This randomized, double-blind clinical trial was conducted over 16 weeks to assess the efficacy and safety profile of intra-articular polynucleotides gel injections in the treatment of knee osteoarthritis associated with persistent knee pain. 60 patients were enrolled and randomized to receive intra-articular polynucleotides (n = 30) or hyaluronan (n = 30); patients received five weekly intra-articular knee injections and the follow-up period was 3 months after the end of treatment. Primary endpoint was to determine polynucleotides (PN) efficacy in reducing knee pain at the end of the study, over baseline value and over standard hyaluronan viscosupplementation (HA). Pain levels were measured using a 0–10 cm Visual Analogue Scale (VAS). Interactions of cationic tetrakis (N, N′, N″, N‴- tetramethyltetra-3, 4-pyridinoporphyrazinatozinc (II) (Zn (tmtppa)) with synthetic polynucleotides, poly (G-C) and poly (A-T), and calf thymus DNA have been characterized in 7.5 mM phosphate buffer of pH 7.2 by UV-Vis absorption and fluorescence spectroscopy. The appearance of hypochromicity more than 30% in UV-Vis spectra of porphyrazine due to interaction of both poly (G-C) and poly (A-T) indicates interaction similar to that of porphyrazine with DNA.The binding constants were determined from the changes in the Q-band maximum of the porphyrazine spectra at various poly (G-C) and DNA concentrations. We provide a brief overview of the structural characteristics of the main groups of conjugated polyelectrolytes (CPEs) as well as the methods of synthesis and their behaviour in solution. Their tendency to form aggregates in solution, which is one of the key points to be taken into account for them to be used in polynucleotide sensing, is also considered and the various strategies adopted to avoid it will be discussed. These include the synthetic one (with the incorporation of charged and/or bulky substituents), the use of organic co-solvents and the addition of surfactants. The main physical chemical changes (optical, photophysical, electrical conductivity and viscosity) observed upon direct binding between polynucleotide and CPE, the kind of interactions involved and their applicability in sensing are considered as a function of the CPE structural rigidity. Moreover, more complex devices developed in CPE-polynucleotide sensing with the involvement of additional spectroscopic probes to induce Förster resonant energy transfer processes (FRET) or superquenching phenomena are reviewed. Finally, the main CPE applications in biosensing and the potential use of these systems in understanding DNA compaction and possible extension to the construction of supramolecular oligonucleotide structures are summarized.

Over 100 regulatory mutations linked to the histidine (his) operon of S. typhimurium have been isolated. They all map in a region estimated to be several hundred base pairs in length located at one end of the his operon ("the hisO region"). The mutations are located at sixteen recombinationally separable sites or are deletions encompassing several sites. Data obtained from pairs of reciprocal three-point tests show that "constitutive" (high enzyme levels) and "promoter-like" (low enzyme levels) hisO mutations are interspersed on the genetic map. In a few crosses, recombination was not observed to occur between markers shown to occupy different sites based on behavior in other recombination tests.

There is an urgent need for more efficient viral vaccines in finfish aquaculture worldwide. Here, we report the use of poly(I:C) stabilized with chitosan as an adjuvant for development of better finfish vaccines. The adjuvant was co-injected with inactivated viral hemorrhagic septicemia virus (VHSV) (CSpIC+iV vaccine) in adult zebrafish and its efficiency in protection against VHSV infection was compared to a live, attenuated VHS virus vaccine (aV). Both free and stabilized poly(I:C) were strong inducers of an antiviral state, measured by transcriptional activation of the genes of viral sensors: toll-like receptors, interferons, and interferon-stimulated genes, such as MXa within 48 h after injection. Polynucleotides (PN) have long been known as an effective supportive therapy for wound healing. The present study investigated whether a hydrogel formulation containing PN and hyaluronic acid (PN + HA) could promote wound healing in an in vitro model of gingival fibroblasts. PN promoted cell growth and viability as assessed by different assays, and PN + HA, though not significantly further increasing cell growth as compared to PN, supported the formation of dense multilayered cell nodules. PN promoted fibroblasts’ clonogenic efficiency and PN + HA further enhanced the formation of more numerous dense colonies. PN + HA appeared to significantly increase the expression of collagen 1a1 and 3a1, while not affecting proteoglycans deposition. Interestingly, when tested in a scratch assay, PN + HA achieved gap closure after 48 h, while cells in the comparison groups had not completely bridged the scratch even after 96 h. Impaired γ-aminobutyric acid (GABA) signaling may contribute to the emergence of cognitive deficits and subcortical dopaminergic hyperactivity in patients with schizophrenia and related psychotic disorders. This review is a comprehensive analysis of the progress made so far on the delivery of polynucleotide-based therapeutics to the eye, using synthetic nanocarriers. Attention has been addressed to the capacity of different nanocarriers for the specific delivery of polynucleotides to both, the anterior and posterior segments of the eye, with emphasis on their ability to (i) improve the transport of polynucleotides across the different eye barriers; (ii) promote their intracellular penetration into the target cells; (iii) protect them against degradation and, (iv) deliver them in a long-term fashion way. Overall, the conclusion is that despite the advantages that nanotechnology may offer to the area of ocular polynucleotide-based therapies (especially AS-ODN and siRNA delivery), the knowledge disclosed so far is still limited. This fact underlines the necessity of more fundamental and product-oriented research for making the way of the said nanotherapies towards clinical translation.

ABSTRACT A THz wave based approach for the label-free characterization of genetic material is presented. Time-resolved THz spectroscopic analysis of genetic sequences (polynucleotides) demonstrate a clearly distinct complex refractive index in the THz frequency range as a function of the hybridization state (hybridized/denatured) of the analysed DNA sequences. By monitoring THz signals one can thus infer the binding state of oligo- and polynucleotides, enabling the label-free determination of the genetic composition of polynucleotides. Recent advances in the development of high sensitivity integrated THz sensors reaching femtomol detection levels and single base mutation detection capabilities are reviewed.

HIV-1 reverse transcriptase (RT) possesses both DNA polymerase activity and RNase H activity that act in concert to convert single-stranded RNA of the viral genome to double-stranded DNA that is then integrated into the DNA of the infected cell. Reverse transcriptase-catalyzed reverse transcription critically relies on the proper generation of a polypurine tract (PPT) primer. However, the mechanism of PPT primer generation and the features of the PPT sequence that are critical for its recognition by HIV-1 RT remain unclear. Here, we used a chemical cross-linking method together with molecular dynamics simulations and single-molecule assays to study the mechanism of PPT primer generation. Efficient intercalation of a luminescent Ir(III) complex exclusively made of polypyridine ligands in natural and synthetic biopolymers is reported for the first time. The emission of the complex is largely enhanced in the presence of [poly(dA-dT)(2)] and strongly quenched in the presence of [poly(dG-dC)(2)]. By comparing the emission decays in DNA and in synthetic polynucleotides, it is proposed that the emission quenching of the title compound by guanine residues in DNA is no longer effective over a distance of four dA-dT base pairs.
