Introduction
CYANStart is an easy to operate semi-automatic biochemistry analyzer. This new instrument was designed
to have powerful features in a compact unit.

1. Description of the analyzer

The CYANStart technical features are:

 Open system
 200 programmable tests.
 Memory for up to 1000 results.
 Integrated pump and flow cell.
 Internal pre-adjusted Peltier element at 25°C, 30°C, 37°C.
 Inbuilt printer

2 Installation
2.1 Packing contents
When you receive your CYANStart biochemistry analyzer, open the shipping box and check the contents
against the following packing list:

a) CYANStart
b) Power supply cable
c) Two fuses
d) Waste bottle and silicone tube
e) One paper roll
f) Instruments cover lining
g) One lamp
h) Push button
i) Peristaltic tube
j) Dust cover

If the instrument is damaged or items are missing, contact your local distributor/importer immediately for
assistance.
OPERATORS SHOULD READ THE MANUAL BEFORE USE. PLEASE ALSO BE REMINDED OF
CHECKING THAT THE TUBING’S (PUMP AND WASTE TUBE) ARE CONNECTED, THAT THE WASTE
BASKET IS CONNECTED, THE PAPER INSERTED AND THAT THE CORRECT POWER SOURCE (110-
230V) IS SELECTED.

2.4 Location of the instrument


The instrument should be located in a clean environment, placed on a stable surface and away from direct
sunlight, which could affect the operating temperature and the quantity of light measured by the instrument.

The following points should be taken into consideration:

 Ensure that the surface is horizontal.


 Avoid positions subject to jerks or vibrations.
 Make sure that the instrument is not placed close to air conditioning or heat sources.
 Indicated temperature:
 For storage: 5°C- 35°C.
 For use: 15°C -25°C.
 See point 8.1 Punctual Maintenance for more details

2.5 Power supply connection


Notice: please check if the setting of the voltage switch is according to your country’s electrical network.

Procedure:
1. Connect the power plug to a grounded AC wall outlet, preferably one that is not shared
with other electric appliances and with low fluctuation of line voltage compared to the
standard voltage specified. (the use of an electrical stabilizer)
2. Keep the instrument away from other appliances that generate high frequency electrical
noises (e.g. radiological instruments).
3. Before connecting the power cord, check that the AC power supply corresponds to the value
that is stated on the voltage switch.
4. Make sure that your AC main line has an efficient ground line. A bad ground line connection
may compromise analysis results and damage the instrument.

2.6 Tube setting

The peristaltic pump is situated on the left side of the instrument. During transport, the peristaltic tube is
disconnected to avoid any damage as indicated below.

Place the tube (1) around the peristaltic pump (2) and make sure that the white plastic connections are set
as shown in the figure below:

Make sure that the transparent tube, connecting the flow-cell with the internal hydraulic circuit is not twisted
or squashed.
At installation, the Inlet pipe (1) and the Push button (2) have to be placed as indicated on the picture below.

Under the measurement block, there is a lever (the PUSH button). Press the lever for no more than half a
second to pump samples into the flow cell.

2.7 Measurement block

Open the measurement block cover lid, positioned on the top left part of the instrument.
Inside you will see:
1. Cover lid
2. Lamp
3. Keyboard (to the right)
4. Flowcell
5. Inlet pipe

2.8 Changing the paper


Procedure:
1. Switch on the instrument.
2. Open the printer cover and locate the blue
lever.
3. Raise the little blue lever located on the right
side of the printer.
4. Take a new paper roll and pass the edge of
the paper under the black rubber cylinder.
5. Pull the lever down and press Feed till the
paper is 4 cm long
6. Pass the edge of the paper through the
aperture and place the printer cover in its
proper position.

2.9 Caution
Procedure:

1. After turning on the instrument, pay attention not to spill liquids or micro solid substances
on the surface around the instrument.
2. Keep the instrument away from young children.

If the above procedures are carefully followed, it will be possible to TURN ON the instrument by
using the switch located on the polysnap block.

