Harderian Gland

A 22-month-old, female rabbit was presented with a 1-day history of acute unilateral exophthalmos. Ultrasonography and computed tomography (CT) of the orbit revealed an orbital mass. Retrobulbar lymphoma was diagnosed following fine-needle aspiration biopsy (FNAB). Thoracic radiographs were normal, and ultrasonography of the abdomen showed focal hypoechoic thickening of the bowel wall and hypoechoic enlarged lymph nodes. The rabbit was euthanized and histopathology identified the retrobulbar mass as B-cell malignant lymphoma of the Harder's gland. Mesenteric lymph nodes, caecum, and both kidneys were also affected. This is the first documented case of malignant lymphoma of the Harder's gland in a rabbit.

The harderian gland was first described in 1694 by Johann Jacob Harder (1656-1711). It occurs in most terrestrial vertebrates and is located within the orbit where, in some species, it is the largest structure. It may be compound tubular or compound tubuloalveolar, and its secretory duct is usually morphologically distinct only after leaving the substance of the gland to open on the surface of the nictitating membrane. The tubules of the gland are formed of a single layer of columnar epithelial cells surrounded by myoepithelial cells. The chief product(s) of the gland varies between different groups of vertebrates, and epithelial cells possess granules or vacuoles whose contents may be mucous, serous or lipid. The harderian gland in the Wistar rat consists of tubules with wide lumina lined by a single layer of columnar epithelial cells possessing myoepithelial cells within their basal laminae. The gland contains porphyrin pigment which is stored as solid intraluminal deposits. The glandular epithelium possesses 2 cell types, termed A and B. These are characterised by an extraordinarily well-developed tubular smooth endoplasmic reticulum and numerous lipid vacuoles. Type A cells can be distinguished from type B by the number, size and content of the lipid vacuoles. Type A cells are more numerous. They contain large lipid vacuoles with dense ribbon-like material identical in form to the material in the luminal masses of porphyrin pigment, whereas those of type B cells are small with crescentic dense lamellar material. The Harderian gland, which is situated within the bony orbit, is usually thought of as a source of lubrication for the eye. However, recent studies have suggested links with reproductive function. In the male golden hamster, both gland histology and activity are known to be under hormonal influence, and the present experiment was undertaken to examine gland weight and activity (as measured by the production of porphyrins) over the oestrous cycle and during pregnancy and early lactation in the female hamster. Gland weight, the number of solid intraluminal porphyrin accretions, and concentrations of copro- and proto-porphyrin were all maximal on day 1 of the cycle (oestrous) and at their lowest on day 2 (or jointly on days 2 and 3), rising gradually thereafter. Development of Harderian gland of larvae of Rana japonica, Bufo bufo japonicus, and Xenopus laevis was studied. In the adult animals, well-developed Harderian glands were invariably present in the orbit. In Rana and Bufo, the gland first appeared at late prometamorphic stage and in Xenopus it appeared around climax stage. In thyroidectomized tadpoles of Bufo and Rana, the Harderian gland was induced by thyroxine. In hypophysectomized tadpoles of Bufo the gland developed when they were treated with thyroxine or TSH.

