Physiochemical properties of biomaterials play critical roles in dictating types of cell behaviour. In this study, a series of poly(ε-caprolactone) (PCL)-derived polymers bearing different small chemical groups was employed as a platform to evaluate chondrogenesis of different cell types. Thin films were prepared by spin-coating PCL derivatives. Rabbit articular chondrocytes (rACs) and rabbit bone marrow-derived mesenchymal stem cells (rMSCs) were seeded on to the films, and cell adhesion, proliferation, extracellular matrix production and gene expression were evaluated. The presence of hydrophilic groups (-NH2 , -COOH, -OH and -C=O) promoted adhesion and proliferation of primary rACs and rMSCs. On these polymeric films, chondrogenesis of primary rACs depended on culture time. For passaged cells, re-differentiation was induced on these films by chondrogenic induction, but less for cells of passage 5 compared to passage 3. This study was designed to examine antioxidant and moisturizing effect of purified GAGS from ascidian tunics as functional cosmetic ingredient. A skin-tonner and essence which contain 1.0% of GAG were manufactured conventionally. These product then were tested for 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, collagenase inhibitory activity, collagen-formation stimulator, and fibroblast-proliferation promoter abilities. The DPPH scavenging activity for skin-tonner and essence were 20.5% and 12.4% respectively. Collagenase inhibitory activity of skin-tonner was higher (94.6%) than the essence (80.1%). Skin-tonner and essence with GAGs showed collagen-formation stimulation ability, which were 129.9% and 120.2% respectively. These products also demonstrated significant (p<0.01) fibroblast-proliferation promoter abilities.

Mucopolysaccharidosis III (MPS III) is characterized by lysosomal accumulation of the glycosaminoglycan (GAG) heparan sulphate (HS). In humans, the disease manifests in early childhood, and is characterized by a progressive central neuropathy leading to death in the second decade. This disease has also been described in mice (MPS IIIA and IIIB), dogs (MPS IIIA), emus (MPS IIIB) and goats (MPS IIID). We now report on dogs with naturally occurring MPS IIIB, detailing the clinical signs, diagnosis, histopathology, tissue enzymology and substrate levels. Two 3-year-old Schipperke dogs were evaluated for tremors and episodes of stumbling. Examination of the animals found signs consistent with cerebellar disease including dysmetria, hind limb ataxia and a wide-based stance with truncal swaying. A double-blinded, controlled clinical study was performed to compare the response of adult dogs affected with hip dysplasia to a placebo and three different dosages of polysulfated glycosaminoglycan (PSGAG): 2.2 mg/kg, 4.4 mg/kg, and 8.8 mg/kg. Dogs were randomly assigned to treatment groups. The drug was administered intramuscularly every 3 to 5 days for a total of eight injections. Response to treatment was analyzed based on changes in lameness, range of motion (ROM), and pain on manipulation of the hip joints. Evaluation for adverse reactions included complete blood cell (CBC) count, blood urea nitrogen (BUN), creatinine, and physical examination. Data were collected on a total of 111 dogs. Eighty-four met all criteria for inclusion in the study. Dogs that were given 4.4 mg/kg of PSGAG showed the greatest improvement in orthopedic scores, whereas dogs in the placebo group showed the smallest improvement; however, the differences in clinical improvement between the four treatment groups were not statistically significant. No local or systemic adverse reactions related to the drug were observed.

The cell line HMC-1, derived from a patient with mast cell leukaemia, is the only established cell line exhibiting a phenotype similar to that of human mast cells. This paper reports on a detailed characterization of the expression of a panel of markers for various types of immature and mature haematopoietic cells in the HMC-1. We also studied the potential of HMC-1 to differentiate upon treatment with conditioned media from the human T-cell line Mo, retinoic acid or DMSO. HMC-1 was found to express several mast cell-related markers. A high expression of Kit, the receptor for stem-cell factor, was detected. The majority of the cells were stained with a MoAb against the mast cell-specific serine protease tryptase. Of particular interest was the finding that beta-tryptase mRNA, but not alpha-tryptase mRNA, was expressed in HMC-1. The following review is focused on a very important wound healing biomaterial class, i.e. glycosaminoglycans, a heteroglycan-based compound. Every biomaterial has its own advantages and disadvantages, but these disadvantages are usually surpassed by blending through different methods with various other compounds. The main advantages of glycosaminoglycans include their water solubility that allows them to maintain the wound environment moist and to absorb the wound exudate. If they are mixed with other materials they can help hemostasis, they do not adhere to the wound bed and thus prevent pain when they are removed from the injured site. They are also known to have antibacterial and antioxidant properties, and low cytotoxicity. This review underlines the benefits glycosaminoglycan application has on wound healing, including the use of several blends that enhance its characteristics.

To investigate their temporal order and interrelationships, we have developed a pulse-chase protocol that follows the synthesis and packaging of a sulfated hormone, pro-opiomelanocortin (POMC). In AtT-20 cells, sulfate is incorporated into POMC predominantly on N-linked endoglycosidase H-resistant oligosaccharides. Subcellular fractionation and pharmacological studies confirm that this sulfation occurs at the trans-Golgi/TGN.