3 Technical control of the CYANStart
3.1 General description and Block Diagram
The motherboard of the CYANStart contains the circuits which carry out most functions of the instrument,
except the printer interface, that is handled by a separate board.

The mother board supplies and controls all instrument’s devices and sensors. This is given in the following Block
Diagram. The location of the different function blocks is given on figure1

3.2 Motherboard
On figure 2, the connections of the different functions are given. More explanation will be given in the
following sections.

3.3 Power supply
The instrument is connected to the main power supply (220 or 110 from a select switch) by a cable with two
plugs; one side of the cable must be inserted into a main socket, the other side in the socket of the
polysnap.

In this polysnap two fuses are located for each phase, with a switch for line and neutral. The power
supply is then leaded to the switching power supply for delivering the right tension in the connector J1
(refer to check points for the tension values, figure 2 and figure 3).

Replacement of the fuses:


When the power supply fuses are burned, it is necessary to replace them with a new one by following this
procedure:
1. Turn off power supply, unplug power supply line
2. Locate the fuse on the rear panel and replace.
3. Under the power supply socket, a second fuse is located. Put out the fuse and replace
with a new one.

3.4 Printer with external board


The operating voltage range for driving the head and motor of the printer is 5.5V to 9.5V. Under the printer,
there is an interface board which contains the control circuit. This operating voltage range is 4.5 V to 5.5 V.
This circuit performs a software and hardware control to detect abnormal temperatures, the position of the
head-up (when the head is in the upper position, the circuit does not activate the heat element) and to detect
when the paper runs out.

3.5 Pump Motor


Perform the preliminary procedure, in order to access to the peristaltic pump tube.
See the figure below for a schematic view of the pump:

3.5.1 Replacement of the pump tube.

To change the tube, disconnect the elastic tube from its 2 adapters by simply pulling them out. Then you can
replace the tube and reconnect the 2 adapters.

Before closing the cover again, be sure that the peristaltic pump elastic tube has a suitable strength. Check this
by aspirating some distilled water and see if there is any drop formation at the intake tip outside the instrument.
No drops must be present.

No drops present!

If you have still some drops, despite already having changed the tube, shorten the tube in order to increase the
strength of the elastic tube. Never shorten the tube if it is not necessary. This will reduce the lifetime of the
peristaltic pump tube.

NOTE: You must recalibrate the peristaltic pump aspiration volume after having changed the pump tube.

3.5.2 Calibration of the pump

This menu enables the user to calibrate the pump. It is advised to calibrate the pump regularly to minimize
bias and cross over. The peristaltic pump is moved by a stepper motor which permits to intake the volume
of sample with high level of precision.

To open the calibration menu:


1. Switch on the instrument with the main power switch located on the rear side of the
instrument.
2. The first screen will display the basic information on the model (name and software version)
3. Press ENTER to proceed. The following display will be shown:

4. Use the UP and DOWN arrows to choose the submenu “Pump Calibrate” and press ENTER
to select it. The following screen will appear.

Calibration Procedure:

NOTE: For a higher precision we advice to use CYANPipettes. Ask your distributor for more
information about the CYAN product range.

1. Select the Pump calibration program.


2. Enter 5000 as the volume that will be aspirated and press ENTER
3. The number of steps registered during the last calibration procedure is shown below.
4. Put the aspiration spout into a tube with exactly 5000µl of distilled water.
5. Press the push button. The peristaltic pump will begin to work.
6. When the target quantity has been aspirated (5000µL) press immediately the push button. The
LCD display shows the number of steps performed by the motor. This value is stored and is
used as reference for the instrument to intake any quantity of sample.
If the obtained value highly differs from the one of the last calibration procedure (see point 3),
something went wrong. Please repeat the procedure.
The optimal value is 8000-15000.
7. Press ESC to return to main menu.

3.6 Optical System
3.6.1 General description

The following figure illustrates the different parts of the optic system. In the next sections, these areas will be
discussed.

3.6.2 Lamp

3.6.2.1 Checking the lamp

The autozero menu enables the user to check the performance of the lamp. It is advised to check the
autozero regularly.