Hamster (Mesocricetus auratus) harderian gland (HG) is a dimorphic orbital gland producing a copious lipid secretion. Two cell-types are present in hamster HG, type I in both sexes, type II only in males. In hamster HGs, we found a marked sexual dichotomy in the expression of uncoupling protein-3 (UCP3), a mitochondrial protein carrier, that probably exports fatty acid anions and fatty acid peroxides from the mitochondrial matrix. Following castration and/or testosterone treatment: (1) UCP3 levels correlated with the type II-cell percentage, not with testosterone levels, (2) in male HGs, UCP3 was comparable to female levels at 30 days post-castration (when the type II-cell percentage had fallen from 50 to 5%), although testosterone was already near zero at 15 days (when neither the type II-cell percentage nor the UCP3 level had fallen), and testosterone-replacement therapy prevented these changes. Testosterone-treated females possessed type II cells and a UCP3 level about twofold higher than in control females. Males displayed more intense UCP3 immunohistochemical positivity in type I HG cells than females. Hence, testosterone may indirectly control UCP3 expression by regulating the gland's morphological and lipid dimorphism. Straight-chain fatty acids [found in alkyl diacylglycerols (ADGs) in males] are oxidized predominantly in mitochondria, branched-chain fatty acids (abundant in ADGs in females) predominantly in peroxisomes, so we speculate that the higher UCP3 expression in males reflects greater fatty acid flux in HG mitochondria. This is supported by our finding that in female (not male) HGs, the peroxisome-rich fraction contained α-methylacyl-CoA racemase (AMACR), an enzyme important in the β-oxidation of branched-chain fatty acids. Two groups of animals were maintained in long days (14 hr light/day). Hamsters in one group were left intact and those in the other were castrated. Another three groups were maintained in short days (8 hr light/day); these animals were either left intact, castrated, or both castrated and pinealectomized. The duration of the experiment was 5 weeks. Castration of long photoperiod-exposed animals resulted in a significant drop in the number of Type II cells and a large increase in the porphyrin deposits (P < 0. 01). However, castrated animals exposed to short photoperiod showed a significant smaller change in both parameters compared with those exposed to long days (P < 0. 05). Pinealectomy prevented the effects of short days in castrated animals. No significant changes were observed in the relative number of mitotic figures or in the number of cell nuclei, indicating that the changes observed were due in part to a transformation of Type II into Type I cells. In a second experiment, male hamsters were injected daily either with 25 μg of melatonin late in the afternoon or with the saline for 8 weeks. The administration of melatonin resulted in a significant (P < 0. 05) increase in the percentage of Type II cells. Effects of δ-aminolevulinic acid (ALA) and melatonin were investigated in the female Syrian hamster Harderian gland. This is an organ physiologically exposed to strong oxidative stress due to the highest porphyrinogenic rates known in nature. Enzyme activities of porphyrin biosynthesis and of antioxidative protection, oxidative protein modification, and histological integrity were studied. In the porphyrin biosynthetic pathway, ALA and melatonin acted synergistically by downregulating ALA synthase (ALA-S) and stimulating product formation from ALA; the combination of ALA and melatonin suppressed ALA-S activity, down to about 1% of that in controls. While ALA effects on porphyrinogenesis can be interpreted in terms of homeostasis, melatonin’s actions may be seen in relation to seasonality and/or reduction of oxidative stress. Among antioxidant enzymes, superoxide dismutase (SOD) and glutathione reductase (GR) activities were diminished by ALA, presumably due to the vulnerability of their active centers to free radicals, whereas melatonin moderately increased SOD. Both ALA and melatonin strongly stimulated catalase (CAT), thereby counteracting the oxidative stress induced by ALA and its metabolites. Nevertheless, exogenous ALA caused a strong net rise in protein carbonyl and considerable damage of tissue. When given together with ALA, melatonin antagonized these effects and largely protected the integrity of glandular structures.

To examine the reliability of quantitative positron emission tomography studies in the rat (Rat–PET), we assessed the influence of radioactivity accumulated in the Harderian glands on PET CMRglc determination. We measured CMRglc by PET and ex vivo dissection methods by using 2-[18F]fluoro-2-deoxy-D-glucose in rats with and without focal brain ischemia. The CMRglc values obtained by PET, after correcting with recovery coefficients, were higher than those measured by the ex vivo method at rostral slices, and reduction of the CMRglc in the ischemic brain was not demonstrated by PET in the frontal cortex. The radioactivity accumulated in the Harderian glands prevents the quantitative determination of CMRglc using Rat–PET.

Monoamine oxidase (MAO) and alcohol dehydrogenase (AD) activities were studied histochemically in the Syrian hamster Harderian gland using tryptamine as substrate and Nitroblue Tetrazolium as the final electron acceptor. No dark: light-related changes were observed. Male type I secretory cells showed an intense MAO reaction. Female type I cells exhibited a moderate MAO activity. Both male and female glands showed a moderate/intense AD-positive reaction. Male type II cells were lacking MAO and AD activities. MAO activity found in the hamster Harderian glands corresponded mainly to MAO type A since treatment with chlorgyline (0.01, 0.1 and 0.5mm) totally inhibited it. The possible role of these two enzymes in Harderian gland indolalkylamine metabolism is discussed.

The Harderian gland (HG), a sero-mucous secreting organ in the eye orbit, has long been recognized as immunologically important in chickens. During experimentation to characterize immune components of the gland, proteomics analysis revealed the presence of hematopoietic prostaglandin D synthase (H-PGDS). Extraction of total RNA followed by RT-PCR produced cDNA of 597 base pairs. DNA sequencing revealed nucleic acid and predicted amino acid sequences that were 99% aligned with the one published sequence for chicken H-PGDS of the spleen. Alignment with murine, rat, and human H-PGDS were 69, 69, and 66%, respectively. Ocular vaccination of chickens with a Newcastle Disease/Infectious Bronchitis vaccine (Mass.-Ark. Strain) induced an increase in H-PGDS expression determined by real-time PCR. Newcastle disease virus, in its most pathogenic form, threatens the livelihood of rural poultry farmers where there is a limited infrastructure and service for vaccinations to prevent outbreaks of the virus. Previously reported studies on the host response to Newcastle disease in chickens have not examined the disease under abiotic stressors, such as heat, which commonly experienced by chickens in regions such as Africa. The objective of this study was to elucidate the underlying biological mechanisms that contribute to disease resistance in chickens to the Newcastle disease virus while under the effects of heat stress.
