatography (GC). The animals were marked with the appropriate number of ear tags on the basis of which we always took samples from the same animals through different periods. Fatty acids in milk were determined by gas chromatography and the following fatty acids composition: butyric acid, caproic acid, caprylic acid, capric acid, stearic acid, oleic acid, linoleic acid, arachidonic acid, eicosapentaenoic acid, docosahexaenoic acid, rumenic acid. The fatty acid content of sheep's milk in this study showed a tendency of variation, both within and between sampling areas, and characterized by its relatively high content of saturated fatty acid (SFA) during the period of harvest.

The serum concentrations of the minerals (calcium, phosphorus and magnesium) were followed over time. Based on the physical-chemical analysis of the sheep milk, we evaluated the milk parameters (milk fat, protein and lactose) and cheese parameters (dry matter, moisture, grease and pH) that can be indicative of the cheese milk quality. The aim of the study was to determine the effect of the serum minerals to the sheep milk quality, and the Livno and Travnik cheese quality, being the final products. Blood, milk and cheese samples were taken in the summer during the summer grazing of sheep. The results were analyzed using the software package/Program SPSS 15.00. The research was performed on 40 chicken broilers of Cobb provenience to investigate the effect of dietary black cumin seed oil on some blood biochemical parameters in chicken broilers. 40 chicken broilers who were included in the 42 days lasting experiment were divided into two groups, each one of 20 individuals. Group K was the control – with no added oil in feed mixture and the second group P was the experimental one – chickens were fed with 0,025 g of p.o. administered black cumin seed oil. Feed and water supplies were ad libitum . Microclimate conditions (light, temperature and airflow) were maintained according to the technological procedure. Metabolizam visokomliječnih krava, obzirom na njihovu genetsku predispoziciju za visoku proizvodnju mlijeka s jedne i reproduktivnih zahtjeva s druge strane, često je opterećen te izložen promjenama koje za posljedicu mogu imati poremećaj funkcionalnog stanja pojedinih organa, a najčešće jetre i genitalnih organa. Fiziološke vrijednosti biokemijskih parametara krvi krava, koje nude različiti izvori, često znaju znatno varirati. The indigenous Travnik (Vlasic) cheese is produced in central Bosnia in the area of Mt. Vlasic. This cheese belongs to the group of white soft cheeses, ripened in brine under anaerobic conditions, and made from raw, thermally untreated sheep’s milk. The production technology is very simple and is adapted to mountainous conditions. Cheese is an important source of essential nutrients, in particular proteins, fat, vitamins and minerals. The aim of this study was to determine the concentration of macroelements (Ca, Na, K, Mg, P) and microelements (Zn, Fe, Cu) in 15 samples of Travnik sheep cheese using the atomic absorption spectrometry (AAS) and ultraviolet–visible spectrometry (UV-VIS) methods. A tendency of increasing Mg and K, Fe and Ca, Fe and Mg, and Fe and P was determined in the examined samples, but without statistical significance. The correlation coefficient values showed statistical validity at the level of high significance of differences (P<0.01).

Brucele u organizmu inficiranih životinja induciraju i humoralnii ćelijski imunološki odgovor. Humoralni imunološki odgovor se bazira na praćenjupojave specifičnih antitijela protiv glatkog lipopolisaharida (S-LPS) ćelijskemembrane brucela. Međutim kod goveda u dokazu specifičnih antitijela klasičnimserološkim metodama mogu se detetkovati antigene determinate za druge vrstemikroorganizama (unakrsna reaktivnost) kao što su: Escherichia coli 0:157, Yersiniaenterocolitica 0:9, Salmonella urban, Pseudomonas malthopilia i Pasteurellae. Cilj ovograda je utvrditi ćelijski imunološki odgovor koji se bazira na upotrebi pročišćenog istandardiziranog alergena, kojem je u potpunosti odstranjen lipopolisaharid te kaotakav ne dovodi do razvoja humoralnog imunološkog odgovora. Istraživanje autohtone proizvodnje Livanjskog i Travničkog sira ima za cilj očuvanje tradicije, organizirane proizvodnje i plasmana na tržišta izvan domicilne regije. Autohtoni sirevi su raznovrsnijeg okusa, arome i konzistencije u odnosu na industrijski proizvedene sireve, gdje je tehnologija definirana, a uvjeti proizvodnje kontrolirani. Nadmorska visina, kvalitetna voda i hrana nisu dovoljni čimbenici za trajno održavanje kvalitete autohtonih sireva. Na osnovi standardizacije tehnoloških postupaka proizvodnje može se proizvesti sir specifičnog obilježja. Cilj istraživanja bio je odrediti i komparirati ukupni dušik i dušične frakcije u dva autohtona sira – Livanjskom i Travničkom, ovisno o terminu uzorkovanja (srpanj, kolovoz i rujan).