To open the autozero menu:

1. Switch on: Start up the instrument from the main power switch located on the rear side of the
instrument.
2. The first screen will display the basic information on the model (name and software version)

5. Aspirate distilled water by pressing the PUSH button. The instrument will begin to zero afterwards.
The first column is the filter wavelength expressed in nm. The second column is the signal-amplified
coefficient (= gain). The third column is the AD signal amplified coefficient. The last column is the
offset-value.

Generally, the gain will be between 2 and 7. The AD initial value is between 26000 and 38000. Offset is
between 1 and 240.

The best results are obtained when:


 The amplified coefficient (gain) is between 3 and 5
 A/D value is between 30000 and 36000
 Offset value is between 15 and 150 AD signal value.

If the results of all the filters are out of range, please check the filter wheel sensor before replacing
the lamp (paragraph 3.7.2). If only one or a few filters are out of range, check the filter wheel sensor
first, then check if those specific filters are broken or damaged (paragraph 3.7.3).

Notice:
1. Do not forget to aspirate distilled water before effectuating the test.
2. The lamp and its socket are already centered. Never touch the lamp with your fingers.

3.6.2.2 Changing lamp procedure

When the lamp is damaged, it is necessary to replace it with a new one by following this procedure:

1. Turn off power supply, unplug power supply line


2. Open the instrument cover
3. Disassemble cuvette tray
4. Unscrew the screw of the lamp socket and disconnect the black and the brown wire on
the power connection board.

5. Install the new lamp and connect the black and brown wire on the power connection
board.
6. Adjust the position of the lamp according to paragraph 3.6.2.3.
7. Check the new lamp according to paragraph 3.6.2.1.
8. If the results are out of range, follow the instructions given in paragraph 3.6.2.4.

3.6.2.3 Adjustment of the position of the lamp

Check the position of the lamp using the following procedure:

1. Open the menu ‘AD value check’ and press ‘1’, the software will select the filter of 340 nm.
2. Optimize the position of the lamp by unscrewing the screw indicated on panel A and pushing
the lamp up or down (see panel B). The optimal position is the position indicating the highest
AD value.
3. If the lamp is properly positioned, fasten the screw.

3.6.2.4 Adjustment of the voltage of the optical system

ATTENTION: This procedure can only be used in case of a new lamp replacement and should not be
used to increase the signal of older lamps!

When replacing a new lamp it is very important to check (and if necessary to adjust) the voltage of the optical
system. Only if the whole optical system auto zero values are too high or too low, it is necessary to
adjust the voltage of the test board.

Please follow the below procedure to adjust the gain and offset value of the new lamp:
1. Adjust the offset value till the value for the 340 nm filter is within the correct range (see below).
2. Subsequently adjust the gain until the filter having the highest AD value is within the correct range
(see below).

Offset adjustment
Offset value refers to the voltage of the circuit when the preamplifier doesn’t receive any signal (= dark filter).
Since we regard it as a starting point of zeroing, the value cannot be zero or negative, neither it can be to
high. The offset value should be between 1 and 240. The best results are obtained when the offset signal
lies between 15 and 150. To decrease the offset value, turn the trimmer screw on the right site of the
adjustment block clockwise. To increase the offset value, turn the trimmer screw counterclockwise. Check by
rerunning the AD autozero.

Preamplifier(test board)voltage adjustment (gain)


As for the whole optical system, the preamplifier has the function of receiving and magnifying the test signal.
In other words, the preamplifier adjusts the voltage of whole optical system. When the auto zero value of the
entire optical system is too high or too low, it is necessary to adjust the voltage of preamplifier. To decrease
the A/D value, turn the trimmer screw on the left site of the adjustment block clockwise. To increase the A/D
value, turn the screw counterclockwise. Check the adjustment by rerunning the AD autozero.

3.7 Filters and filter wheel
3.7.1 General description

Filter wheel assembling


The following table describes the filter wheel assembling. The first column indicates the location of the
different filters on the filter wheel. The second column gives the filter number, programmed in the software.
The fourth column indicates whether the specific wavelength has a pre-filter or not. In the fifth column, the
diaphragm of the pre-filter is given.

