Experiment No. 2 PH and Buffer

The document describes an experiment that measured the pH of various household samples using pH paper and a pH meter. Key results included: - Fresh milk had a pH of 6.5-6.7 by pH paper and 6.4 by pH meter. - Urine had a pH of 6 by pH paper and 5.3 by pH meter. - Vinegar had a pH of 2.5 by pH paper and 0.1 by pH meter. The document also discusses the importance of pH for biological processes and defines buffers as solutions that resist changes in pH when small amounts of acid or base are added.

C. BUFFERS

II. Questions for Research

1. What is the normal pH of blood? Milk? Urine? Tabulate your answer.

2. What is the biochemical relevance of pH?

 pH is biologically important because it affects the structure and activity of macro-
molecules. pH is important in homeostatic processes. For example, most animals
breathe not because they lack oxygen, but because CO2 buildup in the blood increases
the blood acidity beyond normal levels.
3. Which sample in procedure B exhibited buffer action? Why?
 Sodium acetate, because buffer solutions resist a change in pH when small amounts of a
strong acid or a strong base are added. A solution of acetic acid and sodium acetate is
an example of a buffer that consists of a weak acid and its salt.
4. Compare the buffer systems in whole blood and blood plasma. Tabulate your answer

5. Give the Henderson-Hasselbach equation and give its importance in the study of
buffers

 The Henderson-Hasselbalch equation is useful for estimating the pH of a buffer solution and
finding the equilibrium pH in an acid-base reaction. The equation can be used to determine
the amount of acid and conjugate base needed to make a buffer solution of a certain pH.
