Molecular cloning

The MspI restriction-modification system, which recognizes the sequence 5'-CCGG-3', has been previously cloned and sequenced (1). We subcloned the methyltransferase gene (M.MspI) downstream of the ptac promoter in the multicopy vector pUC119 and overexpressed it in E. coli. Upon induction with IPTG, M.MspI constitutes more than 10% of cellular protein. A scheme has been devised to purify large amounts of biologically active M.MspI to apparent homogeneity from these overexpressing E. coli cells. Approximately 0.8 mg of pure M.MspI per gram of cells (wet weight) can be obtained. The apparent molecular weight of M.MspI is 49 kD, by SDS gel electrophoresis and 48-54 kD by gel filtration. In order to identify immunoreactive Bartonella henselae proteins, B. henselae antiserum from an experimentally infected cat was used to screen a B. henselae genomic DNA expression library. One immunoreactive phage clone contained a gene (p26) with significant nucleotide identity with orthologs in brucellae, bartonellae, and several plant-associated bacteria. p26 gene sequences from four B. henselae strains, one B. koehlerae strain, and one B. clarridgeiae strain were cloned. Comparative nucleotide sequence analysis showed that p26 is a potential marker for molecular diagnosis of infection, as well as for identification to species level and genotyping of Bartonella sp. isolates. Alignment of the predicted amino acid sequences illustrated conserved putative protein features including a hydrophobic transmembrane region, a peptide cleavage site, and four dominant antigenic sites. Expression of p26 in Escherichia coli produced two proteins (26 and 27.5 kDa), both of which were reactive with feline anti-B. henselae antisera. Furthermore, murine hyperimmune serum raised against either recombinant protein reacted with both proteins. No reactivity to either recombinant protein was detected in nonimmune serum, and reactivity persisted as long as 20 weeks for one cat. The p26 protein product is an immunodominant antigen that is expressed during infection in cats as a preprotein and is subsequently cleaved to form mature P26.

We used overlapping and nested homozygous deletions, contig building, genomic sequencing, and physical and transcript mapping to further define a approximately 630-kb lung cancer homozygous deletion region harboring one or more tumor suppressor genes (TSGs) on chromosome 3p21.3. This location was identified through somatic genetic mapping in tumors, cancer cell lines, and premalignant lesions of the lung and breast, including the discovery of several homozygous deletions. The combination of molecular manual methods and computational predictions permitted us to detect, isolate, characterize, and annotate a set of 25 genes that likely constitute the complete set of protein-coding genes residing in this approximately 630-kb sequence. Xenopus liver contains a protein inhibitor of glucokinase that, in contrast to the mammalian regulatory protein of glucokinase, is insensitive to fructose 6-phosphate and fructose 1-phosphate [Vandercammen A. & Van Schaftingen, E. SUMMARY In tetrapods, arginine vasopressin and its counterpart, arginine vasotocin(AVT), are involved in renal water conservation through vascular V1a-type and tubular V2-type receptors, and only the former has thus far been cloned in fish. We successfully cloned the V1a-type and V2-type AVT receptor from the kidney of the African lungfish, Protopterus annectens, and the deduced amino acid sequences exhibited high homology with amphibian V1a- and V2-type receptors, respectively. Functional analysis showed that AVT addition to CHO cells transfected with lungfish V1a-type receptor increased[Ca2+]i in a concentration-dependent manner, whereas CHO cells transfected with lungfish V2-type receptor responded with cAMP accumulation after AVT stimulation. Objective
Proteases are among the most important industrial enzymes. Microbial proteases, especially from Bacillus sp., are most widely exploited industrially. The aim of this study was isolation, cloning, sequencing, expression, and bioinformatics study of yyxA serine protease gene extracted from Bacillus licheniformis.

Materials and methods
In this study, after extraction of bacterial DNA, the yyxA serine protease gene was isolated from Bacillus licheniformis using the polymerase chain reaction technique and cloned into the pTG19-T vector. The molecular structure, its biochemical and phylogenetic properties were investigated. The three-dimensional structure of the cloned enzyme was predicted using the I-TASSER, PHYRE2, RAPTORX, and Modeller tools. Confirmation of yyxA gene expression was performed by SDS-PAGE and dot blot analysis.

Results
Cloning was confirmed by sequencing. Based on the results of phylogenetic studies, the obtained protein sequence showed high similarity to the sequences of other Bacillus species, such as B. subtilis, B. gobiensis, and B. pumilus. After evaluating the drawn models, it was found that the models provided by PHYRE2 and I-TASSER software were desirable ones for predicting the three-dimensional structure of this protease. Recombinant protein production was successfully induced by IPTG induction in the host containing the plasmid pET28a-yyxA. Optimization of recombinant protein production was investigated. The highest expression values were obtained at 37 ° C for 4 hours with one mM IPTG.

Conclusions
The nucleotide sequence of the yyxA gene is 1212 nucleotides long, encoding a protein with 403 amino acids. Studies have shown that the enzyme encoded by this gene is in the category of stable enzyme and will be expressed in solution in Escherichia coli. These advantages make the enzyme as a suitable candidate for use in industry.