3.7.2 Checking filter wheel sensor procedure

This procedure must be followed when all filters are out of range during an autozero reading (according to
paragraph 3.6.2.1). Only this way, the movement of the filter wheel and the correct positions of the filters can
be checked.

Procedure:
1. Switch on: Start up the instrument from the main power switch located on the rear side of the
instrument.
2. The first screen will display the basic information on the model (name and software version)
3. Press ENTER to proceed. The following display will be shown:

5. Remove the flow cell carefully.
6. Place a white paper of 1x5 cm in the reading well (as shown on the picture).
7. Select the filter by filter by using the Right and Left keys or by pressing the number of the Filter and
check the color on the paper.

If the colors above are not correct, please change the filter wheel sensor and check the filter wheel
movement again.

3.7.3 Checking filter performance

This procedure is used to check the performance of a specific filter. It must be executed when only one or
few auto zero values are out of range.

Procedure:
1. Switch on: Start up the instrument from the main power switch located on the rear side of the
instrument.
2. The first screen will display the basic information on the model (name and software version).
3. Press ENTER to proceed. The following display will be shown:

5. Aspirate distilled water.
6. Select filter by filter using the Right and Left keys or by pressing the number of the Filter. After
selecting the correct filter, press the DOWN arrow
7. Put the gain value on 3 by pressing the UP arrow and check the AD-value of the selected filter.

The best results are obtained when:


 The amplified coefficient (gain) is equal to 3 or 4
 The A/D value is between 30000 and 38000

If the criteria above are not achieved for one of the filters, it is necessary to adjust this filter by changing the
pre-filter. This procedure is described in paragraph 3.7.4.

3.7.4 Changing the pre-filter

As given in the filter wheel assembling table, most of the filters in the CYANStart have a pre-filter. This is a
non-selective absorbent with a central hole with dimensions from 0.2 mm to 2.5 mm. The pre-filter will
decrease the light intensity of the light source before it passes the wavelength filter. By choosing a different
diaphragm for each filter, the different light intensity is equalized for all the filters. Because of its low light
intensity, 340nm has no pre-filter. This wavelength is thus regarded as the starting point of zeroing. By
replacing the pre-filter of a specific wavelength, it is possible to equalize its light intensity again with the other
filters in order to guarantee the accuracy of all test results.

Pre-filter replacement procedure:


1. Take out the filter locking wreath with forceps. As showed in figure 2.1.2-2.
2. Take out the filter and pre-filter (light barrier). You can use a right angle wrench to raise them behind
filter wheel. As showed in figure 2.1.2-3.
3. After taking out the pre-filter, filter and locking wreath one by one, you can enlarge the diaphragm
hole of the pre-filter by placing another pre-filter.
4. Put every piece back in the correct order and check the AD-value again at gain= 3 according to
paragraph 3.7.3.

3.8. Adjustment of the temperature of the flow cell
This procedure will enable you to adjust the temperature of the flow cell, in case of problem with warming up
or with stability of the temperature.

A. Checking the temperature:

There are two ways to check the temperature of the flow cell:
1. Using the software under the “Temp Display” menu
2. Measuring the real temperature with a thermocouple or mercury thermometer.
Both ways are described hereunder. Please check the two procedures, to detect the cause of the heating
problem and to solve it.

Procedure A1: Using the software under the “Temp Display” menu
- Open the “system setup” menu in the main menu, then open the “Temp Display” menu.

- The temperature of the flow cell can be read next to Cuvette. The real measured temperature by the
instrument (not the selected temperature) is expressed in degrees Celsius. It should be approximately the
same as given in the settings column. Please disregard the Incubator reading. This function is already
included in the hardware design for future options.

Procedure A2: Measuring the real temperature with a thermocouple or mercury thermometer.

Insert a thermocouple thermometer into the flow cell, and then switch on the analyzer. Next picture shows the
thermocouple thermometer. It should be filled with water.

