Characterization of Bacillus megaterium and Bacillus mycoides Bacteria as Probiotic Bacteria in Fish and Shrimp FeedThis study was aimedto identify probiotic characteristics and to test the cellulolytic ability of Bacillus megaterium and Bacillus mycoides bacteria for probiotic microbe candidates in fish and shrimp feed. The description of the cellulolytic and amylolytic abilities of these bacteriawas obtained by non-experimental method and descriptive analysis. Probiotic characteristic identification includes growth curve was obtained through total plate count method, cellular and colony morphology, and cellulase and amylase enzyme activity test using DNS method. Results indicated that the maximum growth of B. megateriumwas observed after six hours at 35.62 x 10 10 (CFU), while B. mycoideswasafter 30 hoursat 42.6 x 10 10(CFU). The macroscopic observation showed that the colony of B. megateriumwas concave and smooth,while B. mycoides was flat, relatively rough, with silken threads around the colony.Both bacteria had milky white color, bacillus shape, Gram positive, and sporous. Sub-Project Completion Report Characterization of Commercial Probiotics being Used for Fish and Shrimp Culture in Bangladesh2019 •
Probiotics are living beneficial microorganisms which confer positive health benefits to the host that has been used in aquaculture in Bangladesh since last decades. However, no comprehensive information on the commercial probiotic products has been found in the literature. Therefore, this study was carried out to isolate and characterize of commercial probiotics being used for fish and shrimp culture in Bangladesh. A total of 31 probiotic products were collected from aqua product sellers and stored of which 27 products were in powder form and 4 in liquid state. All the products were imported from foreign countries like Japan, Thailand, Vietnam and India and in most cases no details label information were found. For isolating the microorganisms, eight different types of selective media such as MRS, LB, NB, PDA, BHI, TSB, Aspergillus differentiated, antibiotic resistance etc. were used. A total of 122 isolates were isolated from twenty-two companies commercial probiotics and stored in 30% glycerol at -220C. For molecular identification of isolates, DNA sequence was performed. The results revealed that diversified genus including 59% Enterobacter, 15% Aeromonas, 13% Enterococcus, 8% Streptococcus and 2% Bacillus, Rhodobacter, Moraxella were identified. The preliminary identification of the selected 122 bacterial isolates were carried out based on morphological and biochemical tests such as gram staining, catalase and oxidase test. Regarding biochemical characterization, 80/122 isolates were found gram positive and 42/122 gram negative, 56/122 oxidase positive and 66/122 oxidase negative, 115/122 catalase positive and 7/122 catalase negative. In case of probiotic properties, different in-vitro assay such as pH tolerance, bile tolerance and auto-aggregation were performed. Most of the tested isolates showed moderate tolerance in acidic condition whereas highly resistant in bile salt. In case of auto-aggregation, most of the isolates showed low auto-aggregation capacity. In addition to that antibiotic resistance and hemolytic assay of different isolates were tested. A total of 56 isolates were screened for antibiotic resistance and showed different level of resistance in response to different antibiotics. To determine the pathogenicity of isolates, a total of 122 isolates were screened out by hemolytic assay and found that 48 isolates showed β hemolysis indicating complete lysis of blood cell. Based on the findings it can be concluded that imported probiotic products are in question in terms of label information, contamination with pathogenic microbes, antibiotic susceptibilities as well as other probiotic properties which ultimately hamper the food safety and quality. More comprehensive study is needed to confirm these properties of commercial probiotic products and emphasis should be given on the evaluation of efficacy of different commercial probiotic products according to their instructions of uses.

Characterization of the three selected probiotic strains for the application in food industry2008 •
Cellulase enzymes have attracted considerable attention in recent years due to their great biotechnological and industrial potential. Cellulase enzymes provide a key opportunity for achieving tremendous benefits of biomass utilization through the bioconversion of the most abundant cellulosic material into the simplest carbohydrate monomer, glucose. Nowadays, the sources of cellulase-producing bacteria have been broadened into the presence of symbiotic bacteria in herbivorous animal and also from marine. Takifugu rubripes or known as Puffer fish is a unique poisonous vertebrate but nevertheless is considered a delicacy in Korea. The diet of the puffer fish includes mostly algae. This dietary habit considers Puffer fish as host of cellulase-producing bacteria, especially on its gut. In the present study an attempt has been made to search for the cellulolytic bacteria in the gut of Takifugu rubripes. Fifty five microorganisms have been isolated using 1% (w/v) Carboxymethyl cellulose (CMC) as substrate. Congo red dye test and DNS method were then used for screening the extracellular cellulase activity of the strains. Among them, TD6 strain has shown the highest performance in term of cellulase activity. In order to evaluate the optimum culture condition of the isolate TD6 for cellulase production, the strain was grown at various temperatures, pH, carbon sources, and nitrogen sources. Under optimum condition, the maximum specific activity of 2.13 U/mg protein was achieved after growth the strain with 1.5% CMC at 45ºC pH 6 for 3 days, respectively. Based on 16S rRNA gene analysis it is proposed that the strain was identified as Bacillus subtilis.

This article examines challenges and opportunities associated with the use of the U.S. military in support of U.S. government efforts to bolster governance and combat insecurity in operations short of combat, in the context of Latin America and the Caribbean. It examines associated challenges with respect to budgetary guidelines and other regulations, and the difficulties of coordinating with partner nations and other parts of the U.S. government. It ends with specific recommendations for U.S. policymakers for how to better use the U.S. military as part of a whole of government approach to strengthening security and governance in the region.