NOTE:

1. Be sure that your thermocouple thermometer is calibrated and with 0.1 °C of accuracy in
measurements. If you set temperature at a wrong value Kinetic analysis results will be mistaken.
2. If you use mercury thermometer you should insert it in a cuvette filled with water and NOT
DIRECTLY INTO THE INCUBATOR. You need 0.1 °C of accuracy in measurements. You need
more time to reach a stable temperature with a mercury thermometer, since the water in the
cuvette needs to be warmed up (typically the waiting time is doubled, i.e. 1h.).

NOTE: The table above can only be used in case of the REAL temperature value (measured with a
thermocouple or thermometer) deviates only a few degrees from the triggered value.

B. Adjustment of the actual temperature in the flow cell and of the displayed temperature

Procedure B1: Adjusting the actual temperature of the flow cell:


Put a thermometer into the flow cell then adjust the adjustable resistor. Clockwise rotation increases
the temperature, counterclockwise rotation decreases the temperature. Keep the temperature on
37.5°C.

Adjust the temperature as below:

Procedure B2: Recalibration of the displayed value:


After the actual temperature is stabilized, check the temperature calibration of the instrument and
adjust the displayed temperature if necessary.

To adjust the displayed temperature, follow the instructions below:

4. Maintenance
The CYANStart requires three levels of maintenance:
 The punctual maintenance: at special moments such as at installation
 The daily maintenance: to maintain the level of precision day in day out

4.1 Punctual Maintenance


The punctual maintenance should be effectuated at installation, before or after a long period of duty.
The following points of attention should be considered:

 Selecting the right voltage (110 or 220V)


 The waste bottle is connected to the instrument
 No visual signs of leakage of the tubes
 Remove dust from the instrument
 Wash the flow cell 10 times with distilled water
 Wash the flow cell with CYANPlus Washing Solution (code CY001-WS) and flush the flow cell for 2
minutes with distilled water.
 In case the instrument will not be used for a long period, remove all liquid inside the instrument by
aspirating air.

4.2 Routine maintenance


4.2.1 Daily washing

At the end of each working session, it is recommended to rinse the instrument with a neutral detergent
solution such as CYANPlus Washing Solution (code CY001-WS) or Tween 20 (5%) via the inlet pipe. Place
a cup of detergent solution under the inlet pipe and carry-out the Washing procedure.

After using the detergent solution, rinse the flow cell abundantly with water. After the washing, it is necessary
to repeat the operation with air so that the water remaining inside the instrument can be expelled completely.
The machine should be dry when turning it off to avoid microbiological contamination.

For normal flow cell washing during work (between contaminated samples or between 2 methods), only use
distilled water. In case of bubbles, use sodium hypochlorite (5 to 10%) and subsequently rinse with plenty of
water, followed by air. This sodium hypochlorite is normally market available. Please use only good
trademarks. Cheap ones contain contaminating solutions.

NOTE: Always dilute the sodium hypochlorite for 5 to 10%! Never use it undiluted!!! This will damage
the instrument!

4.2.2 Weekly maintenance

The Weekly maintenance is also a flow cell cleaning. After users finish a week of work, use distilled water to
wash the flow cell repeatedly, and then clean it with detergent. Pay attention that detergent should remain in
flow cell for about 5-10 minutes, then empty and finally wash repeatedly with distilled water and repeatedly
with air.
The following washing solutions are recommended:
 CYANPlus Washing Solution (code CY001-WS)
 Tween 20 (5%)
 5-10% NaClO

4.3 Periodic Maintenance


4.3.1 Monthly

Every month repeat at least 2 times the washing procedure with sodium hypochlorite (5-10%)
following the procedure of weekly maintenance.

N.B.: Do not use corrosive detergents to wash the instrument.

After the washing, it is necessary to repeat the operation with plenty of distilled water (repeat 5 times),
followed by 5 times with air so that the water remaining inside the instrument can be expelled completely.

Clean the external surface of the instrument every month with a non-abrasive detergent. Clean the surface
near the two fans every month in order to take off dangerous dust.

4.3.2 Special cleaning

Please notice that the instrument, especially the optical system and the electrical circuit, are sensitive to
dust. Avoid using the instrument in dusty spaces. Cover the instrument when not in use and keep the printer
and flow cell cover closed during use.

In dusty places, we advice to clean the dust from inside the instrument. To do so, remove the cover and blow
the dust off the instrument. Open the filter cover and blow the dust off. Finally aspirate inside the instrument
with a vacuum cleaner.

Notice: the instrument cover should only be removed when the CYANStart is unplugged.

We strongly advice to leave this procedure to the technical assistance of the service engineer of your
distributor.

If the instrument is not being used for a long period of time, empty the hydraulic circuit and disconnect the
tubes.

4.3.3 Replacing Spare Parts

Some consumables should be replaced after a fixed period of time to ensure the good performance of the
analyzer. Please check the table here below:

5. Troubleshooting

- The pipette for the manual handling has been correctly calibrated and
the tips are compatible.

2.Check if:
- The peristaltic pump pipe has been connected correctly
- The peristaltic pump is leaking
- The pump is aspirating normally (check the volume)

3.Check if:
- The lamp is stable (procedure auto zero paragraph 3.6.2.1)
- The filter wheel sensor is working (procedure 3.7.2)
- The temperature of the flow cell

Bad accuracy of the results 1.Check if:
- The reagents are detoriated (reagent blank values are out of range)
- The method is correctly calibrated. Perform a new calibration.
- Control samples have been analyzed with the methods specified by
the manufacturer on the control table sheet.
- The settings of the method on the CYANStart (wavelength,
temperature,…) are correct. Check if the value of the standard is
correctly programmed.
- The reagents, controls and standards have been prepared according
to the instructions of the manufacturer.

2.Check if:
- The lamp is working correctly (procedure auto zero paragraph
3.6.2.1)
- The filter wheel sensor is ok (procedure paragraph 3.7.2)
- The filter en pre-filter AD values are correct (procedure paragraph
3.7.3)

Absorbance values are near to 1.Check if:


zero - The reagents have been detoriated (reagent blank values are out of
range)
- The method has been programmed correctly (especially wavelength)
- The lamp is on.
- The flow cell is in the correct direction
- The flow cell is totally inserted in the flow cell holder
- The flow cell is dirty
- The flow cell has been broken
- There are air bubbles in the flow cell
 Check tube connections or leaking and blocking of the tubes
- The peristaltic pump is turning

2.Check if:
- The lamp is working correctly (procedure auto zero paragraph
3.6.2.1)
- The filter wheel sensor is ok (procedure paragraph 3.7.2)

Power supply problem 1.Check if:


- The connection of the power is correct and firm
- The fuses are broken
- The connection of the power switch is correct

2.Check if:
- The transformer is still working. If not, replace it.

Problem with the aspiration 1.Check if:


- The tubes are jammed, fractured or aged and replace if necessary.
- The tubes are obstructed.
- The flow cell is obstructed.

2.Check if:
-The peristaltic pump is correctly calibrated (paragraph 3.5.2)
-The peristaltic pump motor is turning correctly.

Problem with flow cell 1.Check if:
temperature - The temperature of the flow cell is close to 37°C. If so, try to increase
or decrease the temperature by adapting the trimmers on the main
board described in paragraph 3.8.

2.Check:
- If the flow cell does not warm at all, check if the heater is broken and
check the voltage of the heater.

Problems with the screen 1.Check if:


- The connection between the main board and the screen is well
connected. Attention, there are two connections, one for the screen
and one for the keyboard.

- The contrast trimmer on the main board is well set. Turn the little
screw next to the screen connection on the main board to increase the
contrast of the screen.

Problem with the printer 1.Check if:


-The printer is activated in the software menu
-The paper is correctly inserted in the printer
-The green lever is pushed down

2.Check if:
- The connections between the main board and the printer are well
connected.

Problem with the Keyboard 1.Check if:


- The connection between the main board and the keyboard is well
connected. Attention, connecting it the wrong way will give problems!
Loosen the connection and try to connect it the other site.
